exposure of the pig cns for histological analysis: a manual for decapitation, skull opening, and brain removal

Grisar har blivit allt populärare i stora djur translationell neurovetenskaplig forskning som ett ekonomiskt och etiskt genomförbart substitut till icke-mänskliga primater. Den stora hjärnans storlek av grisen tillåter användning av konventionella klinisk hjärnkameror och direkt användning och testning av neurokirurgiska ingrepp och utrustning från den mänskliga kliniken. Ytterligare makroskopisk och histologisk analys, kräver emellertid postmortem exponering av grisen centrala nervsystemet (CNS) och efterföljande avlägsnande av hjärnan. Detta är inte en lätt uppgift, som grisen CNS är inkapslad av en tjock, benig skalle och ryggraden. Målet med detta dokument och instruktionsvideo är att beskriva hur man avslöja och ta bort döden gris hjärnan och hypofysen i en intakt tillstånd, lämplig för efterföljande makroskopisk och histologisk analys.

För att förhindra att vävnadsmaterialet från att torka ut, är det rekommenderat att lagra avlägsnas hjärnan och hypofysen i en burk fylld med fixativ eller isotonisk saltlösning omedelbart efter makroskopisk analys har utförts. Det vävnadsmaterial kan lagras i fixativ för år, medan lagring i isotonisk koksaltlösning, även i kylskåp, kommer att leda till vävnads förfall med tiden. Den avlägsnades hypofysen kan...
