Den sekventielle multipleksering, driftskorrigering og justeringsprosedyrene i madSTORM tillater nøyaktig, høymultiplexert visualisering av heterogene strukturer i celler. 10 Videre unngår madSTORM begrensningene i flerfarget STORM, for eksempel kromatisk avvikelse og suboptimale fotoomkoblings- / utslippsegenskaper for ikke-fargerøde fargestoffer 9 , 12 . Ettersom elueringstrinnet signifikant reduserer sterisk interferens fra sekvens...

En rekke superoppløsningsmikroskopiteknikker har blitt utviklet for å overvinne diffraksjonsgrensen for lysmikroskopi (~ 200 nm). Blant disse er en kategori teknikker kalt single molecule localization microscopy (SMLM) som inkluderer fotoaktivering lokaliseringsmikroskopi (PALM) og stokastisk optisk rekonstruksjonsmikroskopi (STORM). SMLM teknikker deler i bruk av fluoroforer som kan byttes mellom på (fluorescerende) og av (mørke / fotobrytede) tilstander, slik at sekvensiell lokalisering av fluorescens fra enkeltmolekyler 1 , 2 , 3 . På grunn av kompatibiliteten med kommersielt tilgjengelige farger og mikroskoper, har direkte STORM (dSTORM) blitt en allment vedtatt SMLM-teknikk 4 . DSTORM kan rutinemessig oppnå ~ 10 nm lokaliseringspresisjon, definert som usikkerheten ved beregning av sentrum for en diffraksjonIon-begrenset punkt spredning funksjon (PSF). Til tross for den høye presisjonen som ble estimert ved hjelp av lokaliseringsalgoritmer 5 , 6 , 7 , har nøyaktig bestemmelse av den faktiske plasseringen av enkeltmolekyler blitt hindret av en rekke problemer. For det første legger mekanisk bevegelse av mikroskopstadiet under bildeoppkjøpet betydelig usikkerhet til lokaliseringspresisjon. Som SMLM-bilder er oppnådd over tusenvis av tidsrammer, kan nanoskala bevegelser av mikroskopetrinnet betydelige kompromisser med presisjonen til det endelige superoppløsningsbildet 8 . For å kompensere for scenebevakning under bildeoppkjøpet, estimeres trinndrift vanligvis fra regresjonsbasert montering av inndata lokaliseringer fra selve bildet (krysskorrelasjon) eller sekvensielle lokaliseringer fra fiducial-markører (fiducial-korreksjon) 1 , 9 . HowevEr, disse metodene krever optimalisering av flere parametere for hver bildebunke, og kan ikke regne for scenebevegelser på kort tidskalaer som mekanisk vibrasjon. Gullnano-partikler og flerfargede fluorescerende perler har blitt brukt som fiducial markører i SMLM, men de er ikke fotostabile, og resulterer i betydelig lavere presisjon etter driftskorreksjon enn nitrogen-fluorescerende nanodiamanter (FNDs) I madSTORM 10 . I tillegg til diffraksjonsgrensen blir lysmikroskopi ytterligere begrenset av spektrale grenser. Samtidig visualisering av flere mål krever fluorescerende sonder med ikke-overlappende spektrale profiler, generelt begrenser fluorescensbasert lysmikroskopi til 6 farger og SMLM til 2-3 farger 4 , 11 , 12 . Videre forårsaker ikke-lineær kromatisk avvikelse feiljustering av flerfargete bilder, wSom krever omfattende justeringsprosedyrer ved bruk av flerfarget fiducial markører 8 , 13 . For å overvinne disse grensene har tidligere studier avbildet flere mål ved bruk av repeterende fotoblekking eller kjemisk slukning av sekvensielt bundet fluoroforer 14 , 15 , 16 , 17 , 18 , 19 . Selv om disse metodene kan overvinne spektralgrensene for mikroskopi, er fluorescensbleking kjent for å være en toksisk prosess 20 , og langvarig bleking eller quenching kan forårsake uønskede bivirkninger som tverrbinding. Videre kan akkumuleringen av fluorescerende prober føre til sterisk blokkering av bindingssteder i prøven, for å hindre storskala multipleksering og robust målretting av epitoper. For å unngå slike steriske forstyrrelser, en siste sTudy oppnådd multipleksering ved bruk av stokastisk utveksling av fritt diffuserende proteinfragmenter 21 . Mens denne metoden tillater tett merking av cellulære strukturer, krever det omfattende biokjemisk preparat for å isolere peptidfragmenter, kan ikke lokalisere enkeltmolekylposisjoner og letter ikke lettere storskala-multipleksering ved bruk av kommersielt tilgjengelige prober. Vi presenterer en detaljert videoprotokoll som beskriver sekvensiell binding og eluering av fluorescensantistoffer for multiplexert, antistoffstørrelsesbegrenset dSTORM (madSTORM) avbildning, og bruk av fluorescerende nanodiamanter for å oppnå nøyaktig driftskorrigering og justering.

Imaging heterogene cellulære strukturer ved bruk av enkeltmolekyl lokaliseringsmikroskopi er blitt hindret av utilstrekkelig lokalisering presisjon og multiplexing evne. Ved hjelp av fluorescerende nano-diamant fiducial markører, beskriver vi driftskorrigerings- og justeringsprosedyrene som kreves for å oppnå høy presisjon i enkeltmolekyler lokaliseringsmikroskopi. I tillegg er en ny multiplekseringsstrategi, madSTORM, beskrevet hvor flere molekyler er målrettet i samme celle ved bruk av sekvensiell binding og eluering av fluorescerende antistoffer. MadSTORM er demonstrert på en aktivert T-celle for å visualisere plasseringen av forskjellige komponenter i en membranbundet, multi-proteinstruktur kalt T-celle-reseptor-mikroklassen. I tillegg diskuteres anvendelse av madSTORM som et generelt verktøy for visualisering av multi-protein strukturer.

Den sekventielle eluerings- og fargemetode ble brukt til å produsere det multiplekserte madSTORM-bildet av mikroklustere og andre strukturer i en aktivert Jurkat T-celle ( figur 1 , kontrollfigurjusteringer).
