Es wurde eine Vielzahl von Super-Auflösungs-Mikroskopietechniken entwickelt, um die Beugungsgrenze der Lichtmikroskopie (~ 200 nm) zu überwinden. Darunter befindet sich eine Kategorie von Techniken, die als Singlemolekül-Lokalisierungsmikroskopie (SMLM) bezeichnet werden, die eine Photoaktivierungslokalisierungsmikroskopie (PALM) und eine stochastische optische Rekonstruktionsmikroskopie (STORM) umfasst. SMLM-Techniken teilen sich bei der Verwendung von Fluorophoren, die zwischen auf (fluoreszierenden) und aus (dunkel / foto geschalteten) Zuständen umgeschaltet werden können, was eine sequentielle Lokalisierung der Fluoreszenz von einzelnen Molekülen 1 , 2 , 3 ermöglicht . Aufgrund seiner Kompatibilität mit handelsüblichen Farbstoffen und Mikroskopen ist direkt STORM (dSTORM) zu einer weit verbreiteten SMLM-Technik geworden 4 . DSTORM kann routinemäßig ~ 10 nm Lokalisierungsgenauigkeit erreichen, definiert als die Unsicherheit bei der Berechnung des Zentrums eines BeugensIonen-begrenzte Punktspreizfunktion (PSF). Trotz der hohen Genauigkeit, die mit den Lokalisierungsalgorithmen 5 , 6 , 7 geschätzt wird , wurde eine genaue Bestimmung der tatsächlichen Lage einzelner Moleküle durch eine Reihe von Problemen behindert. Zunächst fügt eine mechanische Bewegung des Mikroskopstadiums während der Bildaufnahme eine erhebliche Unsicherheit zur Lokalisierungsgenauigkeit hinzu. Da SMLM-Bilder über Tausende von Zeitraffer-Frames erhalten werden, können nanoskalige Bewegungen des Mikroskopstadiums die Präzision des endgültigen Superauflösungsbildes erheblich beeinträchtigen 8 . Um die Bühnenbewegung während der Bilderfassung zu kompensieren, wird die Bühnendrift üblicherweise aus der regressionsbasierten Anpassung von Binned-Lokalisierungen aus dem Bild selbst (Kreuzkorrelation) oder sequentiellen Lokalisierungen von fiducial-Markern (Referenzkorrektur) 1 , 9 geschätzt. HowevDiese Methoden erfordern die Optimierung mehrerer Parameter für jeden Bildstapel und können keine Bühnenbewegungen bei kurzen Zeitskalen wie mechanische Vibration berücksichtigen. Gold-Nanopartikel und mehrfarbige fluoreszierende Perlen wurden als Fiducial-Marker in SMLM verwendet, sind aber nicht lichtstabil und führen zu einer deutlich geringeren Präzision nach Driftkorrektur als die eingesetzten Stickstoff-Vakanz-Zentrum-Fluoreszenz-Nanodiamanten (FNDs) In madSTORM 10 Neben der Beugungsgrenze wird die Lichtmikroskopie durch spektrale Grenzwerte weiter eingeschränkt. Die gleichzeitige Visualisierung mehrerer Targets erfordert fluoreszierende Sonden mit nicht überlappenden Spektralprofilen, die in der Regel die Fluoreszenz-basierte Lichtmikroskopie auf 6 Farben und SMLM auf 2-3 Farben 4 , 11 , 12 beschränken . Darüber hinaus verursacht eine nichtlineare chromatische Aberration eine Fehlausrichtung von mehrfarbigen Bildern, wDie umfangreiche Ausrichtungsverfahren mit mehrfarbigen Markierungen 8 , 13 erfordern. Um diese Grenzen zu überwinden, haben frühere Studien mehrere Targets unter Verwendung eines wiederholten Photobleichens oder chemischen Quenchens von sequentiell gebundenen Fluorophoren 14 , 15 , 16 , 17 , 18 , 19 abgebildet. Während diese Verfahren die spektralen Grenzen der Mikroskopie überwinden können, ist das Fluoreszenzbleichen bekannt als ein toxisches Verfahren 20 , und ein verlängertes Bleichen oder Abschrecken kann unerwünschte Nebenwirkungen wie den Verlust der Vernetzung verursachen. Darüber hinaus könnte die Anhäufung von fluoreszierenden Sonden zu einer sterischen Blockierung von Bindungsstellen in der Probe führen, was ein großflächiges Multiplexen und ein robustes Targeting von Epitopen verhindert. Um eine solche sterische Störung zu vermeiden,Tudy erreichte Multiplexing mit stochastischem Austausch von frei diffundierenden Proteinfragmenten 21 . Während dieses Verfahren eine dichte Markierung von zellulären Strukturen ermöglicht, erfordert es eine umfangreiche biochemische Präparation, um Peptidfragmente zu isolieren, kann keine Einzelmolekülpositionen lokalisieren und erleichtert nicht leicht das Multiplexen mit großem Maßstab unter Verwendung von handelsüblichen Sonden. Wir präsentieren ein detailliertes Video-Protokoll, das die sequentielle Bindung und Elution von fluoreszierenden Antikörpern für gemultiplexte, antikörpergrßenbeschränkte dSTORM (madSTORM) -Bildgebung und die Verwendung von fluoreszierenden Nanondiamanten beschreibt, um eine präzise Driftkorrektur und Ausrichtung zu erreichen.

Die Imaging heterogener zellulärer Strukturen unter Verwendung einer Einzelmolekül-Lokalisierungsmikroskopie wurde durch unzureichende Lokalisierungsgenauigkeit und Multiplexfähigkeit behindert.
