De første gjær prioner ble identifisert av deres uvanlig fenotyper og forvirrende mønstre av arv. Biologiske systemer opplever ofte forbigående svingninger i protein overflod. Om disse har en varig innvirkning på forme fenotypen av en organisme eller fremtidige generasjoner er fortsatt uklart. De mest kjente forekomstene av denne biologi innebære en sjelden klasse av proteiner, prioner, som styrer fremveksten av arvelige egenskaper uten Genova endringer. I stedet overføre disse proteinaceous og ifectious partikler fenotyper via selvtillit perpetuating endringer til protein konformasjon1,2. Denne typen arv ble oppdaget som forårsaker uvanlig arv mønstre av en ødeleggende neurodegenerative sykdom. Imidlertid har studier i organismer mellom sopp pattedyr3,4,5,6,7,8,9,10 siden avdekket at prion-liknende elementer kan gi adaptive verdi. Likevel har prioner vært betraktet som en fascinerende men sjelden biologiske særhet.
Denne rådende visdom holdes delvis fordi karakterisering av protein-basert arv har lenge vært begrenset av et lite sett med eksempler. Siste systematisk screening innsats har utvidet dette bildet betraktelig ved å identifisere flere nye bona fide prioner11 og nesten to dusin protein domener12 med kapasitet til drivstoff prion-lignende conformational konvertering. Men fordi disse har generelt fokuserer på sterk aminosyre sekvens biases, dele prioner som har blitt oppdaget biokjemiske egenskaper av grunnleggerne gjær prioner [PSI+]13,14, [URE3]15og [RNQ+]11,16. For å undersøke dette spørsmålet, var en høy gjennomstrømning og proteom hele strategi ansatt. Et kjennetegn på alle prioner, inkludert PrPSc, [GAR+], og [Het-s], er at det forbigående overuttrykte kausale proteiner sterkt øker frekvensen av prion oppkjøpet15,23,24,25,26. Vi tok nytte av denne funksjonen å systematisk stille, over hele gjær ORFeome, hvis stabil protein-basert, epigenetic stater kan startes ved å indusere transiently overuttrykte av personlige proteiner. Det er velkjent at protein overuttrykte kan endre fenotyper27. Men er prion proteiner uvanlig fordi deres midlertidig overproduksjon produserer en endring i fenotypen som er arvelig for mange hundre generasjoner etter den første overuttrykte. Vi har tidligere tok nytte av denne funksjonen, samt uvanlige arv mønstre av protein-basert genetiske elementer, identifisere dusinvis av proteiner som kan heritably nytt ledningsnett fenotypiske landskap uten å endre de genom28. Selv om noen identifisert var proteiner tidligere kjent som prioner, de fleste var ikke, understreker kraften i denne tilnærmingen å avdekke nye former for protein-basert arv.

Koding av biologiske informasjon som er tilgjengelig for fremtidige generasjoner er generelt oppnås via endringer i DNA sekvensen. Varige arv kodet i protein konformasjon (snarere enn sekvens) har lenge vært betraktet som paradigme-skifter, men sjelden. De beste preget eksemplene på epigenetic elementer er prioner, som har en selvstendig montering atferd som kan kjøre arvelige manifestasjonen av nye fenotyper. Mange arketypiske prioner viser en slående N/Q-rik sekvens bias og sette sammen til en amyloid fold. Disse uvanlige funksjoner har informert om de fleste screening innsats for å identifisere nye prion proteiner. Imidlertid tre kjente prioner (inkludert den opprinnelige prion, PrPSc) ikke havn disse biokjemiske egenskaper. Protein overexpression is known to dramatically alter cellular phenotypes27. Indeed, with an initial screening approach, hundreds of new phenotypes were reproducibly recovered from the overexpression of clones from the yeast ORFeome using just ten stressors.
