Weefselfusie is een belangrijke stap in de ontwikkeling van meerdere organen. Grote menselijke geboorteafwijkingen zoals gespleten lip en verhemelte, spina bifida en misvormingen van het hart kunnen het gevolg zijn van gebreken in weefselfusie 1 . Muis secundaire palet fusie is uitgebreid bestudeerd om de cellulaire en moleculaire mechanismen die weefselfusie in ontwikkeling 2 , 3 , 4 bepalen, te identificeren. In de muis begint secundaire paletontwikkeling bij ongeveer E11.5 met de uitgroei van een secundaire palatale plank uit elk van de bilaterale maxillaire processen. De initiële groei van de palatale planken vindt verticaal langs de tong plaats, tot ongeveer E14.0, op dat moment worden de palatale planken horizontaal boven de tong verhoogd. Mediaal gerichte groei resulteert in fysiek contact tussen de toepassende epithelia van de twee palatale planken, waardoor de middelste epithelie vormtAl Seam (MES) op E14.5. De tussenliggende MES moet verwijderd worden tussen de secundaire palatale planken om mesenchymale samenvloeiing mogelijk te maken en de ontwikkeling van een intact, volledig gesmolten secundair verhemelte door E15.5 3 te ontwikkelen . Hoe is een gedeelde epitheliale MES cellaag gevormd tussen twee afzonderlijke palatale planken, en vervolgens verwijderd om mesenchymale samenvloeiing te bereiken, is een centrale vraag in de ontwikkeling van de verhemelte. Gebaseerd op histologische en elektronenmicroscopie (EM) studies, explorante cultuurstudies en functionele muisgenetische experimenten, zijn er in dit proces verschillende fundamentele celgedragen betrokken. Filopodia-achtige projecties van het mediale randepitheel MEE van elke palatale plank vergemakkelijkt het eerste contact 5 , 6 , gevolgd door intercalatie van deze epitheliale cellen naar een gedeelde single MES 6 , 7 . Verwijdering van de resulterende gedeelde MESIs voorgesteld om door drie niet-exclusieve mechanismen te gaan. Vroege studies waarbij histologische observaties en ex vivo lijnsporen met vitale kleurstoffen werden gebruikt, wijzen erop dat het MES kan worden verwijderd door epitheliale tot mesenchymale overgang (EMT) van MES-cellen 8 , 9 , alhoewel recentere genetische afstamming van epitheliale cellen onzekerheid heeft opgedaan met betrekking tot De langetermijnbijdrage van epitheelcellen aan de palatale plank mesenchymme 10 , 11 , 12 . Ten slotte, oorspronkelijk gebaseerd op studies met epitheliale etikettering en statische observatie bij progressieve tijdpunten, MES-cellenWerden voorgesteld om te migreren in de oronasale en anteroposterior afmetingen 11 , 13 , maar dergelijke dynamische celgedragen waren aanvankelijk onbevestigd doordat ze niet in staat waren om ze in levend palatale weefsel te observeren. Onlangs waren we in staat om deze gedragingen direct te observeren door een nieuwe live imaging methodologie te ontwikkelen die muisgenetische methoden van fluorescerende etikettering combineert met confocale livebeelden van explanted palatale planken. Ten eerste, voor het visualiseren van dynamisch cellulair gedrag in gomepitheelcellen tijdens de gomfusie , genereren we een epitheliale-specifieke reportermuis door ROSA26-mTmG floxmuizen met Keratin14-cre muizen 14 , 15 te oversteken. Confocale levende beeldvorming van de verhemelte-explorante cultuur van de resulterende embryo&#39;s bevestigde een aantal eerder voorgestelde cellulaire gedragingen en geïdentificeerde nieuwe gebeurtenissen in het fusieproces 6 </sup&gt;. Epitheliale celmembraan uitsteeksels voorafgegaan aan initieel cel-celcontact gevolgd door epitheelconvergentie door celintercalatie en oronasale celverplaatsing. In het bijzonder ontdekten we dat cel extrusie, een proces dat belangrijke rollen speelde in epitheliale homeostase, een belangrijk mechanisme was om muis secundaire paletfusie 6 , 16 te beheren. Deze beeldvormingsmethode kan gebruikt worden bij andere reporterlijnen; We hebben Lifeact-mRFPruby transgene muizen 17 , 18 gebruikt om actin cytoskeletale dynamiek te onderzoeken tijdens het fusieproces. Andere verslaggevers kunnen ook worden gebruikt om andere specifieke aspecten van verhemeltefusie te observeren en deze methode kan worden aangepast aan laserscanning-confocal microscopie of draaibare confocal microscopie, afhankelijk van beeldbehoeften en beschikbaarheid van microscoop. Live imaging wordt steeds meer een keystone-aanpak in ontwikkelingsbiologie. PartiCulariofaciale morfogenese is complex en menselijke geboorteafwijkingen die het gezicht beïnvloeden zijn gebruikelijk. Deze confocal live imaging methode zal bijdragen tot een beter inzicht in onderliggende basisontwikkelingsmechanismen, alsmede de oorsprong van menselijke craniofaciale afwijkingen.

Hier beschrijven we een protocol voor livebeelden van ex vivo secundaire paletfusie in muisembryo&#39;s. Voor het visualiseren van filamenteactine werden Lifeact-mRFPruby- reportermuizen gebruikt. Live beeldvorming van secundaire verhemelingsfusie werd uitgevoerd door dissecteren van recent gehandhaafde secundaire palatale planken van embryonale dag (E) 14.5 stadium embryo&#39;s en culturen in agarose bevattende media op een glasbodemschotel om beeldvorming met een omgekeerde confocale microscoop mogelijk te maken. Met behulp van deze methode hebben we een verscheidenheid aan nieuwe cellulaire gedragingen ontdekt tijdens secundaire paletfusie. Een waardering voor het onderscheidend gedrag van de cellen in ruimte en tijd draagt ​​bij aan ons inzicht in dit dynamische morfogenetische proces.
