The production of macroporous matrices can be beneficial to many different fields. Hydrogels zijn materialen die vormen van onoplosbare 3D netwerken kunnen grote hoeveelheden water bindend. Dergelijke materialen bieden uitstekende omgevingsvoorwaarden voor levende cellen. Op dit moment, er is steeds meer belangstelling in de generatie van driedimensionale hydrogel structuren en de ontwikkeling van procédés voor het aanpassen van hun chemische en fysische eigenschappen. Zodra dit gebeurt, kan een sjabloon voor de groei van cellen en de manipulatie van cellulaire gedrag gegenereerde1,2,3,4. Deze 3D structuren niet alleen maakt een meer natuurlijke en realistische omgeving dan conventionele tweedimensionale benaderingen, maar ze ook onthullen nieuwe mogelijkheden voor de groei van stamcellen of tumor5modellen. Verschillende materialen bezitten een aantal kenmerken die vooral afhankelijk zijn van de poriegrootte van het gel-6. De poriën spelen een cruciale rol in de gestuurde groei van stamcellen cel cultuur toepassingen en weefselkweek. Bijvoorbeeld, zuurstof en voedingsstoffen verspreiden via de matrix, en voldoende hoeveelheden moeten kunnen bereiken van de cellen7. Aan de andere kant, schadelijke metabolieten moeten zo spoedig mogelijk worden verwijderd, en moet voldoende ruimte bieden voor celgroei beschikbaar7. Bijgevolg beïnvloeden de eigenschappen van het materiaal, en dus de poriegrootte, ernstig de potentiële voordelen en mogelijke toepassingen van de matrix. Afhankelijk van de eigenschappen van het materiaal, kunnen verschillende celgroei processen optreden in de 3D-celkweek, met inbegrip van de vorming van neuronale structuren; de groei en differentiatie van huid of bot cellen; en de gestuurde groei van speciale stamcellijnen, zoals hepatocyten of fibroblasten2,3,8,9,10,11. Een ander cruciaal punt beïnvloeden de eventuele toepassing van een materiaal, is zijn stabiliteit naar externe prikkels12. Bijvoorbeeld, moet de hydrogel zijn mechanische integriteit in cel voedingsbodems of het menselijk lichaam.
In de afgelopen jaren onderzoek naar 3D cel cultuur hydrogels geïntensiveerd, en vele studies werden uitgevoerd om te lossen van de 3D-platforms van de systemen-13. Hydrogels bestaat uit chemisch gesynthetiseerde onderdelen zijn meestal onderzocht omdat zij kunnen gemakkelijk gesynthetiseerd en chemisch bewerkt en ze vertonen een hoge stabiliteit (Zie Zhu et al., 2011 herzieningsverzoek)5. Eiwitten hebben echter vele heilzame eigenschappen: als zogenaamde "precisie polymeren," ze zijn biocompatibel; ze hebben een gedefinieerde lengte; ze zijn relatief makkelijk te wijzigen; en ze hebben een groot aantal doel sites14,15. In dit opzicht kunnen zeer specifieke, innovatieve structuren worden gegenereerd voor toepassing op vele terreinen. In deze studie werd een op basis van eiwitten hydrogel16 gebruikt om aan te tonen van het vermogen van gevestigde methoden om de invloed van de 3D-architectuur van het materiaal. Bovendien, het vermogen en de toepasbaarheid op porie generatie werd eveneens onderzocht.
Veel verschillende technieken zijn beschikbaar voor het wijzigen van 3D-structuren, met inbegrip van zowel eenvoudige methoden en geavanceerde, zeer gespecialiseerde technieken uit verschillende gebieden van de materiaalkunde. Een wijdverbreide techniek is het gebruik van electrospinning voor het genereren van welomschreven structuren17. Geladen vezels zijn getrokken uit een oplossing door een elektrisch veld en vervolgens stollen bij blootstelling aan zuurstof. Op deze manier kunnen de vezels in de range van verschillende nanometer tot verschillende micron worden geproduceerd. Additionele technieken om af te stemmen op de grootte, structuur en verdeling van de poriën in de matrix zijn zachte lithografie fotolithografie, hydrodynamische gericht, electro-spuiten, en bio-afdrukken18,19,20. Een belangrijk nadeel van deze technieken is hun afhankelijkheid op specifieke en dure apparatuur en speciale chemische stoffen of materialen. Bovendien ervaring met deze technieken is vaak niet direct overdraagbaar naar materialen op basis van eiwitten, en veel van de chemicaliën en de methoden zijn niet cel compatibel.
Aan de andere kant, vertrouw veel technieken niet op speciale apparatuur, waardoor ze gemakkelijker en goedkoper om toe te passen en te reproduceren. Een wijdverbreide methode voor structuur manipulatie is oplosmiddel gieten21,22,23. Deeltjes zijn toegevoegd voordat de polymerisatie-reactie en homogeen worden verdeeld om te verzadigen van de oplossing. Na de polymerisatie leidt een verandering van omstandigheden, zoals een verdunning of een verandering van de pH, tot het oplossen van de deeltjes, terwijl de poriën binnen het materiaal blijven. De chemicaliën die worden gebruikt in deze technieken, zoals zout, suiker, paraffine, gelatine en krijt, zijn goedkoop en gemakkelijk beschikbaar. In het vriesdrogen, zijn gezwollen hydrogels bevroren. De daaropvolgende sublimatie van de vloeibare fasen onder een vacuüm is dan23,24,25uitgevoerd. Water sublimatie van het netwerk is zacht genoeg om de specifieke 3D structuren van het materiaal. In het gas wordt schuimvorming, wordt een oplossing gestreamd met een gas terwijl de polymerisatie plaats vindt, waardoor poriën binnen de gel-21. De grootte en de verdeling van de poriën kunnen worden aangepast afhankelijk van de gasstroom.
Om de eiwit hydrogel, is BSA reageerde met tetrakis (hydroxymethyl) fosfonium chloride (THPC) in een Mannich-type reactie te voorzien in de vorming van covalente bindingen tussen primaire amines en de hydroxy groepen van de vier-armige linker molecuul26. Mogelijk schadelijke tussenproducten worden verwijderd door overmatig wassen van het materiaal na de reactie optreedt.
Deze studie toont aan dat de mogelijkheid van het behandelen van een BSA gebaseerde materiaal met verschillende technieken om te manipuleren en aanpassen van de grootte van de poriën. Elk van de technieken kan worden gebruikt in elk laboratorium wereldwijd, zoals er geen speciale apparatuur nodig is. Bovendien verschillende parameters, zoals zwelling verhouding, enzymatische afbraak, stabiliteit van de pH en temperatuur gevoeligheid, zijn onderzocht en met elkaar vergeleken, met name de eerbiediging op de invloed van de verschillende technieken op de generatie van 3D platforms. De materialen werden ten slotte matiemaatschappij met cel-lijm peptiden te onderzoeken van de eventuele toepassing van de materialen op de cultuur van de cel. Twee verschillende model-cellijnen werden gebruikt: A549 en MCF7.

easy manipulation of architectures in protein-based hydrogels for cell culture applications

Hydrogels worden erkend als veelbelovende materialen voor cel cultuur toepassingen vanwege hun vermogen om bieden zeer gehydrateerd cel omgevingen. Het gebied van 3D sjablonen stijgt als gevolg van de potentiële gelijkenis van die materialen naar de natuurlijke extracellulaire matrix. Hydrogels op basis van eiwitten zijn bijzonder veelbelovend, omdat ze gemakkelijk kunnen worden matiemaatschappij en gedefinieerde structuren met verstelbare fysisch-chemische eigenschappen kunnen bereiken. De productie van de 3D sjablonen macroporeuze voor cel cultuur toepassingen met behulp van natuurlijke materialen wordt echter vaak beperkt door hun zwakkere mechanische eigenschappen vergeleken met die van synthetische materialen. Hier, verschillende methoden werden geëvalueerd om te produceren macroporeuze bovien serumalbumine (BSA)-gebaseerde hydrogel systemen, met verstelbare porie formaten in het bereik van 10 tot en met 70 µm in straal. Bovendien is een methode voor het genereren van kanalen in dit materiaal op basis van eiwitten die verschillende honderd micron lange opgericht. De verschillende methoden voor de productie van poriën, evenals de invloed van poriegrootte op materiaaleigenschappen zoals zwelling verhouding, pH, temperatuur stabiliteit en enzymatische afbraak gedrag, werden geanalyseerd. Porie maten werden onderzocht in de native, gezwollen Braziliaanse deelstaat de hydrogels met behulp van confocale laser scanning microscopie. De haalbaarheid voor cel cultuur toepassingen werd geëvalueerd aan de hand van een cel-lijm RGD peptide wijziging van het systeem van eiwit en twee model cellijnen: menselijke borstkankercellen (A549) en adenocarcinomic menselijke alveolaire basale epitheliale cellen (MCF7).

Easy Manipulation of Architectures in Protein-based Hydrogels for Cell Culture Applications

The overall goal of this methodology is to create macroporous hydrogels with varying pore sizes for cell culture applications. This method can help answer key questions in the field of 3D cell culture such as the principle feasibility of a novel hydrogel material for 3D cell culture applications. The main advantage of this methodology is that it offers a range of techniques to optimize and adjust a novel material to the requirements of different types of cells.To begin, dilute one milligram of the RGD peptide or an equivalent cell adhesive peptide in 100 microliters of sterile water to obtain the 10 milligram per milliliter solution. Next, remove the bottom of a 96 well plate and replace it with removal plastic wrap. Mix 100 microliters of BSA stock solution and 100 microliters of THPC stock solution in the 96 well plate to obtain 200 microliters of the hydrogel.Mix the components by pipetting up and down at least five times to guarantee a uniform hydrogel after polymerization. Place the 96 well plate at room temperature for about 10 minutes until all hydrogels are properly polymerized. Following polymerization, carefully and slowly remove the plastic wrap from the bottom of the plate.Press the hydrogels out of the 96 well plate using a small stab and transfer them to 1.5 milliliter tubes with sterile PBS pH 7.4. Store the hydrogels in phosphate buffered saline at four degrees Celsius for up to several months. To freeze-dry the hydrogels, first fill 1.5 milliliter tubes with 500 microliters of sterile deionized water and remove the caps.Transfer the hydrogels to the 1.5 milliliter reaction tubes using forceps. Wrap the 1.5 milliliter tubes tightly with paraffin film using at least three layers for each vial. Use a needle to pierce small holes in the film to enable gas release from the tubes.To guarantee that the water and hydrogel completely freeze, transfer the vial to liquid nitrogen solution for five minutes. Immediately after removal from the liquid nitrogen, transfer the vials to the freeze-dryer to prevent the material from thawing. As an alternative freezing step, keep the vials at minus 20 degrees Celsius overnight to slowly freeze the hydrogels.Immediately after removal from minus 20 degrees Celsius, transfer the vials to the freeze-dryer to prevent the material from thawing. After 24 hours and the complete evaporation of the water in and around the hydrogel, thaw the material by removing it from the freeze-dryer. For particle leaching, mix the components of the hydrogel as before.Following mixing, immediately add sodium chloride until saturation occurs. Add salt until salt crystals can be seen in the solution as a white precipitate. Pore sizes can be altered by grinding the salt crystals prior to use.Transfer the 96 well plate to a shaker and shake until polymerization takes place. After about 10 minutes, remove the hydrogels from the plate as before. Incubate the hydrogels for at least 24 hours at room temperature in sterile water on a rotator to elute all salt from the hydrogel template.Store the hydrogels at four degrees Celsius in PBS for up to several months. For gentle formation, prepare hydrogels as described. Then remove a hydrogel from solution using a pincer.Remove the excess water with highly absorbent paper and place it on top of a block of dry ice. Freeze the hydrogel for 30 seconds and carefully remove it from the block. Do not damage the hydrogel.Carefully use a spatula to scrape it off the block. Transfer the hydrogel to a 1.5 milliliter tube and dry it overnight at 37 degrees Celsius. Prepare a Rhodamine B stock solution as well as serial dilutions as described in the text protocol.Remove the hydrogel from the stored solution and transfer it to a 1.5 milliliter tube with one milliliter of 0.01 milligram per milliliter Rhodamine B solution. Stain the hydrogel overnight in Rhodamine B solution at room temperature. The next day, transfer the hydrogel that is to be visualized to 10 milliliters of PBS and wash for at least three hours.Cut small slices out of the hydrogel using a blade. Then transfer the hydrogel onto an eight well microscopic slide and cover it with PBS. Using a confocal laser scanning microscope, visualize the hydrogel at a wavelength of 514 nanometers.Sterile filter all stock solutions with a 0.45 micron filter. Begin preparation of the hydrogel and include the cell adhesive peptide prior to hydrogel polymerization. After mixing the components, immediately pipette the mixture into an eight well microscopic slide until the bottom is completely covered.Transfer the cells into pre-warmed sterile DMEM cell culture medium supplemented with fetal bovine serum, penicillin streptomycin, and nonessential amino acid solution. After counting the cells with a Neubauer counting chamber, carefully pipette 200 microliters of the desired number of cells onto the hydrogel surface. Cover the eight well microscopic slide with a lid and transfer it to an incubator.After at least four hours of cellular attachment, wash the cells twice with 200 microliters of sterile cell culture PBS. Next, fix the cells with 200 microliters of 3.7%formaldehyde for 10 minutes at room temperature and wash twice with PBS. Permeabilize the cells with 200 microliters of 0.1%Triton X for five minutes before washing twice more with PBS.Stain the cells with Toluidine Rhodamine by mixing five microliters of methanolic stock with 195 microliters of PBS and adding it to the cells at room temperature. Stain the cells for 20 minutes in the dark before washing twice with PBS. Investigate the cell adhesion properties using a confocal microscope at 514 nanometers.Analyze the hydrogel properties as described in the text protocol. As shown here, the polymerized and stained hydrogels have no distinctive architectures. After freeze-drying the hydrogels at different temperatures, the average size of the pores within the hydrogel can be extended up to 60 microns.Salt leaching results in uniform networks with pore sizes of about 10 microns. The size can be further adjusted by grinding salt crystals prior to use. The use of a gradient freezing approach on a block of dry ice leads to the formation of channels with a diameter of about 20 microns and a length of several hundreds of microns.Prior to functionalization with the cell adhesive moiety, cells cannot adhere to the surface of the hydrogel and show a round morphology. Only after providing the material with the cell adhesive peptide, the cells find focal adhesion points which enable a proper adhesion of the cell onto a material. This is crucial for use of novel materials in 3D cell culture applications.After watching this video, you should have a good understanding of how to manipulate a hydrogel's architecture with simple methods.
