De eerste gist prionen werden geïdentificeerd door hun ongewone fenotypes en verbijsterende patronen van overerving. Biologische systemen ervaren vaak voorbijgaande fluctuaties in eiwit overvloed. Het is onduidelijk of deze hebben een blijvende invloed bij het vormgeven van het fenotype van een organisme of van de toekomstige generaties. De bekendste voorbeelden van deze biologie betrekken een zeldzame klasse van eiwitten, prionen, die het ontstaan van erfelijke eigenschappen genoom ongewijzigd rijden. In plaats daarvan zenden deze proteinaceous en infectious deeltjes fenotypen via zichzelf onderhoudende wijzigingen aan eiwit conformatie1,2. Dit soort erfenis werd ontdekt als de oorzaak van de ongewone erfenis patronen van een verwoestende neurodegeneratieve ziekte. Echter, studies in organismen variërend van schimmels aan zoogdieren3,4,5,6,7,8,9,10 aangezien is gebleken dat het prion-achtige elementen adaptieve waarde kunnen verlenen. Prionen hebben echter werd bekeken als een fascinerende maar zeldzame biologische eigenaardigheid.

Echter, omdat deze benaderingen over het algemeen op sterke aminozuur reeks vooroordelen gericht hebben, delen de prionen die zijn ontdekt de biochemische eigenschappen van de stichtende gist prionen [PSI+]13,14, [URE3]15en1611,[RNQ+]. Deze omvatten: 1) modulaire domeinen die rijk zijn aan lang polymere stukken van asparagine (N) en glutamine (Q), 2) vergadering in een amyloïde [PRION+] conformatie17,18,19, en 3) volledige afhankelijkheid van disaggregase Hsp104 functie voor trouwe vermeerdering van moeder op dochter13,20,21. Inderdaad, vele bona fide prionen, waaronder [GAR+], [Het-s], en zelfs de oorspronkelijke prion (PrPSc), onder dergelijke strenge criteria zouden worden gemist. Misschien nog belangrijker, zou ze kunnen vangen elke nieuwe mechanismen van overname op basis van eiwitten22. De echte biologische breedte van dergelijke verschijnselen kunnen dus veel vaker in de natuur dan eerder aangenomen.
Om te onderzoeken van deze vraag, een high-throughput, Proteoom-brede strategie werkte. Een kenmerk van alle prionen, met inbegrip van PrPSc, [GAR+], en [Het-s], is dat de voorbijgaande overexpressie van de causale eiwitten sterk verhoogt het tarief van prion overname15,23,24,25,26. We profiteerde van deze functie te systematisch vragen, over de gehele gist ORFeome, als stabiel op basis van eiwitten, epigenetische Staten kon worden gestart door Transient inducerende de overexpressie van individuele eiwitten. Het is bekend dat eiwit overexpressie fenotypen27kunt wijzigen. Prion-eiwitten zijn echter ongebruikelijk omdat hun tijdelijke overproductie produceert een verandering in fenotype thats erfelijke voor vele honderden generaties na de initiële overexpressie. Eerder hebben we voordeel van deze functie, alsook de ongewone erfenis patronen van op basis van eiwitten genetische elementen te identificeren van tientallen eiwitten die kunnen heritably bedrading opnieuw fenotypische landschappen zonder te veranderen van het genoom28. Hoewel sommige geïdentificeerd waren eiwitten voorheen bekend als prionen, de meeste waren niet onderstrepen van de kracht van deze aanpak te ontdekken van nieuwe vormen van overname op basis van eiwitten.

De codering van biologische informatie die toegankelijk is voor toekomstige generaties wordt over het algemeen bereikt via veranderingen in de opeenvolging van DNA. Ingezien langlevende overname gecodeerd in eiwit conformatie (eerder dan reeks) lang als paradigma-shifting maar zeldzaam. De beste gekarakteriseerd voorbeelden van dergelijke epigenetische elementen zijn prionen, die een zelfassemblerende gedrag dat de erfelijke manifestatie van nieuwe fenotypen rijden kan bezitten. Vele archetypische prionen weergeven een opvallende N/Q-rijke reeks bias en monteren in een amyloïde schoot. Deze ongebruikelijke kenmerken hebben laten weten dat de meeste screening inspanningen te identificeren van nieuwe prion-eiwitten. Ten minste drie bekende prionen (met inbegrip van de stichtende prion, PrPSc) doen echter niet deze biochemische kenmerken haven. Daarom ontwikkelden we een alternatieve methode om de sonde van de werkingssfeer van de op basis van eiwitten overname op basis van een eigenschap van massa-actie: de voorbijgaande overexpressie van prion-eiwitten verhoogt de frequentie waarop zij een zelf-templating bevleesdheid verwerven. Dit document beschrijft een methode voor het analyseren van de capaciteit van de gist ORFeome te verduidelijken met overname op basis van eiwitten. Met behulp van deze strategie, eerder vonden we dat &gt; 1% van gist eiwitten kon brandstof de opkomst van biologische eigenschappen die waren langlevende, stabiel, en vaker dan genetische mutatie is ontstaan. Deze aanpak kan worden gebruikt in hoge-doorvoer over de gehele ORFeomes of als een gerichte screening paradigma voor specifieke genetische netwerken of milieu prikkels. Net zoals voorwaartse genetische schermen talrijke ontwikkelings- en signalering trajecten definiëren, bieden deze technieken een methodologie voor het onderzoek naar de invloed van overname op basis van eiwitten in biologische processen.

Protein overexpression is known to dramatically alter cellular phenotypes27.
