Sensorineural Hörverlust (SNHL) spiegelt Schäden am Innenohr oder aufsteigender Gehörgang. Während Hörverlust das häufigste sensorische Defizit bei Menschen 1 ist , existieren heilende Therapien noch nicht 2 . Obgleich Cochlear- oder auditorische Hirnstammimplantate ein gewisses Maß an Hörvermögen für Patienten mit schwerer bis tiefgehender SNHL wiederherstellen können, ist das Hören, das von diesen Geräten zur Verfügung gestellt wird, immer noch sehr verschieden von dem &quot;natürlichen&quot; Gehör, vor allem bei Versuchen, die Sprache im Lärm zu verstehen oder Musik zu hören. Während die Haarzelldegeneration seit langem als die primäre Konsequenz traumatischer auditorischer Ereignisse ( z. B. neonatal murine cochlear explant technique as an in vitro screening tool in hearing research

Während es in den vergangenen Jahrzehnten bemerkenswerte Fortschritte in der Hörforschung gegeben hat, gibt es noch keine Heilung für den sensorineuralen Hörverlust (SNHL), eine Bedingung, die typischerweise eine Beschädigung oder einen Verlust der empfindlichen mechanosensorischen Strukturen des Innenohrs beinhaltet. Ausgefeilte In-vitro- und Ex-vivo- Assays haben sich in den letzten Jahren entwickelt, was es ermöglicht, eine zunehmende Anzahl von potentiell therapeutischen Verbindungen zu screenen und gleichzeitig die Ressourcen zu minimieren und die Bemühungen um die Entwicklung von Kuren für SNHL zu beschleunigen. Die Erhaltung organisierter Zellstrukturen im Innenohr erleichtert die in situ- Bewertung verschiedener Komponenten der Cochlea-Infrastruktur, einschließlich innerer und äußerer Haarzellen, Spiralganglion-Neuronen, NeurItes und unterstützende Zellen. Hier präsentieren wir die Vorbereitung, Kultur, Behandlung und Immunfärbung von neonatalen murinen Cochlea-Explantaten. Die sorgfältige Vorbereitung dieser Explantate erleichtert die Identifizierung von Mechanismen, die zu SNHL beitragen und ein wertvolles Instrument für die Hörforschungsgemeinschaft darstellen.

Während viele Protokolle auf Orgel von Corti explants konzentrieren, versucht diese Technik, die Anatomie der gesamten Cochlear-Umdrehung zu bewahren, einschließlich der SGNs.
