PD è una delle più comuni malattie neurodegenerative, che interessa principalmente i neuroni dopaminergici nel midbrain, conseguente disfunzione dei sistemi motore in persone anziane1. Mentre la maggior parte dei pazienti sviluppano PD in maniera sporadica, ci sono famiglie di ereditare la malattia. Mutazioni in diversi geni sono state trovate per essere collegati con FPD2. Uno dei geni causativi per FPD è LRRK2, in cui otto mutazioni di senso sbagliato (N1437H, R1441C/G/H/S, Y1699C, G2019S e I2020T) collegate a un dominante ereditato FPD chiamato PARK8 finora sono stati segnalati3,4,5. Diversi studi di associazione genome-wide (GWAS) di pazienti sporadici di PD sono identificati varianti genomiche del locus LRRK2 come un fattore di rischio per PD, suggerendo che l'anomalia nella funzione di LRRK2 è una causa comune di neurodegenerazione in entrambi sporadici e PARK8 FPD6,7,8.
LRRK2 è una grossa proteina (2.527 aminoacidi) costituito da un dominio ripetizione leucina-ricche, un Ras GTP-legante del dominio di proteine complesse (ROC), un C-terminale del dominio, un dominio della chinasi serina/treonina proteina e un dominio ripetere di WD409ROC (COR). Fosforilazione della proteina viene generalmente rilevata dal immunoblotting o enzima-collegata dell'immunosorbente (ELISA) utilizzando anticorpi che riconoscono specificamente lo stato fosforilato di proteine o dall'analisi di spettrometria di massa. Tuttavia, la strategia ex a volte non può essere applicata a causa delle difficoltà nella creazione di anticorpi specifici per fosforilazione. Etichettatura metabolica delle cellule con fosfato radioattivo è un'altra opzione per esaminare i livelli fisiologici di fosforilazione quando fosforilazione specifici anticorpi non sono prontamente disponibili. Un sottoinsieme della famiglia Rab GTPase tra cui Rab10 e Tab8 recentemente è stato segnalato come substrati fisiologici diretti per LRRK2 basano sul risultato di analisi su larga scala fosfoproteomica15. Abbiamo quindi dimostrato che la fosforilazione Rab10 è stata aumentata da mutazioni di FPD in fibroblasti embrionali di topo e nei polmoni dei topi16di knockin. Non siamo riusciti a rilevare la fosforilazione di endogeno Tab8 a causa della scarsa selettività di anticorpi attualmente disponibili per Tab8 totale. La fosforilazione di Rab8A di LRRK2 inibisce il legame di Rabin8, un fattore di scambio di nucleotide della guanina (GEF) che attiva Rab8A scambiando il PIL associato con GTP15. Fosforilazione di Rab10 e Rab8A di LRRK2 inibisce anche il legame degli inibitori di PIL-dissociazione (GDIs), che sono essenziali per l'attivazione delle proteine Rab estraendo le proteine Rab PIL associati da membrane15. Collettivamente, è supposto che la fosforilazione delle proteine Rab di LRRK2 impedisce loro di attivazione anche se il preciso meccanismo molecolare e conseguenze fisiologiche della fosforilazione rimangono poco chiare.
P-tag SDS-PAGE è stato inventato da Kinoshita et nel 2006: In questo metodo, acrilammide in covalenza è stato accoppiato con tag P, una molecola catturando fosfati con alta affinità, che copolymerized in SDS-PAGE gel17. Poiché le molecole di P-tag in un gel di SDS-PAGE ritardano selettivamente mobilità elettroforetica delle proteine fosforilate, tag P SDS-PAGE possibile separare proteine fosforilate da quelli non-fosforilate (Figura 1). Se la proteina di interesse è fosforilata su residui multipli, sarà osservata una scaletta delle bande corrispondenti alle forme differenzialmente fosforilate. Nel caso di Rab10, osserviamo soltanto una fascia spostata, che indica che Rab10 è fosforilata solo a Thr73. Il vantaggio principale di SDS-PAGE tag P sopra immunoblotting con anticorpi specifici per fosforilazione è che Rab10 fosforilata può essere rilevato dall'immunoblotting con gli anticorpi non specifici di fosforilazione (cioè, riconoscendo Rab10 totale) Dopo essere stato trasferito sulle membrane, che è generalmente più specifico, sensibile e disponibile da fonti commerciali/accademico. Mutazioni in leucina-ricche ripetere chinasi 2 (LRRK2) sono state indicate per essere collegati con la malattia del Parkinson familiare (FPD). Poiché l'attivazione anormale di attività della chinasi di LRRK2 è stato implicato nella patogenesi del PD, è essenziale per stabilire un metodo per valutare i livelli fisiologici di attività della chinasi di LRRK2. Il presente protocollo non solo fornisce un esempio della metodologia che utilizza il tag P ma consente anche la valutazione di come le mutazioni così come inibitore trattamento/amministrazione o altri fattori alterano la segnalazione a valle di LRRK2 in cellule e tessuti .
