Ovariotomie en -transplantatie van de eierstokken intact en geslachtscellen-verarmd zijn uitgevoerd in muizen variërend van 21 dagen tot 18 maanden. Bij het uitvoeren van een ovariotomie waar het wenselijk om de ovariële bursa intact is, moet worden gezorgd als het bursal weefsel gebeuren. Bij muizen die ouder zijn dan zes maanden oud, wordt veel van de bursa bedekt met het vet pad, waardoor de gebieden die kunnen worden gebruikt voor toegang wordt beperkt. In deze gevallen is het vaak voordelig te scheuren van de burs...

De invloed van reproductie op gezondheid en levensduur wordt vaak ten onrechte gedacht zo beperkt slechts tot de periode omvat reproductieve competentie. Terwijl de levensstijl en de fenotypes gekoppeld aan de reproductief bevoegde fase van de levensduur gemakkelijk van de pre- en postreproductive fasen onderscheiden worden, beïnvloedt de status van de reproductieve gezondheid in alle fasen van de levensduur. De periode van reproductieve bekwaamheid is vaak aangeduid als de reproductieve levensduur, onderscheiden zich van de chronologische levensduur. Deze terminologie leent zich voor een kunstmatige scheiding van de twee benamingen van de levensduur wanneer in werkelijkheid, beide zijn nauw gebonden aan elkaar met gebeurtenissen die van invloed is, vaker wel dan niet, waardoor de andere ook.
De afgelopen tien jaar blijkt dat iemands reproductieve status wordt geassocieerd met een verhoogd risico op het ontwikkelen van chronische gezondheid voorwaarden1. Een studie een vereniging tussen kortere levensduur en reproductieve mislukking voor een cohort van mannen2gedocumenteerd. De vereniging is nog meer opvallend bij vrouwen. Hart-en vaatziekten is zeldzaam in premenopausal vrouwen3, maar ovarieel falen verhoogt hart-en vaatziekten fors bij menopauze4 en bij jonge vrouwen met prematuur ovarieel falen5. Insuline resistentie6 en bot verlies in menopauze7 toenemen en bijna tweederde van de Amerikanen met de ziekte van Alzheimer zijn vrouwen8. Well-defined veranderingen in ovariële signalering markeert de overgang van een premenopausal, ziekte-resistente staat, naar een postmenopauzale, ziekte-belast staat. Deze stabiele overgang, biedt een kans om te identificeren van mechanismen die ziekte risico's in de ouder te versnellen.
Transplantatie van cryopreserved eierstokweefsel is onlangs gebruikt om te herstellen van de vruchtbaarheid bij vrouwen die medische procedures die gevaar voor de geslachtscellen voor de lijn, zoals chemotherapie opleveren kunnen ondergaan. Transplantatie van eierstokweefsel kan ook worden gebruikt voor de productie van levensvatbare nakomelingen in dieren met niet-ovariële gebaseerde onvruchtbaarheid, met inbegrip van transgene muizen9. Ovariële transplantatie is een efficiënte methode om te scheiden van de invloed van de voortplantingsfunctie of reproductieve veroudering van chronologische veroudering per se.
De gevestigde ondersteunende rol voor de eierstokken hormonen in vele aspecten van de gezondheid van vrouwen impliceert het verlies van hormoonproductie van het actief fietsen kiemcellen, als de voornaamste oorzaak van ziekte van de verhoogde risico's bij menopauze. Terwijl de waarde van de eierstokken hormonen in de gezondheid van vrouwen onbetwistbaar is, ter vervanging van deze hormonen in de peri- en postmenopauzale vrouwen geweest onbetrouwbaar in het herstel van de gezondheidsvoordelen voor jonge vrouwen met jonge eierstokken. Deze voordelen voor de gezondheid zijn betrouwbaar hersteld door heterochronic transplantatie van jonge eierstokken postreproductive vrouwelijke muizen10,11,12,13,14.
Reproductieve status van zowel de donor als de ontvanger muizen moet worden bepaald vóór en na de transplantatie of ovariotomie procedures. Vermoedelijke postreproductive muizen waarin tekenen van gonadale input voorafgaand aan de operatie op 12 maanden oud zijn uitgesloten van deze experimenten. Gonadale input wordt bepaald door vaginale cytologie. Gegevens over vaginale cytologie zijn verzameld voor ten minste 10 opeenvolgende dagen vóór chirurgie om ervoor te zorgen een postreproductive staat in ontvangende muizen en actief-Fiet & Mountainbike eierstokken in donor muizen. Een oestrische cyclus wordt gedefinieerd als de periode van de dag nucleated epitheliale cellen voor het eerst verschijnen (dat wil zeggen, proestrus) tot de dag voorafgaande aan de volgende verschijning van genucleëerde epitheliale cellen in de vaginale uitstrijkjes, mits er een periode van leukocytic aanwezigheid (dat wil zeggen, diestrus) in tussen. Estrus wordt bepaald door de aanwezigheid van grote, squamous epitheliaale cellen, met of zonder kernen. Muizen weergeven die geen cyclische activiteit gedurende een periode van 10 dagen vóór en/of na de operatie worden bepaald dat geen gonadale input voor de genoemde periode. Muizen die ten minste één volledige oestrische cyclus in een periode van 10 dagen vóór en/of weergegeven na chirurgie zijn vastbesloten om gonadale inbreng hebben voor de genoemde periode. Vrouwelijke muizen moeten worden gehuisvest in aanwezigheid van mannelijke muizen of blootgesteld aan bevuilde beddengoed van een man in de kooi om consistente blootstelling aan cyclus-inducerende prikkels.
Deze transplantatie procedures zijn zo efficiënt mogelijk bereikt met twee chirurgen werken tegelijkertijd op donor- en ontvangende muizen. Echter, de procedures zijn eenvoudig te realiseren door een chirurg, met de donor eierstokken meestal hersteld voorafgaand aan het begin van de procedure van de ovariotomie op de ontvangende muis. Dit protocol beschrijft de procedure voor de enkele chirurg. Blootstelling van het voortplantingskanaal en de sluiting van de site wordt normaal gedaan zonder een Microscoop, overwegende dat de verwijdering en de vervanging van de eierstokken wordt gedaan onder een microscoop. Wanneer met het leren van elke chirurgische procedure het, incisie sites groter worden kunnen en chirurgie kunnen terwijl het eerste leren de procedure langer duren en zal over het algemeen kleiner met ervaring.

Ovariële transplantatie werd voor het eerst uitgevoerd in Utah State University in 1963. In meer recente werk wordt heterochronic transplantatie van zoogdieren eierstokken gebruikt om te onderzoeken van de gezondheid-beschermende effecten van jonge eierstokken bij jonge vrouwen. De huidige procedures gebruik van een orthotopic transplantatie methode, waar allogene eierstokken terug naar hun oorspronkelijke positie in de eierstokken bursa worden getransplanteerd. Dit is in tegenstelling tot de meer algemeen gebruikte heterotopic transplantatie van de eierstokken/eierstokweefsel subcutaan of onder de capsule van de nier. Alle drie locaties bieden efficiënte revascularisatie van het getransplanteerde weefsel. Echter orthotopic transplantatie biedt de meest natuurlijke signalering omgeving de eierstok en is de enige procedure die de mogelijkheid voor het dier biedt te reproduceren natuurlijk post-operatively. Men moet zorgen om alle endogene eierstokweefsel verwijderen tijdens de procedure ovariotomie. Als een endogene weefsel blijft of slechts één eierstok wordt verwijderd, blijft het getransplanteerde weefsel slapende zijn totdat het bestaande weefsel verouderend wordt. Terwijl revascularisatie van de getransplanteerde eierstokken zeer snel optreedt, kan de ontvanger van de transplantatie nemen een aanzienlijke hoeveelheid tijd aan te passen aan een nieuwe hypothalame/hypofyse/gonadale/bijnier (HPG/A) as signalering regime geassocieerd met de getransplanteerde weefsel. Dit duurt normaal gesproken ongeveer 100 dagen in de muis. Daarom moeten transplantatie experimenten worden ontworpen om deze aanpassingsperiode tegemoet. Typische resultaten met ovariële transplantatie zal omvatten veranderingen in de gezondheid van de ontvanger die overeenkomen met de leeftijd van de getransplanteerde eierstok, in plaats van de chronologische leeftijd van de ontvanger.

Ovariotomie op de leeftijd van 21 dagen zullen grote up-regulatie van het reproductieve systeem bij het begin van de puberteit voorkomen en elimineren van andere invloeden die de vrouwelijke gonaden bovendien directe effecten van gonadale hormonen wellicht.
