Işık mikroskopisinin (~ 200 nm) kırınım sınırını aşmak için çeşitli süper-çözünürlüklü mikroskopi teknikleri geliştirilmiştir. Bunların arasında, foto-aktivasyon lokalizasyonu mikroskopisi (PALM) ve stokastik optik rekonstrüksiyon mikroskopisi (STORM) içeren tek molekül lokalizasyon mikroskopisi (SMLM) adı verilen bir teknik kategorisi bulunmaktadır. SMLM teknikleri, (floresan) ve kapalı (karanlık / foto-anahtarlamalı) durumlar arasında değiştirilebilen, floresanın tekli moleküller 1 , 2 , 3&#39;den sıralı lokalizasyona olanak tanıyan flüorofor kullanımını paylaşır. Ticari olarak mevcut boyalar ve mikroskoplar ile uyumluluğu nedeniyle doğrudan STORM (dSTORM) yaygın bir şekilde benimsenen SMLM tekniği haline gelmiştir. DSTORM, bir kırınım merkezinin hesaplanmasında belirsizlik olarak tanımlanan ~ 10 nm lokalizasyon hassasiyetini rutin olarak gerçekleştirebilirIyon sınırlı nokta yayılım fonksiyonu (PSF). Bununla birlikte, lokalizasyon algoritmaları 5 , 6 , 7 kullanılarak yüksek hassasiyet tahmini yapılmasına rağmen, tek moleküllerin gerçek konumunun doğru olarak belirlenmesi bir takım hususlarla engellenmiştir. SMLM görüntüleri binlerce zaman aşımı karesinden elde edildiğinde, mikroskop kademesinin nano ölçekli hareketi, son süper-çözünürlüklü görüntünün 8 hassasiyetini önemli ölçüde tehlikeye atabilir. Görüntü yakalama işlemi sırasında sahne hareketini telafi etmek için sahne kayması yaygın olarak, kendiliğinden yerleşimlerin resmin kendisinden regresyon temelli uydurulması (çapraz korelasyon) veya referans işaretlerinden sıralı lokalizasyonlar (referans düzeltme 1 , 9 ) ile tahmin edilir. ancak buradanBu yöntemler, her görüntü yığıtı için birden fazla parametrenin optimizasyonunu gerektirir ve mekanik titreşim gibi kısa zaman ölçeklerinde sahne hareketlerini hesaba katamaz. SMLM&#39;de, altın nano parçacıkları ve çok renkli flüoresan boncuklar referans işaretleyiciler olarak kullanılıyor ancak foto-kararlı değiller ve sürüklenme düzeltildikten sonra kullanılan azot boşluğu-merkez flüoresan nano-elmas (FND) değerlerinden çok daha düşük hassasiyetle sonuçlanıyorlar MadSTORM 10&#39;da . Kırılma sınırına ek olarak, ışık mikroskopisi spektral sınırlarla daha da sınırlandırılmıştır. Birden fazla hedefin eşzamanlı olarak görselleştirilmesi, örtüşmeyen spektral profilleri olan flüoresan sondaları gerektirir ve genellikle 6 renk ve SMLM&#39;den flüoresan esaslı ışık mikroskopisi 4 , 11 , 12&#39;ye kısıtlanır. Ayrıca, lineer olmayan renk sapmaları, çok renkli görüntülerin hizalamasına neden olur, wÇok renkli referans işaretleri 8 , 13 kullanarak geniş hizalama prosedürleri gerektirir. Ayrıca, flüoresan probların birikimi, numunedeki bağlanma alanlarının sterik olarak bloke edilmesine ve epitopların geniş çaplı çoğullama ve sağlam hedeflemesine engel olabilir. Bu tür sterik parazitlerden kaçınmak için, son zamanlardaTitizlikle serbestçe yayılmakta olan protein parçalarının stokastik değişimi kullanılarak çoğullama elde edilmiştir 21 . Bu yöntem, hücresel yapıların yoğun şekilde etiketlenmesine izin verirken, peptid parçalarını izole etmek için kapsamlı biyokimyasal hazırlık gerektirir, tek molekül konumlarını bulamaz ve piyasada bulunan problar kullanılarak büyük ölçekli çoğullamayı kolayca kolaylaştırmaz.
