Vi tog fordel af denne funktion til systematisk spørge, på tværs af hele gær ORFeome, hvis stabil protein-baseret, epigenetiske stater kunne indledes af forbigående inducerende overekspression af individuelle proteiner. Kodning af biologiske oplysninger, der er tilgængeligt for fremtidige generationer er generelt opnået via ændringer i DNA-sekvens. Langlivede arv kodet i protein kropsbygning (snarere end sekvens) har længe været set som paradigme-skift, men sjældne. De bedste karakteriserede eksempler på sådanne epigenetiske elementer er prioner, som besidder en selvsamlende adfærd, der kan drive den arvelige manifestation af nye fænotyper. Mange arketypiske prioner vise en slående N/Q-rige sekvens bias og samle til en amyloid fold. Disse usædvanlige egenskaber har informeret de fleste screening bestræbelserne på at identificere nye prion-proteiner. Dog havnen mindst tre kendte prioner (herunder den stiftende prioner, PrPSc) ikke disse biokemiske egenskaber. Vi har derfor udviklet en alternativ metode til at probe omfanget af protein-baserede arv baseret på en ejendom massevirkningsloven: den forbigående overekspression af prioner proteiner øger den hyppighed, hvormed de erhverve en self-templating kropsbygning. Dette papir beskriver en metode til at analysere gær ORFeome evne til at fremkalde proteinbaseret arv. Bruger denne strategi, vi tidligere har konstateret, at &gt; 1% af gær proteiner kunne brændstof fremkomsten af biologiske træk, der var langlivet og stabil, og rejste sig oftere end genetiske mutation. Protein overexpression is known to dramatically alter cellular phenotypes27. Indeed, with an initial screening approach, hundreds of new phenotypes were reproducibly recovered from the overexpression of clones from the yeast ORFeome using just ten stressors.
