Hier beschrijven we een facile en robuuste methode voor het opsporen van Rab10 fosforylering door LRRK2 endogene niveau op basis van de methodologie van de P-tag. Omdat het op dit moment beschikbare antilichaam tegen gefosforyleerd Rab10 alleen met overexpressie eiwitten15 werkt, is de huidige methode met behulp van P-tag SDS-pagina de enige manier om te beoordelen van endogene niveaus van Rab10 fosforylering. PD is één van de meest voorkomende neurodegeneratieve ziekten, voornamelijk op het gebied van Dopaminerge neuronen in de veroorzaakt, resulterend in een dysfunctie van de motor systemen in ouderen1. Terwijl de meeste patiënten PD in een sporadische manier ontwikkelen, zijn er gezinnen overnemen van de ziekte. Mutaties in meerdere genen zijn bevonden om te worden gekoppeld aan FPD2. Een van de oorzakelijke genen voor FPD is LRRK2, waarin acht missense mutaties (N1437H, R1441C/G/H/S, Y1699C, G2019S en I2020T) gekoppeld aan een dominant erfelijke FPD genaamd PARK8 dusver gerapporteerde3,4,5. Verschillende genoom-brede vereniging studies (GWAS) van sporadische PD-patiënten hebben ook genomic variaties op de LRRK2 locus geïdentificeerd als een risicofactor voor PD, suggereert dat de afwijking in de functie van LRRK2 een veelvoorkomende oorzaak van neurodegeneratie in beide sporadische is en PARK8 FPD6,7,8.
LRRK2 is een groot eiwit (2,527 aminozuren), die bestaat uit een rijk aan leucine herhalen domein, een Ras van de GTP-bindende van complexe eiwitten (ROC) domein, een C-terminal van ROC (COR) domein, een serine/threonine proteïne kinase domein, en een WD40 herhalen domein9. De acht FPD mutaties zoeken in deze functionele domeinen; N1437H en R1441C/G/H/S in het domein van de ROC, Y1699C in het domein van het CVDR, G2019S en I2020T in het domein kinase. Aangezien de G2019S mutatie, die het vaakst vlindertje mutatie PD patiënten10,11,12, verhoogt het kinaseactiviteit van LRRK2 door 2-3 vouwen in vitro13, is het veronderstelde dat de abnormale toename van fosforylering van de LRRK2 substrate(s) giftig voor zenuwcellen is. Het is echter onmogelijk om te bestuderen of de fosforylatie van fysiologisch relevante LRRK2 substraten is gewijzigd in familiale/sporadische PD-patiënten als gevolg van het gebrek aan methoden evalueren het in patiënt afgeleide monsters.
Proteïne Fosforylatie is over het algemeen herkend door immunoblotting of enzyme-linked immunosorbent assay (ELISA) met behulp van antilichamen specifiek herkennen de gefosforyleerd staat van eiwitten of door massaspectrometrische analyse. Echter, de voormalige strategie niet soms worden toegepast wegens de moeilijkheden bij het creëren van fosforylering-specifieke antilichamen. Metabole labeling van cellen met radioactieve fosfaat is een andere optie te onderzoeken fysiologische niveaus van fosforylering fosforylering-specifieke antilichamen niet dadelijk beschikbaar zijn. Echter het vereist een grote hoeveelheid radioactief materiaal en daarom gaat sommige gespecialiseerde apparatuur voor stralingsbescherming14. Massaspectrometrische analyse is gevoeliger in vergelijking met deze immunochemical methoden en werd populair in het analyseren van proteïne fosforylatie. Echter, de bereiding van de monsters is tijdrovende en dure instrumenten nodig zijn voor de analyse.
Een subset van de familie van de Rab GTPase met inbegrip van Rab10 en Rab8 werd onlangs gemeld als directe fysiologische substraten voor LRRK2 op basis van het resultaat van een grootschalige phosphoproteomic analyse15. We toonden dan dat Rab10 fosforylering door FPD mutaties in muis embryonale fibroblasten en in de longen van knockin muizen16werd verhoogd. In dit verslag, kozen we om een polyacrylamide-gelelektroforese voor natrium dodecyl sulfaat (SDS-pagina) in dienst-op basis van de methode waarbij een P-tag molecuul mede gepolymeriseerde in SDS-pagina gelen (P-tag SDS-PAGE is) voor het opsporen van de endogene niveaus van fosforylering van de Rab10, omdat een hooggevoelige antilichaam specifiek voor gefosforyleerd Rab10 ontbrak nog. We hebben gefaald om te ontdekken de fosforylatie van endogene Rab8 te wijten aan de slechte selectiviteit van momenteel beschikbare antilichamen voor totale Rab8. Daarom besloten hebben we om zich te concentreren op de Rab10 fosforylering. LRRK2 kinaseenzym Rab10 op Thr73 zoeken in het midden van de regio zeer geconserveerde "Wissel II". Hoge instandhouding van de fosforylatie sites onder Rab eiwitten misschien wel een van de redenen waarom phosphospecific antilichamen herkennen van afzonderlijke Rab eiwitten zijn moeilijk om te maken.
De fosforylering van de Rab8A door LRRK2 remt de binding van Rabin8, een guanine nucleotide uitwisseling factor (GEF), die Rab8A activeert door de afhankelijke BBP met GTP15uit te wisselen. Fosforylering van de Rab10 en Rab8A door LRRK2 remt ook de binding van BBP-dissociatie-remmers (GDIs), die essentieel voor de activering van Rab eiwitten zijn door winning van BBP-gebonden Rab eiwitten uit membranen15. Collectief, is het veronderstelde dat de fosforylatie van Rab eiwitten door LRRK2 hen activering verhindert hoewel de exacte moleculaire mechanisme en fysiologische gevolgen van de fosforylatie onduidelijk blijven.
P-tag SDS-pagina werd uitgevonden door Kinoshita et al. in 2006: bij deze methode wordt acrylamide covalent gekoppeld is met P-tag, een molecule vastleggen van fosfaten met hoge affiniteit, die copolymerized in SDS-pagina gels17. Omdat de moleculen van de P-tag in een SDS-pagina gel retard selectief elektroforetische mobiliteit van gefosforyleerd eiwitten, P-tag SDS-pagina gefosforyleerd eiwitten kan scheiden van degenen die niet phosphorylated (Figuur 1). Als de eiwit-of-interest is phosphorylated op meerdere residuen, worden een ladder van bands overeenkomt met differentieel gefosforyleerd formulieren nageleefd. In het geval van Rab10 observeren wij slechts één verschoven band, die aangeeft dat de Rab10 is phosphorylated alleen op Thr73. Het grote voordeel van P-tag SDS-pagina over immunoblotting met fosforylatie-specifieke antilichamen is dat gefosforyleerd Rab10 kan worden gedetecteerd door immunoblotting met niet-fosforylatie-specifieke antilichamen (d.w.z., herkennen totale Rab10) Na overgedragen worden op membranen, dat is over het algemeen meer specifieke, gevoelige en beschikbaar uit commerciële/academische bronnen. Een ander voordeel van het gebruik van de P-tag SDS-pagina is dat men benaderende raming van de stoichiometrie van fosforylering verkrijgen kan, wat onmogelijk door immunoblotting met fosforylatie-specifieke antilichamen of metabole labeling van cellen met radioactieve is fosfaten.
Afgezien van het gebruik van goedkope P-tag acrylamide en een aantal kleine wijzigingen daaraan, volgt de huidige methode voor de detectie van Rab10 fosforylering door LRRK2 een algemeen protocol voor immunoblotting.Daarom moet het eenvoudig en gemakkelijk uitvoerbare in de laboratoria waar immunoblotting is een gebruikelijke praktijk, met alle soorten monsters met inbegrip van gezuiverde eiwitten, cel lysates en weefsel homogenates.

rab10 phosphorylation detection by lrrk2 activity using sds-page with a phosphate-binding tag

Mutaties in leucine-rijke herhalen kinase 2 (LRRK2) hebben aangetoond dat het familiale Parkinson's disease (FPD) worden gekoppeld. Aangezien abnormale activering van het kinaseactiviteit van LRRK2 is betrokken bij de pathogenese van PD, is het essentieel een methode om te evalueren van de fysiologische niveaus van het kinaseactiviteit van LRRK2 vast te stellen. Recente studies is gebleken dat LRRK2 kinaseenzym leden van de Rab GTPase-familie, waaronder Rab10, onder fysiologische omstandigheden. Hoewel de fosforylatie van endogene Rab10 door LRRK2 in gekweekte cellen kon worden opgespoord door massaspectrometrie, is het moeilijk om het te ontdekken door immunoblotting vanwege de slechte gevoeligheid van momenteel beschikbare fosforylatie-specifieke antilichamen voor geweest Rab10. Hier beschrijven we een eenvoudige methode voor het opsporen van de endogene niveaus van fosforylering van de Rab10 door LRRK2 van gebaseerd op immunoblotting met behulp van elektroforese natrium dodecyl sulfaat polyacrylamidegel (SDS-PAGE) gecombineerd met een fosfaat-bindende tag (P-tag), die N-(5-(2-aminoethylcarbamoyl)pyridin-2-ylmetyl) -N,N',N'- tris (pyridin-2-yl-methyl) - 1,3 - diaminopropan-2-ol. Dit protocol biedt niet alleen een voorbeeld van de methode met behulp van de P-tag maar maakt het ook mogelijk de beoordelingvan hoe mutaties alsmede remmer behandeling/administratie of andere factoren veranderen de stroomafwaartse signalering van LRRK2 in cellen en weefsels .

Overexpressie systeem: Fosforylatie van HA-Rab10 door 3 × vlag-LRRK2 in de HEK293 cellen:
