Netvlies regeneratie/degeneratie in de zebravis is onderzocht door verschillende benaderingen zoals cytotoxin-gemedieerde cel dood22, mechanische schade23en thermische schade24. We een 532 nm diodelaser om schade aan het netvlies zebrafish in dienst. Daarmee is ons model biedt diverse voordelen. Visie is waarschijnlijk de meest essentiële betekenis van de mens en zijn bijzondere waardevermindering heeft een hoge sociaal-economische gevolgen. In de geïndustrialiseerde wereld goed retinale degeneratieve ziekten voor de meerderheid van gezichtsverlies en blindheid onder de volwassen bevolking1. Retinitis pigmentosa (RP) is de meest voorkomende erfelijke oorzaak van blindheid bij mensen tussen de leeftijden van 20 en 60, die ongeveer 1,5 miljoen mensen wereldwijd2,3. Het is een heterogene familie van overgenomen retinale aandoeningen gekenmerkt door progressieve verlies van de researchdieren (PRs) gevolgd door degeneratie van het retinale pigment epitheel en, vervolgens, gliosis en remodeling van innerlijke neuronen4. Het verloop van de ziekte kan worden verklaard door de incrementele verlies van de twee PR celtypes, meestal beginnend met staven, die verantwoordelijk zijn voor Achromatische visie in het schemerige licht en kegels, die essentieel zijn voor kleur visie en gezichtsscherpte5. Een enkel gen-defect is voldoende om het veroorzaken van RP. Tot nu toe zijn meer dan 130 mutaties in meer dan 45 genen geassocieerd met de ziekte6. Dit leidt tot verschillende fenotypes van de ziekte en is een van de redenen dat gentherapie niet-gegeneraliseerd is en dus een ingewikkelde therapeutische aanpak. Daarom is er dringend behoefte aan de ontwikkeling van nieuwe algemene therapeutische benaderingen voor de behandeling van retinale degenerations in verblindende ziekten.
Retina degeneratie vaak gaat om PR verlies; Dus, de dood van de cel van de PR is een kenmerk van de degeneratieve processen in de retina-7. Het is al gebleken dat de dood van de cel van de PR Müller glia cellen (MC) activering en proliferatie8 stimuleert. MCs, het grote gliale celtype in het netvlies van gewervelde, werden ooit beschouwd als niets meer dan een "lijm" tussen retinale neuronen. In de afgelopen jaren hebben veel studies aangetoond dat MCs fungeren als meer dan louter structurele steun9. Onder de verschillende functies, MCs ook deelnemen aan de neurogenese en repareren van10. Inderdaad, in antwoord op diffusible factoren van de degenererende netvlies, MCs aanzienlijk verhogen gliale fibrillary zuur eiwit (GFAP) expressie. GFAP etikettering kan dus worden gebruikt als een marker voor MC activering als een secundaire reactie op netvlies letsel en degeneratie11.
Onlangs, ontwikkelden we een nieuwe aanpassing van focal letsel met behulp van een laser voor het opwekken van Retina degeneratie in zebrafish (Danio rerio). Focal letsel is voordelig voor de studie van bepaalde biologische processen zoals de migratie van de cellen in de plaats van de gewonden en de precieze timing van gebeurtenissen die tijdens de retinale regeneratie12 plaatsvinden. Bovendien, de zebravis belangrijker geworden in het visuele onderzoek vanwege de overeenkomsten tussen het visuele systeem en dat van andere gewervelde dieren. Bruto morfologische en histologische kenmerken van menselijke en teleost retinae weer enkele verschillen. Dienovereenkomstig, mens en zebrafish retinae bevatten de dezelfde grote cel klassen georganiseerd in gelaagde hetzelfde patroon, waar licht-sensing researchdieren de buitenste laag, bezetten terwijl het netvlies projectie neuronen, de peesknoopcellen, bevinden zich in het binnenste veld neuronale laag, proximale aan de lens. Het netvlies interneuronen, amacrine, bipolaire, en horizontale cellen, localiseren tussen de fotoreceptor en de ganglion cel lagen13. Bovendien is het netvlies zebrafish kegel gedomineerde en dus dichter bij het menselijke netvlies dan, bijvoorbeeld, het intensief bestudeerde knaagdier netvlies. Een fascinerende verschil tussen teleost en zoogdieren is de aanhoudende neurogenese in vis netvlies en netvlies regeneratie na beschadiging. MCs kunnen zebrafish, dedifferentiate en bemiddelen van regeneratie in benadeelde netvlies14,15. In kip hebben MCs sommige capaciteit ook tot Re-Enter de celcyclus en dedifferentiate. Na retinale letsel in volwassen vis, MCs nemen bepaalde kenmerken van voorlopercellen en stamcellen, migreren naar het beschadigde retinale weefsel en produceren van nieuwe neuronen16. Onverwachte overeenkomsten met retinal progenitoren gen expressie profilering van zoogdieren MCs geopenbaard en bewijs voor intrinsieke neurogene potentieel voor MCs in kip, knaagdieren, en zelfs menselijke netvlies groeit17. Echter is waarom de regeneratieve reactie in vogels en zoogdieren is beduidend goedkoper in vergelijking met de robuuste reactie in vis nog niet begrepen. Daarom begrijpen de endogene herstelmechanismes in zebrafish kan stellen voor strategieën voor stimulerende retinale regeneratie bij zoogdieren en mensen. Met betrekking tot de regeling van de endogene reparatie voor MCs als een therapeutisch instrument voor de behandeling van patiënten met Retina degeneratie zou een uitstekende gevolgen voor onze maatschappij hebben.
Hierin bieden we de stappen die nodig zijn om het model van de degeneratie/regeneratie in ophthalmic onderzoek in dienst. We gericht eerst op inducerende focal schade in de hormoonfuncties retina, dan op de beeldvorming van gebeurtenissen op de site van de schade, en ten slotte visualiseren betrokkenheid van de aangrenzende MCs te ontwikkelen. Het algemene protocol is relatief gemakkelijk uit te voeren en een breed scala aan mogelijkheden voor de evaluatie van het netvlies daarna opent.

m&#252;ller glia cell activation in a laser-induced retinal degeneration and regeneration model in zebrafish

Een fascinerende verschil tussen teleost en zoogdieren is het levenslang potentieel van het teleost netvlies voor retinale neurogenese en regeneratie na ernstige schade. Onderzoeken van de regeneratie-trajecten in zebrafish zou brengen nieuwe inzichten te ontwikkelen innovatieve strategieën voor de behandeling van retinale degeneratieve ziekten bij zoogdieren. Hierin zijn we gericht op de inductie van een focal laesie aan het buitenste netvlies in volwassen zebrafish door middel van een 532 nm diodelaser. Een gelokaliseerde blessure kunt onderzoek naar biologische processen die tijdens de retina degeneratie en regeneratie rechtstreeks op het gebied van schade plaatsvinden. Met behulp van niet-invasieve optische coherentie tomografie (OCT), konden we de locatie van het beschadigde gebied en monitor volgende regeneratie definiëren in vivo. Inderdaad, OCT imaging produceert met hoge resolutie, transversale beelden van het netvlies van de zebravis, het verstrekken van informatie die eerder alleen beschikbaar met histologische analyses was. Om te bevestigen de gegevens uit real-time LGO, histologisch secties werden uitgevoerd en regeneratieve reactie na de inductie van de retinale schade werd onderzocht door immunohistochemie.

Real-time OCT: om het analyseren van de rol van MCs in netvlies reparatie, gebruikten we een laser letsel model inducerende een goed afgebakende zone van schade in de zebravis retina. The overall goal of this video is to show how to monitor cellular changes in vivo following a focal, laser-induced retinal damage in Zebrafish. This model can help you answer key questions of retinal regeneration, such as morphological changes, kinetics, as well as cell types involved. The main advantage of this technique is that by inducing a focal injury, biological processes can be investigated directly at the site of injury.Though this method can provide insight into regeneration of Zebrafish retina, it can also be applied to study the repair mechanism in different animal models. Prepare a stock solution of anesthetic by dissolving 400 milligrams of tricaine powder in 97.9 millimeters of tank water, and 2.1 millimeters of one molar TBS. Adjust to pH 7.0 with one molar tris at pH 9.To make the working solution, dilute the tricaine stock solution 1 to 25 in tank water and transfer 50 milliliters to a Petri dish. Then, place the Zebrafish into the anesthetic solution for two to five minutes until they become immobile and do not respond to external stimuli. Transfer each fish by hand to a custom made silicone pin holder for laser treatment.Appropriate anesthesia is critical for the well-being of the animal and the success of procedure. Therefore, freshly prepared tricaine solution is pivotal, and the time out of the water should not far exceed 10 minutes. Set up the output power of the 532 nanometer dialed laser to 70 milliwatts, the pulse duration to 100 milliseconds, and the aerial diameter to 50 microns.Then, apply one to two drops of 2%hydroxypropyl methylcellulose to the eye. When apply methylcell to the eye, ensure that the viscous solution doesn't go in the gills. Next, use a 2 millimeter fundus laser lens to focus the laser aiming beam on the retina.Place four laser spots around the optic nerve on the left eye, and use the right, untreated eye as internal control. Immediately after laser induction, place the still anesthetized Zebrafish on a custom made silicone pin holder in the imaging area. To obtain optimal images, cut a commercially available hydrogel contact lens to fit the Zebrafish eye by means of a hole punch.Fill the concave surface of the lens with methylcellulose, then place it over the cornea. Equip the optical coherence tomography system with a 78D non-contact slit lamp lens. Focus the infrared image in IR plus OCT mode to visualize the fundus of the eye.Then, take the IR pictures by clicking the acquire button to localize the laser spots on the retina. Then, visualize a three dimensional section of the retinal layers in IR plus OCT mode, and take the pictures by clicking the acquire button. Observe the severity of injury in the outer nuclear layer in these images.To reverse anesthesia after treatment and imaging, place the zebrafish in a container containing tank water. To support recovery, create a flow of fresh tank water over the gills by moving the zebrafish back and forth in the water. Immediately after laser treatment, a diffuse hyper-reflective signal was localized to the outer retina.It extended from the retinal pigmented epithelium to the outer plexiform layer. The diffuse hyper-reflective signal is absent in the retina from the control un-lesioned eye. A similar diffuse hyper-reflective signal detected on day one after injury.After day three, this diffuse signal became more organized and dense. It was consistently seen in the outer nuclear layer, extending into the photoreceptor layer. Following the first week, there was a significant decrease in average lesion size, and only a small hyper-reflective signal was detected.Starting from day 14 until the latest time point investigated, the laser spots were no longer visible in IR and OCT images, and the morphology of the retina is comparable to the control side, seen here. H&E staining was employed to investigate the extent and kinetics of retinal degeneration and regeneration. This image shows the control section.This image shows H&E staining three days after injury, when maximum photoreceptor loss is apparent. Immunohistochemistry to visualize the glial cell markers, glutamine synthetase, in red, and glial fibrillary acidic protein, in green, was performed. This image shows a control retina where there is very little green fluorescence, which is indicative of GFAP.The GFAP signal was up-regulated at day three post injury, whereas the red glutamine synthetase signal remained unchanged. Following this procedure, other methods like two photon microscopy as well as cellular and molecular analysis can be performed in order to study the involvement of molecular cells in endogenous repair mechanisms. After watching this video, you should have a good understanding how to induce focal damage to the zebrafish retina and to monitor in vivo the following degenerative and regenerative processes.
