Migratie van eukaryotische cellen is bevooroordeeld door middel van diverse chemische en fysische signalen in het milieu, met inbegrip van gradiënten van oplosbare of substraat-gebonden chemoattractants, variabele stijfheid van de substraten, elektrische velden of schuintrekken stroom. Hoewel er al veel vooruitgang in ons begrip van de moleculaire mechanismen chemotaxis rijden, minder is gekend over andere soorten gestuurde migratie en hoe deze uiteenlopende signalen zijn geïntegreerd op cellulair niveau tot een uniforme trekkende reactie.
Regie migratie naar een toenemende concentratie van een chemoattractant omvat drie gedrags componenten: beweeglijkheid, directionele sensing en polariteit1. Motility verwijst naar willekeurige verkeer van cellen bereikt door pseudopod uitsteeksel. Directionele sensing is de mogelijkheid van een cel om de oorzaak van een chemoattractant worden opgespoord, die kan optreden zelfs in cellen geïmmobiliseerd. Polariteit verwijst naar de meer stabiele asymmetrische distributie van intracellulaire componenten tussen de leading en lagging rand van een cel, die tot verhoogde persistentie in beweging leidt.
Cellulaire reactie op een chemoattractant hangt af van de activiteit van vier conceptueel gedefinieerde regulerende netwerken: receptor / G eiwit, signaaltransductie, actine cytoskelet en polariteit1. Chemoattractant binding aan de G eiwit-gekoppelde receptor stuurt het signaal via heterotrimeric G eiwitten α en βγ op het stroomafwaarts signaaltransductie netwerk, dat het signaal directionele versterkt. Meerdere trajecten binnen het netwerk signaaltransductie handelen in parallel en worden meegenomen bij het actine cytoskelet netwerk aan bias actine polymerisatie, en daaruit voortvloeiende pseudopod uitsteeksel, in de richting van het verloop. Belangrijke regulatoren van chemotaxis behoren tot Ras GTPase, TorC2, phosphoinositide 3-kinase (PI3K) fosfatase en Tenzin homolog (PTEN) en guanylyl cyclase. Feedbackmechanismen binnen het netwerk signaaltransductie en tussen de signaaltransductie en actine cytoskelet netwerken verder versterken het antwoord. Ten slotte, het slecht gedefinieerde polariteit netwerk ontvangt input van het cytoskelet actine, en verder vooroordelen het signaaltransductie netwerk ter bevordering van permanente migratie in de richting van het verloop.
Veel van onze mechanistische begrip van chemotaxis werd mogelijk gemaakt door de ontwikkeling van fluorescently-tagged biosensoren voor verschillende onderdelen van de regulerende netwerken. Vele chemotaxis toezichthouders hebben een asymmetrische verdeling van de regelgevende molecuul zelf of haar activiteit. Bijvoorbeeld, biosensoren die herkent geactiveerd versies van kleine GTPases Ras en Rap1-Ras-bindende domeinen van Raf1 (hierna aangeduid als RBD hier) en RalGDS, respectievelijk – lokaliseren naar de voorrand van een chemotaxing cel2,3. Evenzo PI3K en haar product phosphatidylinositol (3,4,5)-trisphosphate (PIP3), erkend door een pleckstrin-homologie (PH) domein, Toon ook lokalisatie aan de voorzijde van een cel4,5. In tegenstelling, een 3-fosfatase-PTEN, die worden omgezet in PIP3 terug phosphatidylinositol (4,5)-difosfaat, lokaliseert aan de rand van de achterstand van de cel-6. Nog belangrijker is, is deze biosensoren wijzigen hun lokalisatie in reactie op de wereldwijde stimulatie met een chemoattractant. Leading edge markers, die cytosolische of op de toppen van de uitsteeksels in een cel rusten, relocalize naar de cortex, overwegende dat achtergebleven rand markeringen, die corticale lokalisatie en zijn afwezig vanaf de toppen van de uitsteeksels in een cel rusten, worden cytosolische na stimulatie. Analyse van biosensor distributie in reactie op de wereldwijde stimulatie met een chemoattractant minimaliseert de bijdrage van de motiliteit en polariteit, die vaak de opmerkingen verwarren. Globale of uniforme stimulatie van een celsuspensie met een chemoattractant wordt ook gebruikt als een instrument voor het beoordelen van wijzigingen voor de gehele bevolking in eiwit activering, vaak gedetecteerd door proteïne fosforylatie7,8,9 . Deze biochemische bepaling wordt voornamelijk gebruikt voor temporele informatie, terwijl microscopie wordt gebruikt om zowel temporele en ruimtelijke informatie verzamelen over het gedrag van verschillende onderdelen van de regulerende netwerken.
Het signaaltransductie netwerk omvat functies van een prikkelbaar systeem10,11. Reacties op supra-drempel Chemotactische stimuli zijn "all-or-none" en vuurvaste punten weergegeven. Reacties zijn ook stochastically geactiveerd en oscillerende gedrag kunnen tonen. Signaaltransductie gebeurtenissen zijn gelokaliseerd aan regio's van de cortex die als golven12,13,14,15 verspreiden. Voorste, terug, markeringen worden gerekruteerd voor of distantiëren van, de actieve zones van het teeltmateriaal golven. Als gevolg van de achterstand van de actieve zone vuurvaste regio vernietigen de tegengesteld gerichte golven als ze elkaar ontmoeten. Het teeltmateriaal signaaltransductie golven ten grondslag liggen aan de cellulaire uitsteeksels die cel migratie10bemiddelen.
Veel van de bovengenoemde informatie over chemotaxis kwam van de studies op de sociale Amoebe Dictyostelium discoideum, hoewel soortgelijke regulerende mechanismen zijn ook van toepassing op de neutrofielen en andere zoogdieren cel typen16 . Dictyostelium is een reeds lang gevestigde modelorganisme dat een robuuste Chemotactische reactie tijdens hongersnood, heeft wanneer duizenden afzonderlijke cellen naar een aggregatie midden migreren, uiteindelijk de vorming van een meercellig fruiting body met sporen. Chemotaxis is ook essentieel in de fase van de eencellige groei van dit organisme voor het lokaliseren van bacteriële voedselbronnen. Belangrijker, is migratie van afzonderlijke Dictyostelium cellen opvallend vergelijkbaar met de migratie van zoogdieren neutrofielen of metastatische kankercellen, die allemaal zeer snelle Wortelpotigen-type migratie ondergaan. In feite, worden zowel de algemene topologie van de regulerende netwerken, evenals veel van de individuele signaaltransductie trajecten die betrokken zijn bij chemotaxis bewaard tussen Dictyostelium en zoogdieren leukocyten17. Bovendien andere cellen, zoals fibroblasten, receptor tyrosine kinases (RTK) gebruiken in plaats van GPCRs; RTKs kunnen echter voeden in soortgelijke netwerken.
In tegenstelling tot chemotaxis ontbreekt grondig inzicht in de signalering mechanismen die verschillende andere vervoerswijzen gestuurde migratie rijden. Ook naar cellen migreren in een chemoattractant verloop verscheidene studies hebben gemeld activering en/of de lokalisatie van typische voorrand markers, met inbegrip van actine polymerisatie, PIP3 en/of extracellulaire signaal-gereglementeerde kinase (ERK) 1/2, op de voorkant van cellen ondergaan regisseerde migratie in reactie op stroom of veranderingen in elektrische velden18,19,20,21schuintrekken. Echter, in deze studies continue blootstelling aan de prikkel ook resulteerde in cel migratie, bloot de vraag, bijvoorbeeld, de toonaangevende markers lokaliseren speciaal in reactie op een prikkel, of als ze gewoon lokaliseren op de voorrand vanwege toegenomen aantal pseudopods aan de voorzijde van een migreren cel.
We ontwikkeld testen die ons toelaten te observeren de reactie van cellen op acute mechanische perturbation geleverd als schuintrekken stroom zowel op het bevolkingsniveau en als afzonderlijke cellen22. Vergelijkbaar met de globale stimulatie met een chemoattractant, acute stimulation met schuintrekken stroom maakt de studie van een cellulaire reactie op een mechanische prikkel zonder de storende bijdrage van beweeglijkheid of polariteit. Deze biochemische en microscopische tests te combineren met genetische of farmacologische verstoringen ons toelaat om te leren over hoe mechanische stimuli worden waargenomen en verzonden. Bovendien biedt deze aanpak ook een nieuwe methode voor te tikken in het systeem stroomafwaarts van de chemoattractant receptor in het ontbreken van een chemoattractant, waardoor het isoleren van de netwerken van de signaal transductie en actine de cytoskelet van de receptor/G eiwit netwerk.
Met behulp van de technieken die hieronder we onlangs aangetoond dat acute shear stress leidt tot activatie van meerdere onderdelen van de Chemotactische signaal transductie en actine cytoskelet netwerken22. Door de acute mechanische stimulus toe te passen op verschillende tijdstippen, we laten zien dat ook aan chemoattractants, reactie op mechanische stimuli ook kenmerken van een prikkelbaar systeem vertoont, met inbegrip van het all-or-none gedrag van de reactie onder verzadigen van de voorwaarden en de aanwezigheid van een refractaire periode. Tot slot, door een combinatie van mechanische en chemische stimulatie we bleek dat de twee prikkels aandeel signaal transductie en actine cytoskelet netwerken, waardoor waarschijnlijk voor de integratie van meerdere stimuli bias cel migratie.

Chemotaxis, of de migratie van een verloop van een chemoattractant, is het best te begrijpen wijze van gestuurde migratie. Studies met behulp van sociale amoeba Dictyostelium discoideum bleek dat een complex signaaltransductie netwerk van parallelle trajecten de respons op chemoattractants versterkt, en leidt tot vooringenomen actine polymerisatie en uitsteeksel van een pseudopod in de richting van een verloop. Moleculaire mechanismen rijden van andere soorten gestuurde migratie, bijvoorbeeld als gevolg van blootstelling aan het schuintrekken stroom of elektrische velden, zijn daarentegen niet bekend. Vele regulatoren van chemotaxis vertonen lokalisatie bij de regelafstand of de achterblijvende hoek van een cel, migreren, evenals tijdelijke wijzigingen worden weergegeven in de lokalisatie of activering na globale stimulatie met een chemoattractant. Om te begrijpen van de moleculaire mechanismen van andere soorten gestuurde migratie ontwikkelden we een methode waarmee onderzoek van cellulaire reactie op acute mechanische stimulatie op basis van korte (2-5 s) blootstelling aan stroom schuintrekken. Deze stimulatie kan geleverd worden in een kanaal terwijl imaging cellen uiten fluorescently-geëtiketteerden biosensoren te onderzoeken van de individuele cel gedrag. Bovendien kan celpopulatie worden gestimuleerd in een plaat, lysed, en immunoblotted met behulp van antilichamen die actieve versies van proteïnen van belang herkent. Door het combineren van beide tests, kan men een breed scala van moleculen geactiveerd door veranderingen in subcellular localisatie en/of fosforylatie onderzoeken. Using zulks werkwijze vastbesloten wij dat acute mechanische stimulatie activering van de Chemotactische signaal transductie en actine cytoskelet netwerken activeert. De mogelijkheid te onderzoeken van cellulaire reacties op acute mechanische stimulatie is belangrijk voor het begrijpen van de initiatiefnemende gebeurtenissen nodig zijn voor de beweeglijkheid van de stroom-geïnduceerde schuintrekken. Deze aanpak biedt een instrument voor de studie van de Chemotactische signaaltransductie netwerk zonder de storende invloed van de chemoattractant receptor.
