De huismussklok , Suncus murinus , behoort tot de orde Insectivora en de familie Soricidae. Dit kleine zoogdier is verspreid over heel Zuidoost-Azië en woont in huizen en grasgebieden die zich bevinden in de buurt van menselijke woningen of veestokken 4 . Deze soort vertoont algemene morfologische kenmerken die vergelijkbaar zijn met mensen dan andere laboratoriumdieren, zoals de muis, de rat en het konijn 5 . Vroeger werd immunostaining met een perifere neuronmarkering voor S. murinus neurofilament eiwit (NFP) gebruikt om perifere zenuwen in de alvleesklier 6 , hoofdduodenale papilla 7 , pylorus 8 , galblaas 5 en extrahepatische galwegen 9 te markeren. NFP is een macromoleculaire complex die deel uitmaakt van het volwassen neuronale cytoskelet. Neurofilament-gerelateerd eiwitS bemiddelen interacties tussen NFP en de zygosoom. Zowel de enzymfunctie als de structuur van linkerproteïnen worden gereguleerd door NFP 10 . Het NFP-complex is gemaakt van drie polypeptiden: NF-L (70 kDa), NF-M (150-160 kDa) en NF-H (200 kDa). NFP kan worden gevonden in neuronale axonen in zowel het centrale als perifere zenuwstelsel. Anti-menselijk NFP antilichaam is aangetoond om axonen van het centrale en perifere zenuwstelsel te vermelden. Anatomische en elektrofysiologische onderzoeken hebben aangetoond dat de sfincter, galblaas en proximale maagdarmkanaal 11 , 12 , 13 , 14 verbonden zijn. Echter, de morfologische kenmerken van hun neuronale verbindingen zijn nog niet gedefinieerd. Dit werk demonstreert het gebruik van een volledig-mount immunohistochemische kleuring methode om de innervation van de S. murinus bilIary kanaal met een NFP antilichaam.

Dit werk beschrijft de gehele berg immunohistochemie kleuring methode in detail, met behulp van neurofilament eiwit antilichaam voor het labelen van de innervatie van het galweg in Suncus murinus ( S. murinus &#160; ). Ten eerste werd het monster van S. murinus gescheiden en in 4% paraformaldehyde (PFA) gefixeerd. Ten tweede werd een enzymatische behandeling en potentiële endogene peroxidase-inactivering uitgevoerd. Het monster werd vervolgens blootgesteld aan het primaire antilichaam, anti-neurofilament eiwit antilichaam, gedurende 3-6 dagen. Het werd vervolgens geïncubeerd met het secundaire antilichaam dat is geconjugeerd met mierikswortelperoxidase. De kleurreactie werd geopenbaard door het monster te laten reageren met een 3,3&#39;-diaminobenzidine (DAB) substraat. Deze methode kan worden toegepast om de innervering van alle viscerale organen te analyseren. Bovendien kan dit protocol ook worden aangepast om andere neuronale antilichamen te testen, maar optimalisatie van de antibodiesHet moet eerst gedaan worden. Deze methode werd oorspronkelijk geïntroduceerd door Kuratani en Tanaka 1 , 2 , 3 .
