Rigenerazione/degenerazione retinica in zebrafish è stata studiata da diversi approcci quali citotossina-mediata delle cellule morte22, danno meccanico23e lesione termica24. Visione è probabilmente il senso più essenziale dell'essere umano e la sua svalutazione ha un alto impatto socio-economico. Nei paesi industrializzati, le malattie degenerative retiniche rappresentano la maggior parte della perdita della vista e cecità fra la popolazione adulta1. Retinite pigmentosa (RP) è la causa ereditaria più comune di cecità nella gente fra le età di 20 e 60, che colpisce circa 1,5 milioni di persone in tutto il mondo2,3. È una famiglia eterogenea di malattie retiniche ereditarie caratterizzata da perdita progressiva dei fotorecettori (PRs) seguita da degenerazione dell'epitelio retinico del pigmento e, successivamente, il gliosis e il rimodellamento di neuroni interno4. Il corso della malattia può essere spiegato dalla perdita incrementale dei due tipi cellulari PR, solitamente a partire con i coni retinici, che sono responsabili della visione acromatico nella luce fioca e coni, che sono essenziali per la visione e l'acuità visiva del colore5. Un singolo difetto di gene è sufficiente per causare la RP. Finora più di 130 mutazioni in oltre 45 geni sono state associate con la malattia6. Questo conduce a diversi fenotipi di malattia ed è una ragione per cui la terapia genica è non generalizzabile e così un approccio terapeutico intricato. Di conseguenza, c'è un urgente bisogno di sviluppare nuove strategie terapeutiche generali per il trattamento di degenerazioni retiniche in accecante malattie.
Degenerazione retinica spesso comporta perdita di PR; Pertanto, la morte delle cellule di PR è un marchio di garanzia di processi degenerativi della retina7. Già è stato dimostrato che la morte delle cellule PR stimola Müller glia cella (MC) attivazione e proliferazione8. MCs, il tipo di cellule gliali principali nella retina dei vertebrati, una volta sono stati considerati per essere nient'altro che una "colla" tra neuroni retinici. Negli ultimi anni, molti studi hanno dimostrato che MCs agire come più di mera strutturali sostengono il9. Tra le diverse funzioni, MCs partecipare anche nella neurogenesi e10di riparazione. Infatti, in risposta a fattori diffusibili dalla degenerazione retina, MCs significativamente aumentare l'espressione di proteina silicea fibrillare glial (GFAP). Di conseguenza, GFAP etichettatura può essere utilizzato come un indicatore per l'attivazione di MC come una risposta secondaria a lesioni retiniche e degenerazione11.
Recentemente, abbiamo sviluppato un adattamento del romanzo di lesioni focali usando un laser per indurre la degenerazione retinica in zebrafish (Danio rerio). Lesione focale è vantaggioso per lo studio di alcuni processi biologici come la migrazione delle cellule nel sito di feriti e la tempistica precisa degli eventi che si svolgono durante la rigenerazione della retina12. Inoltre, il pesce zebra è diventato importante nella ricerca visiva a causa delle somiglianze tra il sistema visivo e quello di altri vertebrati. Lorda caratteristiche morfologiche ed istologiche di retine umane e Teleostei visualizzano alcune differenze. Di conseguenza, retine umane e zebrafish contengono le stesse classi principali delle cellule organizzate nello stesso modello a strati, dove la luce-sensing fotorecettori occupano lo strato più esterno, mentre i neuroni di proiezione retinica, le cellule del ganglio, risiedono nella parte più interna un neurone strato, prossimale alla lente. Gli interneuroni retinici, amacrine, bipolare e cellule orizzontali, localizzano tra strati di cellule del fotoricettore e del ganglio13. Inoltre, la retina di zebrafish è dominata dal cono e quindi più vicino alla retina umana che, ad esempio, la retina del roditore intensamente studiata. Un'affascinante differenza tra teleosteo e mammiferi è la persistente neurogenesi nella retina di pesce e rigenerazione della retina dopo danni. In zebrafish, MCs può dedifferentiate e mediare la rigenerazione in retina feriti14,15. Nel pollo, MCs hanno alcune capacità anche di inserire nuovamente il ciclo cellulare e di dedifferentiate. A seguito di lesione retinica in pesci adulti, MCs adottare determinate caratteristiche di cellule staminali e progenitrici, la migrazione verso il tessuto retinico danneggiato e produrre nuovi neuroni16. Profili di espressione genica di mammiferi MCs ha rivelato somiglianze inaspettate ai progenitori della retina, e prova per potenziale neurogenico intrinseco di MCs nella pollo, roditore e nella retina umana sta sviluppandosi17. Tuttavia, perché la risposta rigenerativa negli uccelli e nei mammiferi è inferiore rispetto con la risposta robusta nel pesce non è ancora capito. Di conseguenza, la comprensione dei meccanismi di riparazione endogena in zebrafish può suggerire strategie per stimolare la rigenerazione della retina in mammiferi ed esseri umani. Impiegando il meccanismo di riparazione endogena di MCs come strumento terapeutico per il trattamento dei pazienti con degenerazione retinica avrebbe un impatto eccezionale per la nostra società.
Qui, forniamo i passaggi necessari per utilizzare il modello di degenerazione/rigenerazione nella ricerca oftalmica. Prima ci siamo concentrati sulla induzione del danno focale nella retina neurosensoriale, quindi sulla formazione immagine degli eventi in via di sviluppo presso il sito di lesione e infine visualizzante coinvolgimento di MCs adiacente. Il protocollo generale è relativamente facile da effettuare e si apre una vasta gamma di possibilità per la valutazione della retina in seguito.

m&#252;ller glia cell activation in a laser-induced retinal degeneration and regeneration model in zebrafish

Un'affascinante differenza tra teleosteo e mammiferi è il potenziale permanente della retina Teleostei per neurogenesi retinica e la rigenerazione dopo gravi danni. Studiando le vie di rigenerazione in zebrafish potrebbe portare a nuove comprensioni di sviluppare strategie innovative per il trattamento di malattie degenerative retiniche nei mammiferi. Qui, ci siamo concentrati sull'induzione di una lesione focale a retina esterna in zebrafish adulto per mezzo di un laser a diodo 532 nm. Una lesione localizzata permette di indagare i processi biologici che avvengono durante la degenerazione retinica e rigenerazione direttamente presso l'area di danno. Usando la tomografia a coerenza ottica non invasiva (OCT), siamo stati in grado di definire la posizione della rigenerazione successiva area e monitor danneggiata in vivo. Infatti, la formazione immagine OCT produce immagini ad alta risoluzione, a sezione trasversale della retina zebrafish, fornendo informazioni che era precedentemente disponibile solo con le analisi istologiche. The overall goal of this video is to show how to monitor cellular changes in vivo following a focal, laser-induced retinal damage in Zebrafish. This model can help you answer key questions of retinal regeneration, such as morphological changes, kinetics, as well as cell types involved. The main advantage of this technique is that by inducing a focal injury, biological processes can be investigated directly at the site of injury.Though this method can provide insight into regeneration of Zebrafish retina, it can also be applied to study the repair mechanism in different animal models. Prepare a stock solution of anesthetic by dissolving 400 milligrams of tricaine powder in 97.9 millimeters of tank water, and 2.1 millimeters of one molar TBS. Adjust to pH 7.0 with one molar tris at pH 9.To make the working solution, dilute the tricaine stock solution 1 to 25 in tank water and transfer 50 milliliters to a Petri dish. Then, place the Zebrafish into the anesthetic solution for two to five minutes until they become immobile and do not respond to external stimuli. Transfer each fish by hand to a custom made silicone pin holder for laser treatment.Appropriate anesthesia is critical for the well-being of the animal and the success of procedure. Therefore, freshly prepared tricaine solution is pivotal, and the time out of the water should not far exceed 10 minutes. Set up the output power of the 532 nanometer dialed laser to 70 milliwatts, the pulse duration to 100 milliseconds, and the aerial diameter to 50 microns.Then, apply one to two drops of 2%hydroxypropyl methylcellulose to the eye. When apply methylcell to the eye, ensure that the viscous solution doesn't go in the gills. Next, use a 2 millimeter fundus laser lens to focus the laser aiming beam on the retina.Place four laser spots around the optic nerve on the left eye, and use the right, untreated eye as internal control. Immediately after laser induction, place the still anesthetized Zebrafish on a custom made silicone pin holder in the imaging area. To obtain optimal images, cut a commercially available hydrogel contact lens to fit the Zebrafish eye by means of a hole punch.Fill the concave surface of the lens with methylcellulose, then place it over the cornea. Equip the optical coherence tomography system with a 78D non-contact slit lamp lens. Focus the infrared image in IR plus OCT mode to visualize the fundus of the eye.Then, take the IR pictures by clicking the acquire button to localize the laser spots on the retina. Then, visualize a three dimensional section of the retinal layers in IR plus OCT mode, and take the pictures by clicking the acquire button. Observe the severity of injury in the outer nuclear layer in these images.To reverse anesthesia after treatment and imaging, place the zebrafish in a container containing tank water. To support recovery, create a flow of fresh tank water over the gills by moving the zebrafish back and forth in the water. Immediately after laser treatment, a diffuse hyper-reflective signal was localized to the outer retina.It extended from the retinal pigmented epithelium to the outer plexiform layer. The diffuse hyper-reflective signal is absent in the retina from the control un-lesioned eye. A similar diffuse hyper-reflective signal detected on day one after injury.After day three, this diffuse signal became more organized and dense. It was consistently seen in the outer nuclear layer, extending into the photoreceptor layer. Following the first week, there was a significant decrease in average lesion size, and only a small hyper-reflective signal was detected.Starting from day 14 until the latest time point investigated, the laser spots were no longer visible in IR and OCT images, and the morphology of the retina is comparable to the control side, seen here. H&E staining was employed to investigate the extent and kinetics of retinal degeneration and regeneration. This image shows the control section.This image shows H&E staining three days after injury, when maximum photoreceptor loss is apparent. Immunohistochemistry to visualize the glial cell markers, glutamine synthetase, in red, and glial fibrillary acidic protein, in green, was performed. This image shows a control retina where there is very little green fluorescence, which is indicative of GFAP.The GFAP signal was up-regulated at day three post injury, whereas the red glutamine synthetase signal remained unchanged. Following this procedure, other methods like two photon microscopy as well as cellular and molecular analysis can be performed in order to study the involvement of molecular cells in endogenous repair mechanisms. After watching this video, you should have a good understanding how to induce focal damage to the zebrafish retina and to monitor in vivo the following degenerative and regenerative processes.
