La recherche sur l&#39;hypoxie comporte de nombreuses applications. Il s&#39;agit notamment d&#39;enquêter sur la pathogenèse et de développer des traitements pour les affections médicales caractérisées par une hypoxie 1 et une maladie aiguë de haute altitude 2 . Le stress hypoxique entraîne des changements métaboliques majeurs chez tous les organismes nécessitant de l&#39;oxygène. Le stress hypoxique influence également la croissance et le développement du fœtus et la pathogenèse de plusieurs maladies humaines, y compris la restriction de la croissance intra-utérine 3 . Le stress hypoxique peut non seulement entraîner une réduction du poids à la naissance, la mortalité du foetus et du néonatale, mais aussi entraîner de nombreuses complications dans la vie adulte, telles que les maladies cardiovasculaires, le diabète de type 2, l&#39;obésité et l&#39;hypertension 4 . Le stress hypoxique est également souvent observé lors du développement de la tumeur solide, lorsque le tissu tumoral dépasse son apport sanguin. Il est donc crucial de pouvoir étudier les effets de l&#39;hypoxie in vivo et directement pendant l&#39;embryon<html> Développement yonique. Parmi les méthodes les plus connues qui ont été utilisées pour étudier les effets de l&#39;hypoxie au cours du développement, il y a l&#39;utilisation de chlorure de cobalt dans le milieu de croissance ou l&#39;incubation de l&#39;organisme dans une chambre hypoxique. Le chlorure de cobalt induit artificiellement une réponse hypoxique sous une concentration normale d&#39;oxygène, en raison de son rôle dans la stabilisation du facteur 1 inducible par l&#39;hypoxie (HIF-1α) en empêchant sa dégradation protéosomale 5 , 6 , 7 . Cependant, étant une méthode pratique 8 , l&#39;utilisation de chlorure de cobalt ainsi que d&#39;autres mimétiques d&#39;hypoxie chimique similaires peut avoir un effet délétère non spécifique sur les cellules et les tissus, par exemple , l&#39;apoptose 9 . Par conséquent, les chambres hypoxiques sont une meilleure méthode pour l&#39;induction de «l&#39;hypoxie naturelle» dans les organismes vivants au cours du développement normal. Nous nous sommes concentrés sur le développement d&#39;un système d&#39;induction de l&#39;hypoxie chez les embryons d&#39;animaux aquatiques. Les grenouilles et le poisson zèbre sont devenus des organismes modèles informatifs à base de vertébrés pour des études sur de nombreux processus biologiques, ainsi que des modèles pour diverses maladies humaines. Se développent à l&#39;extérieur, éliminant la complication de la compensation maternelle. En outre, un développement rapide permet de manipuler les facteurs environnementaux et d&#39;observer les changements phénotypiques dans la formation des organes en temps réel. Le principal avantage de l&#39;utilisation de grenouilles et d&#39;embryons de poissons zèbres pour étudier les effets de l&#39;hypoxie sur le développement des vertébrés est que tous les processus peuvent être surveillés directement, car l&#39;oxygène pénètre rapidement dans les embryons. Ainsi, dans les grenouilles et le poisson zèbre, contrairement aux autres organismes modèles tels queEmbryons de souris, l&#39;influence d&#39;une concentration d&#39;oxygène spécifique peut être étudiée dans le tissu d&#39;intérêt, sans tenir compte de la présence ou du manque de système vasculaire fonctionnel. La plupart des configurations disponibles dans le commerce pour l&#39;incubation hypoxique présentent l&#39;inconvénient d&#39;être relativement importantes et d&#39;avoir des coûts de fonctionnement élevés. Outre leur coût initial élevé et leur consommation de gaz, l&#39;équilibrage et l&#39;entretien des chambres communes d&#39;hypoxie nécessitent un maintien de l&#39;atmosphère hypoxique constante contre le gradient de gaz qui se produit naturellement dans ces chambres en raison de leur plus grande taille et / ou de leur respiration. Cela nécessite l&#39;emploi de ventilateurs à gaz et un système de refroidissement, ce qui augmente la quantité d&#39;équipement nécessaire supplémentaire, entrave la dextérité du chercheur et diminue globalement la simplicité de la procédure expérimentale. En revanche, la configuration que nous présentons ici est relativement robuste mais très rentable, petite, facile à établir et permet fÉquilibre économique des gaz, atmosphère hypoxique stable et échange simple de matériaux et de solutions dans la chambre. Notre système peut être utilisé pour être utilisé avec n&#39;importe quel organisme modèle aquatique d&#39;intérêt. Nous avons construit une chambre hypoxique qui est commodément petite et donc peut être placée dans un incubateur de laboratoire commun, ce qui permet facilement des procédures expérimentales à n&#39;importe quelle température spécifique. Offrant un contrôle pratique de la température ainsi que de la concentration d&#39;oxygène dans le milieu, l&#39;avantage de notre système contre les incubateurs d&#39;hypoxie disponibles dans le commerce réside dans sa petite taille et son rapport coût-efficacité. Ainsi, notre configuration peut être établie en utilisant des fournitures de laboratoire générales disponibles pour la majorité des laboratoires de recherche et ne nécessite aucun matériau coûteux. En outre, notre installation ne génère pas de chaleur, contrairement aux incubateurs d&#39;hypoxie disponibles dans le commerce, et permet une utilisation à des températures inférieures à la température ambiante placées dans un incubateur. La laSt est particulièrement critique pour le travail avec des organismes à sang froid tels que les grenouilles et les poissons où les taux de développement et métabolisme dépendent fortement de la température. Étant très rentable et facilement construit, notre chambre d&#39;incubation de gaz est néanmoins très polyvalente dans l&#39;établissement de diverses conditions hypoxiques ou hyperoxiques, tout en permettant une administration rapide et facile de différents supports et solutions pour un grand nombre de conditions expérimentales. En outre, en utilisant une plaque de 24 puits au lieu des plats couramment utilisés ou des réservoirs de laboratoire 10 , 11 , 12 , notre système permet l&#39;observation et le traitement expérimental de plusieurs conditions mutantes à la fois. Pour contrôler l&#39;induction correcte de l&#39;hypoxie, nous avons surveillé les niveaux de la protéine HIF-1α par détection Western Blot. En outre, le nombre de cellules proliférantes avant et après incubationN dans la chambre hypoxique peut être utilisé pour déterminer si l&#39;hypoxie a été induite dans le tissu. Cette méthode est basée sur nos résultats précédemment publiés 13 , montrant que la prolifération du nerf embryonnaire de cellules souches rétiniennes diminue lors de l&#39;induction de l&#39;hypoxie. Ainsi, nous avons surveillé le niveau de prolifération des cellules souches de la rétine en ajoutant de la 5-éthynyl-2&#39;-désoxyuridine (EdU) au milieu embryonnaire et en mesurant son incorporation dans l&#39;ADN des cellules nouvellement proliférantes.

Ici, nous introduisons un nouveau système pour l&#39;induction de l&#39;hypoxie, que nous avons développé pour étudier les effets de l&#39;hypoxie chez les organismes aquatiques tels que les embryons de grenouille et de poisson zèbre. Notre système comprend une chambre avec une configuration simple qui est néanmoins robuste pour induire et maintenir une concentration et une température d&#39;oxygène spécifiques dans n&#39;importe quelle solution expérimentale de choix. Le système présenté est très rentable mais très fonctionnel, il permet l&#39;induction et le maintien en puissance de l&#39;hypoxie pour des expériences directes in vivo et pour différentes périodes jusqu&#39;à 48 h. Pour surveiller et étudier les effets de l&#39;hypoxie, nous avons utilisé deux méthodes: la mesure des niveaux de facteur 1alpha inducible à l&#39;hypoxie (HIF-1α) dans des embryons entiers ou des tissus spécifiques et la détermination de la prolifération des cellules souches de la rétine par 5-éthynyl-2&#39;- Incorporation de désoxyuridine (EdU) dans l&#39;ADN. Les niveaux de HIF-1α peuvent servir de marqueur d&#39;hypoxie générale dans l&#39;ensemble de l&#39;embryon ou du tissuDe choix, ici la rétine embryonnaire. L&#39;incorporation d&#39;EdU dans les cellules proliférantes de la rétine embryonnaire est une production spécifique d&#39;induction d&#39;hypoxie. Ainsi, nous avons montré que les progéniteurs de rétine embryonnaire hypoxique diminuent la prolifération dans les 1 h d&#39;incubation sous 5% d&#39;oxygène des embryons de grenouille et de poisson zèbre. Une fois maîtrisé, notre installation peut être utilisée pour être utilisée avec de petits organismes modèles aquatiques, pour des expériences directes in vivo , une période de temps donnée et sous une concentration d&#39;oxygène normale, hypoxique ou hyperoxyde ou sous tout autre mélange de gaz donné.

L&#39;utilisation du système de chambre hypoxique que nous présentons ici permet d&#39;étudier les effets de l&#39;hypoxie individuellement et in vivo chez les animaux vivants entiers. The overall goal of this invention is to study the effects of hypoxia or hyperoxia on the embryonic tissues of any aquatic organism of interest. This method can help answer key questions in the field of developmental and cell biology, such as embryonic growth restriction, fetal hypoxia, high-altitude adaptation, obesity and solid tumor development. The main advantages of this technique are simplicity, cost-effectiveness and robustness.It allows induction and sustainment of hypoxi in vivo for long time periods. After collecting and raising xenopus and zebrafish embryos, prepare a mesh-bottom 24-well plate according to the text protocol. Attach plastic rods to the mesh-bottom 24-well plate and place it into the aquatic tank of choice, then using silicone tubing, attach a gas tank with the desired oxygen nitrogen mixture, or another gas mixture of choice, to a distributor valve using an appropriate gas regulator.Next connect the silicone tubing and the distributor valve to a ceramic disk diffuser inside the aquatic tank. Depending on the type of embryos used in the experiment, fill the hypoxic chamber tank with the appropriate embryo medium, then close the aquatic tank and use silicone grease to coat the lid and carefully seal it. Start diffusing the gas mixture through the ceramic disk diffuser.Allow the system to equilibrate for 10 to 15 minutes. Adjust the oxygen flow rate in the chamber medium to 0.0017 to 0.0019 cubic meters per second. Under this gas flow rate, no disturbance of the medium should be seen in the wells of the plate.The oxygen flow rate depends on the internal pressure of the gas tank and the outlet pressure of the gas regulator. Both these parameters should be adjusted according to the monitor of the gas regulator. After equilibration, use an oximeter or oxygen probe to measure the oxygen concentration in the water.If a particular non-room temperature is required, place the hypoxic chamber into a laboratory incubator of the required temperature. Carefully select the embryos for the experiment using whole and live embryos for hypoxia incubation. Place the embryo dishes into the incubator containing the gas incubation chamber, and let them equilibrate to the temperature of the incubator.Then use a plastic pipette to carefully transfer the embryos into the wells of the meshed 24-well plate of the gas incubation chamber. Carefully label the wells with different genotypes. Ensure minimal reoxygenation of the chamber while placing the embryos into the wells.Transfer the sample swiftly, and quickly reseal the chamber. Maintain the gas incubation chamber under a constant infusion with the gas mixture of choice for five hours, or for a longer time that suits the experiment. A mixture of 5%oxygen and 95%nitrogen is used here.Carefully transfer the embryos from the gas incubation chamber back to the normoxic medium corresponding to the embryo type. After preparing buffers and media, and anesthetizing the embryos according to the text protocol, use a tissue homogenizer or a sonicator to homogenize the embryo tissue. Then centrifuge the homogenates to remove debris.Collect the supernatant and denature the sample at 85 degrees Celsius for five minutes, then add Laemmli gel loading buffer to one X volume by volume. Use the supernatant for western blot analysis, or freeze the samples at negative 20 degrees Celsius. To incorporate EdU into the embryos, fill the gas incubation chamber with 400 to 500 milliliters of five millimolar EuD solution supplemented with 1%volume by volume DMSO.Connect the gas tubing to the gas cylinder containing a gas mixture of 5%oxygen and 95%nitrogen. Incubate the solution for 15 minutes. Pre-equilibrate the solution with the same gas mixture used in the experiment for 15 minutes prior to the experiment.Transfer the embryos to the hypoxic chamber, distributing them evenly between the wells, and incubate the embryos for two hours. Finally, analyze the embryos for EdU incorporation according to the text protocol. As shown in this table, desired oxygen concentrations in the incubation medium were induced at different time points throughout the experiments, as monitored by a fiber optic oxygen sensor.In this experiment HIF-1 alpha which is stabilized under low oxygen concentrations, was measured in whole frog embryo lysates kept under normal oxygen concentrations, or subjected to 5%hypoxia and in lysates of their isolated retinas. In both samples HIF-1 alpha was stabilized under hypoxia. Here frog and zebrafish embryos were incubated in a hypoxic chamber maintained at 5%oxygen, and retinal proliferation was assessed by monitoring EdU incorporation.As expected normoxic control retinas of frog and zebrafish embryos showed intensive staining in the CMZ, where proliferating progenitors reside. Following the oxygen incubation in the hypoxic chamber a strong decrease in the CMZ progenitor proliferation was observed in both frog and zebrafish embryos. By carrying out a time course it could be shown that the decrease in retinal progenitor proliferation was acute, and greatest within two hours, and persisted for many hours while embryos developed normally according to their developmental stage.Once mastered, this technique can be done in 30 minutes if it is performed properly. While attempting this procedure it's important to remember to ensure correct gas flow rate, and to avoid unnecessary and excessively long reopening of the chamber. Following this procedure other methods like QPCR can be performed in order to answer additional questions like activation, or down-regulation of certain target genes of HIF-1 alpha and hypoxia, or hyperoxia respectively.After its development this technique paved the way for the researchers in the field of developmental biology to explore retinal cell proliferation and hypoxia and restricted nutrition in frog and zebrafish embryos. After watching this video you should have a good understanding of how to induce hypoxia or hyperoxia in any aquatic organism, and how to study its effects on the tissue and phenomenon of interest, such as retinal cell proliferation. Don't forget that working with oxygen gas mixtures can be extremely dangerous, and precautions such as ensuring correct use and attachment of gas regulators should always be taken while performing this procedure.
