I in vitro- SUMOylation-protokollet som beskrivs här används rutinmässigt att etablera SUMOylation status identifierade Ubc9 substrat. Den största begränsningen använda standardprotokollet för att studera funktionen SUMOylation av SUMO E3 ligase är överflödet av aktivering av SUMO E1 och E2 konjugera enzymer Ubc9 som maximerar SUMO konjugationen i in vitro- system. SUMO modifiering identifierades inledningsvis som en reversibel posttranslationella modifieringen (PTM) som reglerar protein stabilitet1. Förutom direkta konjugation, kan SUMO också bifogas ett protein genom icke-kovalenta interaktion av SUMO interaktion motiv (SIMs)2. Liknar den bindningen av tyrosyl-fosforylering av molekyler som härbärgerat Src homologi 2 (SH2) eller phosphotyrosine bindande (PTB) domäner3,4, SUMO modifiering ger en ytterligare interaktion plattform för selektiv rekrytering av SIM-innehållande effektor proteiner5,6. Förutom regleringen av signaltransduktion fungerar SUMOylation av transkriptionell faktorer och kromatin remodeling molekyler att modulera gen uttryck7,8. Studier av genome-wide SUMOylation mönster har visat att SUMO modifiering är associerade med antingen positiva9,10 eller negativa10,11,12 transkription förordning, delvis på grund av SUMOylation paralogue specificitet.
Det finns tre stora SUMO isoformer för protein konjugera närvarande i däggdjursceller; Dessa inkluderar SUMO-1, och mycket homologa SUMO-2 och SUMO-3 (kallad SUMO-2/3)13. I in vitro- SUMOylation-test, som utvärderar renat Ubc9 förmåga att katalysera SUMOylation av substrat proteiner, är en väl accepterade analys för att studera SUMO ändring17. Dock SUMO E3 ligase aktivitet i de flesta fall inte kan upptäckas eller kan endast upptäckas med muterade SUMO ligase med hög SUMO E3 ligase aktivitet och låg substrat specificitet av standardprotokollet på grund av närvaron av en riklig mängd Ubc918 . Efter en småskalig genomgång av de samverkande proteinerna av viral SUMO E3 ligase K-bZIP, p53 identifierades som romanen substrat. I detta protokoll visar vi i detalj i insekt celler stegen inblandade i uttryck och rening av vild typ K-bZIP, en viral SUMO E3-ligase med SUMO-2/3 specificitet. Den renade K-bZIP förmåga att förbättra SUMOylation av p53, en välkänd SUMO substrat, utvärderas i närvaro av reducerade mängder E1 och E2 enzymer. Liten ubiquitin-liknande modifierare (SUMO) ändring är en viktig post-translationella ändring (PTM) som medierar cellsignalering främst genom modulerande protein-protein interaktioner. Liknar ubiquitin modifiering, SUMOylation är regisserad av en sekventiell enzym kaskad inklusive E1-aktiverande enzym (SAE1/SAE2), E2-konjugation enzym (Ubc9) och E3-ligase (dvs, PIAS familj, RanBP2 och Pc2). Dock skiljer sig från ubikvitinering, en E3-ligase är icke-väsentliga för reaktionen men inte ger precision och effekt för SUMO konjugation. Proteiner modifieras av SUMOylation kan identifieras genom in-vivo analys via immunoprecipitation med substrat-specifika antikroppar och immunoblotting med SUMO-specifika antikroppar. Demonstration av protein SUMO E3 ligase aktivitet kräver dock in vitro- beredning av SUMOylation analyser med renade enzymer, substrat och SUMO proteiner. Eftersom i in vitro- reaktioner, oftast SAE1/SAE2 och Ubc9, ensam är tillräcklig för SUMO konjugation, är förbättring av SUMOylation av en förmodad E3-ligase inte alltid lätt att upptäcka. Här beskriver vi en modifierad in vitro- SUMOylation protokoll som konsekvent identifierar SUMO ändring med hjälp av ett in vitro- beredning-system. En steg för steg-protokollet att rena katalytiskt aktiva K-bZIP, en viral SUMO-2/3 E3-ligase, presenteras också. SUMOylation verksamhet den renade K-bZIP visas på p53, ett välkänt mål för SUMO.
