혈관은 몸 전체에 산소와 영양분을 공급하며 혈관 네트워크의 형성은 엄격한 발달 조절하에 있습니다. 혈관의 효율적인 생체 내 시각화 및 그 복잡성의 신뢰할 수있는 정량화는 혈관 네트워크의 생물학 및 질병을 이해하는 데 중요합니다. 여기서는 상업적으로 이용 가능한 형광 염료 인 인간 혈장 아세틸 화 저밀도 지단백 DiI 복합체 (DiI-AcLDL)를 사용하여 혈관을 시각화하고 Xenopus tropicalis 의 복잡성을 정량화하는 자세한 방법을 제공합니다. 혈관은 DiI-AcLDL을 배아의 심장 박동에 간단히 주입하여 표시 할 수 있으며 전체 배아의 혈관을 생체 또는 고정 배아에서 영상화 할 수 있습니다. 핵산의 표적 미세 주입 및 / 또는 약리학 적 시약의 배양 적용에 의한 유전자 교란과 결합하여, 혈관 발생에 대한 유전자 또는 신호 전달 경로의 역할을 수행 할 수있다정교한 유전자 조작 동물에 의지하지 않고 1 주일 이내에 징계 조치를 취했다. 잘 정의 된 Xenopus의 정맥 시스템과 그 고정 관념의 혈관 신생, 기존 혈관의 싹 트기 때문에 혈관의 복잡성은 섭동 실험 후에 효율적으로 정량화 될 수 있습니다. 이 비교적 단순한 프로토콜은 심혈관 연구 분야의 다양한 분야에서 쉽게 접근 할 수있는 도구가되어야합니다.
