Hemos desarrollado un modelo clínico relevante totalmente funcional de CPC en un ratón. Con más de 30 cepas de ratones con enfermedades cardiovasculares, nuestro modelo podría ser un punto de partida para el desarrollo de nuevos protocolos prospectivos relacionados con el CPB. Desde la introducción en la clínica de puente cardiopulmonar (CPB), jugó un papel esencial en la cirugía cardíaca1. En la cirugía cardiaca moderna, tiempo de CEC prolongado es esencial para llevar a cabo reconstrucciones aórticas extensa y procedimientos combinados. Aunque los avances tecnológicos han sido tremendos, el uso de circulación extracorpórea se asocia con intra y postoperatorias sistémicas y daño de órgano local2,3.
Grandes modelos animales se han desarrollado para investigar el papel de la CPB en procesos fisiológicos4,5. Aunque estos modelos han proporcionado una visión de la CPB complicaciones asociadas, son extremadamente costosos y herramientas moleculares (p. ej., anticuerpos) son muy limitadas. Una alternativa más costo-eficiente se ha desarrollado en pequeños animales. Desde su desarrollo, se han realizado múltiples estudios para optimizar un modelo CPB en ratas y conejos5,6,7,8,9. Estos modelos proporcionan una buena base para la medición de los procesos fisiopatológicos de la enfermedad; sin embargo, son todavía insuficientes para investigar Inmunología celular y humoral debido a la falta de los correspondientes anticuerpos y reactivos. Esto deteriora su papel en este campo de investigación.
Recientemente hemos desarrollado un modelo de ratón de CPB. Debido a una gran variedad de reactivos específicos de ratón y ratones genéticamente modificados, modelos de ratón son en general el modelo de elección para la investigación fisiológica, molecular e inmunológica10,11. Por lo tanto, nuestro modelo facilitará el estudio de la CPB en relación con diversas comorbilidades ya que hay muchas cepas de ratones con enfermedades clínicamente relevantes12,13. Por consiguiente, este artículo describe, en detalle, cómo realizar CPB en ratones. Oxígeno y parámetros hemodinámicos son vigilados de cerca después de la detención de la profundidad respiratoria y circulatoria.

cardiopulmonary bypass in a mouse model: a novel approach

Como bypass cardiopulmonar prolongada se vuelve más esencial durante intervenciones cardiológicas, se presenta una demanda creciente de la clínica para la optimización del procedimiento y para reducir al mínimo daño a los órganos resultantes de la circulación extracorpórea prolongada. El objetivo de este trabajo fue demostrar un modelo completamente funcional y clínicamente relevante de puente cardiopulmonar en un ratón. Divulgamos sobre el diseño del dispositivo de optimización de circuito de perfusión y técnicas microquirúrgicas. Este modelo es un modelo agudo, que no es compatible con la supervivencia debido a la necesidad de múltiples dibujos de sangre. Debido a la gama de herramientas disponibles para los ratones (p. ej., marcadores, agujeros ciegos, etc.), este modelo facilitará la investigación sobre los mecanismos moleculares de daño del órgano y el efecto de puente cardiopulmonar en relación con otros comorbilidades.

Este protocolo describe el circuito de perfusión, procedimientos quirúrgicos y el seguimiento de los parámetros fisiológicos durante la CEC de un ratón. Cuando es realizada por un microsurgeon adecuadamente capacitado, los resultados son consistente y reproducible obtenidos.
