Chez les rats âgés de moins de jour 11, n’on observe aucun potentiel d’action en réponse à une stimulation sonore au cortex auditif6,7. Deux fonctions distinctes de cellules de cheveux sensoriels cochléaires sont essentielles pour l’audience chez les mammifères : mechanoelectric transduction (MET) et electromotility1,2. Par MET les canaux situés dans le faisceau de cheveux, CSI (CSI) et contaminants organohalogénés convertir les vibrations sonores des changements potentiels de membrane, ainsi que les signaux électriques des neurones du ganglion spiral innervés. Contaminants organohalogénés changent leur longueur avec le potentiel de membrane des cellules et amplifient la vibration du son à basse altitude. Cette activité appelée electromotility est dérivée de la protéine motrice prestin situé dans la paroi latérale des contaminants organohalogénés3.
Chez de nombreuses espèces, y compris les rongeurs, la fonction auditive est immature à l’époque postnatale précoce4,5. Aucun potentiel d’action en réponse aux signaux sonores ne pourraient être détectée dans le cortex auditif avant l’audience début6,7. Développement de la morphologie et le fonctionnement de la cochlée a été largement étudiées en souris, gerbille et rat4,5,8. La mécanotransduction et electromotility des cellules ciliées sont également développées dans la première époque de vie5.
Afin d’évaluer la sensibilité auditive des rats à différents âges postnatals, nous avons développé une méthode de réaction auditif du tronc cérébral (PEATC) enregistrement chez les ratons. Cellule entière patch clamp d’est une technologie idéale pour enquêter sur les contaminants organohalogénés électrophysiologique. Toutefois, le faible taux de cellules entières d’étanchéité en comparaison avec le patch clamp de jouée dans les neurones et autres cellules épithéliales, limité l’enquête d’electromotility des contaminants organohalogénés isolés.
Ici, nous décrivons une procédure pour enquêter sur les contaminants organohalogénés morphologiquement et électrophysiologique dans la cochlée aiguë dissociée des rats postnatals. Cette méthode peut être modifiée afin d’étudier les mécanismes moléculaires qui régissent la fonction et le développement des cellules ciliées internes.

Les cellules ciliées externes est l’un des deux types de sensorielle des cellules ciliées de la cochlée mammifères. Ils modifient leur longueur des cellules avec le récepteur potentiel pour amplifier la vibration faible du signal sonore à basse altitude. La morphologie et la propriété électrophysiologique des cellules ciliées externes (contaminants organohalogénés) se développent dans l’âge postnatal précoce. La maturation des cellules ciliées externes peut-être contribuer au développement du système auditif. Toutefois, le processus de développement de contaminants organohalogénés n’est pas bien étudié. C’est en partie à cause de la difficulté à mesurer leur fonction par une approche électrophysiologique. Dans le but de développer une méthode simple pour résoudre le problème ci-dessus, nous décrivons ici un protocole étape par étape pour étudier la fonction des contaminants organohalogénés cochlée aiguë dissociés des rats postnatals. Avec cette méthode, nous pouvons évaluer la cochléaire réponse à des stimuli de sons purs et examiner le niveau d’expression et la fonction de la protéine motrice de prestin en contaminants organohalogénés. Cette méthode peut également être utilisée pour étudier les cellules ciliées internes (CSI).

ABR peut être déclenché de rat anesthésié petits plus vieux que le jour après la naissance 7 (P7) à l’aide de salves de sons purs (Figure 1 a).
