שינויים פתולוגיים של מערכת העצבים הנגרם על ידי הטראומה, תפקוד מטבולי, דלקת, זיהום, איסכמיה, או מחלות אוטואימוניות מדי פעם לגרום כאב נוירופטי, אשר מוגדר כאבים הנובעים כתוצאה ישירה של פגיעה או מחלה להשפיע על מערכת המגע1. כאב נוירופטי הוא בדרך כלל בלתי נסבל, למרבה הצער, משככי כאבים רגילים בדרך כלל אינם מייצרים מספיק כאב הקלה2. תכונה מרכזית של כאב נוירופתי מכיל ספונטנית ועוררו על-ידי גירוי כאבים (קרי, allodynia ו- hyperalgesia). Allodynia היא תגובה nociceptive המתרחשת לגירויים שאינם כואבים בדרך כלל, כגון במגע קל או גירוי חמים.
