For å avhøre effekten av formaldehyd (FA) innånding på atopisk dermatitt (AD) progresjon, vi etablert en protokoll for AD induksjon og utviklet en konvensjonell sirkulasjon kammer for kronisk eksponering av FA.
Annonsen er en kronisk inflammatorisk hudsykdom knyttet til komplekse interaksjoner mellom genetisk disposisjon og miljømessige utløsere1,2. En skikkelig dyremodell må studere annonse i preklinisk staten. Dette er de viktigste når Klargjørende patogenesen av AD påvirket av miljøfaktorer. Videre bedre undersøke effekten av miljømessige utløsere på AD progresjon, er det viktig å skille de enkelte miljøfaktorer og kvantifisere eksponeringsgraden.
Vi har vist en roman rotte modell av AD indusert av neonatal capsaicin behandling. Denne Annonsen modell3 har noen fortjeneste over andre AD modeller som AD modeller av epicutaneous allergi, genmodifiserte AD modeller og spontan musen modeller av AD4. Først utvikler denne modellen AD-lignende symptomer i en alder av 3 uker, som tilsvarer spedbarn AD og er tidligere enn andre spontan musen modeller av Annonsen som NC/Nga musen. I noen tilfeller, i motsetning til genmodifiserte AD modeller symptomene på AD modellene avta og tilbakefall spontant over en 20-ukers periode. Videre er induksjon prosessen med AD-lignende symptomer i modellen lettere enn den epicutaneous sensibilisering modellen, som er mer lik den allergisk kontakteksem i perspektiv av induksjon prosessen.
Vi har innført romanen protokollen for FA eksponering for en dyr modell med en akryl glass chamber og konvensjonell medisinsk oksygen regulator5 for å studere virkningene av FA innånding på utvikling av AD i infantile og unges perioder. Regulatoren har tillatt oss å avsløre dyr til en konstant konsentrasjon av gass ved å justere flyt priser og løsning konsentrasjonen. Tidligere studier har rapportert flere måter å eksponering for FA dyremodeller. Xu et al. beskrevet metoden eksponering i som FA løsning var direkte brukes til huden6. En fersk studie adressert protokollen FA gass eksponering ved hjelp av en spesialisert flyktige løsemidler generator, modell 49127. Sammenlignet med metoder for tidligere studier, den nye metoden er lettere utstyrt og kan brukes for andre løsninger i tillegg til FA.

Atopisk dermatitt er kronisk relapsing pruritic eksem og råder over hele verden spesielt i utviklede land. Komplekse interaksjoner mellom genetiske og miljømessige faktorer er kjent for å spille en viktig rolle i Patofysiologien ved atopisk dermatitt. Men mangler vi fortsatt et detaljert bilde av patogenesen av denne sykdommen. Dermed er det viktig å utvikle riktige dyr modeller for Klargjørende progresjon av atopisk dermatitt. Videre utvider undersøker effekten av miljømessige faktorer som luftforurensende på atopisk dermatitt forståelse av sykdommen. Her beskriver vi en metode for å indusere atopisk dermatitt i rotter neonatal capsaicin behandling og en protokoll for eksponering for en konstant konsentrasjon av formaldehyd rotter å avsløre effekter på utvikling av atopisk dermatitt i infantile og ungdom perioder. Disse protokollene er vellykket brukt til flere eksperimenter og kan brukes for andre stoffer.
