Meer dan 200 onnatuurlijke aminozuren van verschillende chemische en fysische eigenschappen zijn genetisch in eiwitten opgenomen in E. coli , gist en zoogdiercellen 3 . Het onnatuurlijke aminozuur is opgenomen in reactie op een specifiek stopcodon via een orthogonaal ontwikkeld tRNA / synthetasepaar. De genetische benadering van het modificeren van eiwitten heeft waardevolle inzichten gegeven in eiwitstructuur en -functie. Hier presenteren we een protocol voor het gebruik van Voltage-Clamp Fluorometry (VCF) in combinatie met een fluorescerende UAA. In VCF staat de gelijktijdige observatie van functionele data en structurele omgevingen rond de fluorescerende sonde (~ 5 Å) ons in staat dynamische informatie te verkrijgen met millisecondresolutie 1 . De fluorescerende probes veranderen hun blusstaat op gelokaliseerde beweging van het eiwit. Een beweging van slechts 1-2 Å is voldoende om te leiden tot significante veranderingen in de fluorescentieIntensiteit 4 . Na identificatie van de bezienswaardigheid in het doelproteïne wordt de plaats gemuteerd door puntmutatie. Klassiek was het residu gemuteerd naar een cysteïne, terwijl nu een amber stopcodon (TAG) geïntroduceerd wordt voor genetische fUAA-incorporation. Het eiwit wordt vervolgens in vitro getranscribeerd. Terwijl andere expressiesystemen (bijvoorbeeld zoogdiercellen) 5 , 6 , 7 kunnen gebruiken, hebben Xenopus- oocyten de voorkeur aan structurele-functiestudies vanwege hun grotere grootte, wat leidt tot eenvoudiger manipulatie en hogere fluorescentie-intensiteit (meer fluorophoren) Ruisverhouding. Bovendien hebben Xenopus- oocyten lage achtergrond van endogene eiwitten 2 , 8 en de donkere pigmentatie op de dierenpoolschilden tegen achtergrondfluorescentie van tHij cytosol. De Xenopus- oocyten worden chirurgisch verwijderd en DNA dat codeert voor het orthogonale tRNA / tRNA-synthetase-paar dat specifiek is voor het fUAA, wordt geïnjecteerd in de kern van de oocyten. Na een incubatietijd van 6-24 uur wordt het eiwit RNA gecojecteerd met het fUAA in het cytosol van de oocyten, gevolgd door een incubatieperiode van 2-3 dagen. Om eventuele schade aan de fUAA te voorkomen (fotobladen) moeten de procedures inclusief Anap onder rood worden uitgevoerd om fluorofore excitatie te vermijden. Oocyten worden bestudeerd op een open-oocyte spanningsklemopstelling, die op een oprechte fluorescentiemicroscoop is gemonteerd. Elektrische stroom- en fluorescentieveranderingen worden gelijktijdig opgenomen 9 , 10 . Als alternatief kunnen twee-elektrode spanningsklem 1 of patch-klemconfiguraties 11 gebruikt worden. Fluorescentie wordt opgewonden door geschikte golflengten met laag RMS geluid en<html> Emissie opgenomen met behulp van een fotodiode gekoppeld aan een versterker met hoge amplificatie. voltage-clamp fluorometry in xenopus oocytes using fluorescent unnatural amino acids

Voltage-Clamp Fluorometry (VCF) is de gewenste techniek om de structuur en functie van elektrogenische membraanproteïnen te onderzoeken, waarbij real-time metingen van fluorescentie en stromingen gelijktijdig rapporteren over lokale omgevingen en globale functie respectievelijk 1 . Terwijl hoge-resolutie structurele technieken zoals cryo-elektronenmicroscopie of röntgenkristallografie statische beelden van de eiwitten van belang bieden, biedt VCF dynamische structurele gegevens die ons in staat stellen de structurele omzettingen (fluorescentie) te koppelen aan dynamische functionele data (elektrofysiologie). Tot voor kort beperkte de thiol-reactieve chemie die wordt gebruikt voor plaatsgerichte fluorescente etikettering van de eiwitten de reikwijdte van de aanpak, omdat alle toegankelijke cysteinen, waaronder endogene, worden gemerkt. Het was derhalve verplicht om eiwitten vrij te maken van endogene cysteinen. Etikettering was ook beperkt tot sites die toegankelijk waren voor de extracellulairekant. Dit veranderde met behulp van Fluorescent Unnatural Amino Acids (fUAA) om specifiek een kleine fluorescerende sonde in te nemen in reactie op stopcodononderdrukking met behulp van een orthogonale tRNA en tRNA synthetase pair 2 . De VCF-verbetering vereist alleen een tweetraps injectieprocedure van DNA-injectie (tRNA / synthetase-paar) gevolgd door RNA / fUAA co-injectie. Nu is het mogelijk om zowel intracellulaire als begraafde plaatsen te labelen, en het gebruik van VCF is aanzienlijk uitgebreid. De VCF techniek wordt daardoor aantrekkelijk voor het bestuderen van een breed scala van eiwitten en - nog belangrijker - maakt het mogelijk om talloze cytosolische regulerende mechanismen te onderzoeken.

Figuur 4 toont een voorbeeld van VCF-opnamen verkregen uit een oocytenuitdrukkende Shaker kanalen met snelle inactivatie verwijderd (IR), L382stop-W434F in aanwezigheid van pAnap en Anap. De W434F mutatie blokkeert de ionische kaliumstromen, waardoor het mogelijk is om de transiente gatenverplaatsingen (gatenstromen) te meten.
