O sistema nervoso de Hydra consiste em uma rede nervosa, com neurônios associados a ambas as camadas de tecido epitelial 1 . A rede nervosa é mais densa no hipossênero e pedúnculo e menos densa na coluna 2 do corpo. As células nervosas são originárias de células estaminais intersticiais, que são células-tronco multipotentes que originam células secretoras, nematócitos, células germinativas e neurônios 1 . É possível eliminar as células intersticiais de Hydra vulgaris através do tratamento com colchicina 3 , 4 , uma toxina derivada de plantas que mata células em divisão. Embora a colicicina tenha inibido a polimerização de microtúbulos em outros organismos, um estudo anterior mostrou que os microtúbulos estão presentes na Hydra ao longo de todo o tratamento, sugerindo que a colchicina não age dessa maneira na Hydra 3 . Outra stSugere que a colchicina não se liga de forma eficiente à tubulina em alguns organismos, incluindo Tetrahymena pyriformis, Zea mays, Chlamydomonas e Schizosaccharomyces pombe, o que pode explicar essa diferença 5 . O tratamento com colchicina induz a fagocitose das células intersticiais pelas células epiteliais endodérmicas 3 e, portanto, permite a criação de animais que faltam células nervosas, células da glândula e nematócitos. Não está claro por que as células intersticiais são particularmente suscetíveis ao tratamento com colchicina. Dado que as células intersticiais pós-mitóticas e a linhagem das células estaminais intersticiais são danificadas e fagocitadas, Campbell concluiu que a colicicina não afetou diretamente a atividade mitótica 3 . Notavelmente, o tratamento com colchicina funciona bem em Hydra vulgaris, mas foi demonstrado que não funciona também em outras espécies, como Hydra oligactis 6 . </Em&gt; Um tratamento modificado com colchicina e hidroxiureia pode ser usado para produzir Hydra viridis livre de nervos 7 . A hidra sem nervos (também às vezes referida como &quot; Hydra epitelial&quot; 8 ) é, portanto, uma ferramenta útil para estudar os papéis desses tipos de células especializadas a partir da linhagem celular intersticial na homeostase e regeneração do tecido. Hydra pode ser o único exemplo conhecido de um animal capaz de viver sem um sistema nervoso. Hydra sem nervos serve como um modelo particularmente útil para dissecar o papel da rede nervosa na regulação da regeneração, homeostase e comportamento da Hydra . Por exemplo, a introdução de células intersticiais na hidra livre de nervos através do enxerto permitiu a caracterização da diferenciação de células nervosas como altamente específico da região 9 . Além disso, uma vez que Hydra livre de nervos pode se regenerar, elesPermitir a investigação de caminhos de regeneração independentes do sistema nervoso. Um desses exemplos é a neurogênese apical e a formação da cabeça, que demonstrou depender da função cnox-2 no sistema nervoso em Hydra de tipo selvagem, mas parece ser dispensável na hidra livre de nervos, sugerindo que pode haver um processo alternativo de regeneração da cabeça 10 . A hidra sem nervos também tem sido utilizada para estudar a expressão das células epiteliais e a regulação de genes neurogênicos e de neurotransmissão após a perda de neurogênese 11 . A hidra sem nervos não exibe rajadas de contração espontâneas 12 , indicando que essas explosões são reguladas pelo sistema nervoso. Hydra do Nerve-free, no entanto, contrata em resposta a beliscar a coluna do corpo com fórceps, sugerindo que a contração em resposta a estímulos mecânicos é mediada por acoplamento através deJunções gap gh nas células epiteliais, enquanto o comportamento contrátil espontâneo é mediado pelo acoplamento através de junções gap nas células nervosas 13 . A hidra sem nervos não abre a boca quando é apresentada com alimentos ou glutationa reduzida 3 , sugerindo que os neurônios sensoriais são necessários para detectar a presença de alimentos e sinalizar a boca para abrir. Além disso, a rede nervosa parece desempenhar um papel na detecção da pressão osmótica, porque os animais sem nervos são incapazes de regular de forma autônoma sua pressão hidrostática interna através da abertura da boca, causando a aparência característica do balão 3 , 4 ( Figura 1B ). O regulamento da pressão hidrostática na hidra livre de nervos por deflação manual freqüente levou à perda de alguma morfologia anormal na coluna de hipossetro e corpo. No entanto, a deflação crônica levou à interferência com o crescimento,Organização de articulação, brotação e tecido 8 . Embora a hidra não livre de nervos seja incapaz de alimentar e, por exemplo, sozinha, é possível mantê-los indefinidamente no laboratório manualmente forçando e erguendo cada animal. Publicações anteriores descreveram métodos de alimentação forçada e eructação de Hydra sem nervos, no entanto, esses protocolos envolveram o uso de dicas de micropipetas que devem ser puxadas manualmente para o tamanho apropriado, bem como o uso de um bocal conectado à pipeta pela tubulação 14 . Aqui, é descrito um método de alimentação e burping mais simples, seguro e com maior tempo. Além disso, estudos anteriores envolveram verificar a ausência de células nervosas através da dissociação de animais fixos em células individuais e exame de morfologia celular 3 , 4 , 15 . HAntes, foi utilizada a imuno-histoquímica com um anticorpo monoclonal contra o terminal carboxilo tirosinado da alfa-tubulina como método complementar para maceração para verificar a depleção de neurônios no hiposstoma 13,16. Estudos anteriores mostraram que os neurônios no pedúnculo também podem ser visualizados usando este anticorpo 13 , no entanto, esses neurônios, bem como aqueles na coluna do corpo, são mais difíceis de distinguir. Embora a imuno-histoquímica seja suficiente para confirmar a ausência de células nervosas no hiposstoma e não requer experiência em morfologia do tipo celular, não pode ser usado para verificar a ausência das células estaminais intersticiais e os outros derivados dessas células. Os estudos de dissociação e morfologia celular são mais rigorosos e podem fornecer uma contagem quantitativa dos números de cada tipo de célula que permanece após cada etapa do tratamento.

A linhagem celular intersticial de Hydra inclui células estaminais multipotentes e seus derivados: células da glândula, nematócitos, células germinativas e células nervosas. As células intersticiais podem ser eliminadas através de dois tratamentos consecutivos com colchicina, uma toxina derivada de plantas que mata células em divisão, eliminando o potencial de renovação das células diferenciadas derivadas das células estaminais intersticiais. Os protocolos previamente publicados para a manutenção da Hydra sem nervos envolvem técnicas desatualizadas, como a pipetagem bucal com micropipetas tIps para alimentar e limpar a Hydra . Aqui, é introduzido um protocolo melhorado para a manutenção da hidra livre de nervos. As pinças de ponta fina são usadas para forçar a abertura da boca e inserir Artemia recentemente matada. Após a alimentação forçada, a cavidade do corpo do animal é lavada com meio fresco usando uma seringa e uma agulha hipodérmica para remover material não digerido, aqui referido como &quot;eructo&quot;. The overall goal of this procedure is to generate and efficiently maintain nerve-free hydra by manual force feeding and burping. This method can help answer key questions in the hydra field about the role of the nerve and various behaviors and biological processes. The main advantage of this technique is that it is simpler, safer and more time efficient than previously published protocols for feeding and burping.Using forceps to manipulate the hydra on the microscope can be difficult and frustrating at first, but it quickly gets easier with practice. Begin by adding Artemia cysts to a tall, narrow glass container that tapers at the bottom. Fill the container with Artemia water, taking care not to exceed one gram of cysts per one liter of water for a higher hatch yield.Cover the top of the container with parafilm and insert a 10 milliliter serological pipette through the parafilm, into the container. Taking care that the pipette tip reaches the bottom of the container. Attach a piece of tubing to an aquarium air pump, then fit the tubing around the pipette to provide air ration to the container.When the Artemia have hatched use an Artemia net to strain the shrimp from the culture and wash the animals for about 20 seconds in deionized water. At the end of the wash, transfer the Artemia into a dish with hydra medium. Under a stereo microscope, use a pair of fine forceps to euthanize the shrimp with light squeezing.Placing the freshly sacrificed Artemia close to the hydra to facilitate a fast feeding. Now grasp the first hydra by the foot with one pair of forceps and pinch the body of the hydra with a second pair of forceps to make the hydra contract. Tap the center of the hypostome to make the hydra open its mouth.If the mouth does not open, puncture the center of the hypostome with the forceps and slightly release the pressure on the tips to stretch out the mouth opening made by the puncture. Quickly insert as many Artemia as possible into the hydra's gastric cavity, until the cavity is full and no more Artemia can be inserted without damaging the hydra, accidentally pulling out any Artemia from inside the cavity or preventing the mouth from closing. If the mouth begins to close, stretch the mouth open with the forceps as just demonstrated, taking care that no Artemia already inside the animal, come out.If the hydra is too small for a whole Artemia, cut the shrimp with the scalpel and fit the hydra smaller pieces. If the Artemia does not stay inside the hydra, it maybe necessary to press the forceps against the Artemia, to hold the Artemia inside until the mouth closes. When all of the hydra have been fed, use a glass pasteur pipette to transfer the hydra into a dish with fresh hydra medium, supplemented with rifampicin, taking care to avoid accidental burping.Between eight and 20 hours after the feeding, use P320 or similar grid sandpaper, to smooth down the point of a 30 gauge hypothermic needle until the tip is flat. Attach the needle to a disposable one milliliter plastic syringe and fill the syringe with fresh rifampicin supplemented hydra medium. Place the hydra culture dish under the microscope and use fine forceps to hold the foot of the hydra.Tap at the center of the hypostome with the syringe needle. If the mouth doesn't open, insert forceps into the hypostome to open the mouth as just demonstrated. Next, use the syringe to very gently flush the medium into the gastric cavity, taking care not to blow away the hydra until all of the debris has been expelled.Then, use a glass pasteur pipette to transfer the burped hydra into a dish with fresh rifampicin supplemented hydra medium. Immediately following an initial eight hour culture treatment, all hydra survived. Some animals will have only tentacle stubs remaining, while others will have completely lost their tentacles.About one week following treatment, the hydra will demonstrate signs of small, stub-like tentacle regrowth before their eventual full regeneration, over the next seven days. After a second treatment, about 10%of the original number of animals that have undergone both treatments recover and grow to a size suitable for experimentation. Dual-treated hydra exhibit tentacles that appear thinner than those of untreated animals, have tissue that is more transparent and appear bloated due to the inability to open their mouths to relieve osmotic pressure.The nerve nets in the heads of untreated hydra display distinct fibers that radiate outward from the mouth, as well as obvious cell bodies. While the same fibers are absent in nerve-free hydra after double treatment with culturezine. Further co staining with DAPI reveals a reduced number of nuclei in nerve-free hydra compared to untreated controls, due to the loss of interstitial cell lineage cells induced by dual culture zine treatment.Once mastered, a single hydra feeding can be completed in under one minute if it is performed properly. After watching this video, you should have a good understanding of how to force feed and burp nerve-free hydra.
