Het protocol hier beschreven voor de inductie van AD-achtige symptomen bij ratten kan worden aangepast voor het bestuderen van AD pathofysiologie en gebruikt als een instrument voor screening voor drugtest. Om de effecten van formaldehyde (FA) inademing op atopische dermatitis (AD) progressie te ondervragen, we een protocol voor AD inductie opgericht en ontwikkeld van een conventionele omloop kamer voor chronische blootstelling voor FA.
AD is een chronische inflammatoire huidziekte toegeschreven aan de complexe interacties tussen genetische aanleg en milieu triggers1,2. Een goede diermodel is vereist voor het bestuderen van AD in de preklinische staat. Dit is het belangrijkste bij het ophelderen van de pathogenese van AD onder invloed van de omgevingsfactoren. Bovendien, om te beter onderzoeken de effecten van het milieu triggers op de progressie van de advertentie, het is essentieel te scheiden van de individuele milieufactoren en kwantificeren van de mate van blootstelling.
We hebben een nieuwe rat model van AD geïnduceerd door neonatale capsaïcine behandeling aangetoond. Deze advertentie model3 heeft sommige verdiensten ten opzichte van andere AD-modellen zoals de modellen van de AD geïnduceerd door overgevoeligheid van de epicutaneous, de genetisch gemanipuleerde AD-modellen en de spontane Muismodellen van AD4. Eerst, ontwikkelt dit model AD-achtige symptomen op de leeftijd van 3 weken, die correspondeert met baby AD en ouder is dan andere spontane Muismodellen van advertentie zoals de NC/Nga muis. In sommige gevallen, in tegenstelling tot de genetisch gemanipuleerde AD modellen, de symptomen van de AD-modellen verdwijnen en terugvalpreventie spontaan over een periode van 20 weken. Bovendien, het proces van de inductie van AD-achtige symptomen in het model is eenvoudiger dan die van het epicutaneous sensibilisatie model, dat meer vergelijkbaar met de allergische contactdermatitis in het perspectief van de inductie-proces is.
Met behulp van een acryl glas-kamer en een conventionele medische zuurstof regelgever5 aan het bestuderen van de effecten van FA inhalatie betreffendedeontwikkeling van AD in infantiele en adolescent periodes hebben we het nieuwe protocol voor FA blootstelling aan een dierlijk model geïntroduceerd. De regelgever heeft konden we bloot van de dieren een constante concentratie van het gas door het aanpassen van het debiet en de concentratie van de oplossing. Eerdere studies hebben verschillende manieren van blootstelling van FA aan diermodellen gemeld. Xu et al. beschreven de methode van de blootstelling in welke FA oplossing voor het rechtstreeks op de huid6is toegepast. Een andere recente studie gericht het protocol van FA gas blootstelling met behulp van een gespecialiseerde vluchtige oplosmiddelen generator, model 49127. Vergeleken met de methoden van eerdere studies, de nieuwe methode gemakkelijker is uitgerust en kan worden toegepast voor andere oplossingen naast FA.

Atopische dermatitis is chronisch relapsing pruritic eczeem en heerst over de hele wereld vooral in ontwikkelde landen. Complexe interacties tussen genetische en omgevingsfactoren is bekend dat een belangrijke rol spelen in de pathofysiologie van atopische dermatitis. Echter wij nog steeds een gebrek aan een gedetailleerd beeld van de pathogenese van de ziekte. Het is dus van belang voor het ontwikkelen van passende dierlijke modellen voor het ophelderen van de progressie van atopische dermatitis. Bovendien breidt het onderzoeken van het effect van milieufactoren zoals luchtverontreiniging op atopische dermatitis begrip van de ziekte. Hier beschrijven we een methode voor het inducerende atopische dermatitis bij ratten met neonatale capsaïcine behandeling en een protocol voor Gasbedwelming met behulp van een constante concentratie van formaldehyde ratten te onthullen van de effecten op de ontwikkeling van atopische dermatitis in infantiele en adolescent perioden. Deze protocollen zijn met succes toegepast op verschillende experimenten en kunnen worden gebruikt voor andere stoffen.

De AD-achtige huidontsteking van de capsaïcine behandeld AD rat model op 7 weken van leeftijd wordt gepresenteerd in figuur 1A.
