For at afhøre virkningerne af formaldehyd (FA) indånding på atopisk dermatitis (AD) progression, vi etableret en protokol til annonce induktion og udviklet en konventionel omsætning kammer for kronisk eksponering af FA.
Annonce er en kronisk inflammatorisk hudsygdom tilskrevet det komplekse samspil mellem genetiske disposition og miljømæssige udløser1,2. En ordentlig dyremodel er forpligtet til at studere annonce i prækliniske tilstand. Dette er det vigtigste, når belyse patogenesen af annonce under indflydelse af miljøfaktorer. For at bedre undersøge virkningerne af de miljømæssige udløser på AD progression, er det desuden vigtigt at adskille de enkelte miljøfaktorer og kvantificere graden af eksponering.
Vi har demonstreret en roman rotte model af annonce induceret af neonatal capsaicin behandling. Denne annonce model3 har nogle fortrin over andre AD modeller såsom AD modeller induceret af epicutaneous sensibilisering, genetisk manipuleret AD modeller og de spontane musemodeller af annonce4. Første, denne model udvikler annonce-lignende symptomer i en alder af 3 uger, som svarer til spædbarn AD og er tidligere end andre spontane musemodeller af annonce som NC/Nga musen. I nogle tilfælde, i modsætning til de genetisk manipuleret AD modeller, symptomerne for AD modeller stilne og tilbagefald spontant over en periode på 20 uger. Derudover er induktion processen med AD-lignende symptomer i modellen lettere end epicutaneous sensibilisering model, som mere ligner den allergisk kontakteksem i perspektiv af induktion processen.
Vi har indført den nye protokol for FA eksponering for en dyremodel ved hjælp af en akrylglas kammer og en konventionel medicinsk ilt regulator5 for at studere virkningerne af FA indånding på udviklingen af annonce i infantile og Ungdomspsykiatrisk perioder. Den regulerende myndighed har tilladt os at udsætte dyr for en konstant koncentration af gas ved at justere strømningshastigheder og opløsningens koncentration. Tidligere undersøgelser har rapporteret adskillige måder af eksponering af FA til dyremodeller. Xu et al. beskrev metoden eksponering i hvilken FA var løsningen direkte påføres huden6. En anden nylig undersøgelse rettet i protokollen af FA gas eksponering ved hjælp af en specialiseret flygtige opløsningsmidler generator, model 49127. I forhold til metoder til tidligere undersøgelser, den nye metode er lettere udstyret og kan anvendes for andre løsninger ud over FA.

Atopisk dermatitis recidiverende kronisk kløende eksem og hersker rundt om i verden især i de udviklede lande. Komplekse samspil mellem genetiske og miljømæssige faktorer er kendt for at spille en vigtig rolle i Patofysiologi af atopisk dermatitis. Vi mangler dog stadig et detaljeret billede af patogenesen af denne sygdom. Det er således vigtigt at udvikle passende dyremodeller for belyse progression af atopisk dermatitis. Derudover udvider undersøger effekten af miljøfaktorer såsom luft forurenende stoffer på atopisk dermatitis forståelse af sygdommen. Her, beskriver vi en metode til at inducere atopisk dermatitis i rotter med neonatal capsaicin behandling og en protokol for eksponering af en konstant koncentration af formaldehyd til rotter kan afsløre virkninger på udviklingen af atopisk dermatitis i infantile og Ungdomspsykiatrisk perioder. Disse protokoller har været anvendt med succes til flere eksperimenter og kan bruges til andre stoffer.
