PD é uma das mais comuns doenças neurodegenerativas, predominantemente, afetando os neurônios dopaminérgicos no mesencéfalo, resultando em disfunção dos sistemas de motor em idosos1. Enquanto a maioria dos pacientes desenvolvem PD de forma esporádica, há famílias herdar a doença. Mutações em vários genes foram encontradas para ser ligado com FPD2. Um dos genes causadores de FPD é LRRK2, em que oito mutações missense (N1437H, R1441C/G/H/S, Y1699C, G2019S e I2020T), ligadas a um FPD predominantemente hereditária chamada PARK8 até agora têm sido relatados3,4,5. Vários estudos de associação de genoma-larga (GWAS) de pacientes com DP esporádicos também identificaram variações genômicas no locus LRRK2 como um fator de risco para a polícia, sugerindo que a anormalidade na função do LRRK2 é uma causa comum de neurodegeneração em ambos os casos esporádicos e PARK8 FPD6,7,8.
LRRK2 é uma proteína grande (2.527 aminoácidos), consistindo de um domínio de repetição rico em leucina, um Ras-GTP-vinculação do domínio de proteínas complexas (ROC), um C-terminal do domínio, um domínio da quinase serina/treonina proteína e um domínio de WD409ROC (CR). As oito mutações FPD localizar estes domínios funcionais; N1437H e R1441C/G/H/S no domínio do ROC, Y1699C no domínio do CR, G2019S e I2020T no domínio de cinase. Desde que a mutação G2019S, que é o mais frequentemente encontrada mutação em PD pacientes10,11,12, aumenta a atividade da quinase de LRRK2 por 2-3 vezes em vitro13, supor que o aumento anormal da fosforilação de LRRK2 substrate(s) é tóxico para os neurônios. No entanto, tem sido impossível estudar se a fosforilação de substratos LRRK2 fisiologicamente relevantes é alterada em pacientes com DP familiar/esporádicas devido à falta de métodos para avaliá-lo em amostras derivadas de pacientes.
Fosforilação de proteínas é geralmente detectada pelo immunoblotting ou ensaio imunoenzimático (ELISA), utilizando anticorpos especificamente, reconhecendo o estado fosforilado de proteínas ou por análise de espectrometria de massa. No entanto, a estratégia anterior às vezes não pode ser aplicada devido às dificuldades em criar anticorpos específicos de fosforilação. Etiquetando metabólica das células com fosfato radioativo é outra opção para examinar os níveis fisiológicos de fosforilação quando anticorpos específicos fosforilação não estão prontamente disponíveis. No entanto, ele requer uma grande quantidade de materiais radioativos e, portanto, envolve alguns equipamentos especializados para protecção contra as radiações14. No entanto, a preparação da amostra é demorada e caros instrumentos são necessários para a análise.
Um subconjunto da família Rab GTPase, incluindo Rab10 e Rab8 foi relatado recentemente como substratos fisiológicos diretos para LRRK2 com base no resultado de uma análise de phosphoproteomic em grande escala15. Em seguida demonstramos que a fosforilação de Rab10 foi aumentada por mutações FPD em fibroblastos embrionários de rato e nos pulmões de ratos16de knockin. A fosforilação de Rab8A por LRRK2 inibe a ligação do Rabin8, um fator da troca do nucleotídeo guanina (GEF) que ativa Rab8A trocando o PIB ligado com GTP15. A fosforilação de Rab10 e Rab8A por LRRK2 também inibe a ligação dos inibidores de PIB-dissociação (GDIs), que são essenciais para a ativação de proteínas Rab extraindo as proteínas Rab ligados a PIB de membranas15. Coletivamente, supor que a fosforilação de proteínas Rab por LRRK2 impede que a ativação embora o mecanismo molecular preciso e consequências fisiológicas da fosforilação permanecem obscuros.
P-marca SDS-PAGE foi inventado por Kinoshita et al em 2006: neste método, acrilamida covalentemente foi acoplada com P-tag, uma molécula capturando fosfatos com alta afinidade, que copolymerized em SDS-PAGE geles17. Porque as moléculas de P-etiqueta em um gel de SDS-PAGE seletivamente retardam mobilidade eletroforética de proteínas fosforiladas, P-marca SDS-PAGE pode separar proteínas fosforiladas aqueles não-fosforilada (Figura 1). Se a proteína de interesse é fosforilado em múltiplos resíduos, uma escada de bandas correspondentes às formas diferencialmente fosforiladas será observada. No caso de Rab10, observamos apenas uma banda deslocada, indicando que o Rab10 é phosphorylated apenas em Thr73. A grande vantagem de P-marca SDS-PAGE sobre immunoblotting com anticorpos específicos fosforilação é que Rab10 fosforilada pode ser detectado pelo immunoblotting com anticorpos de fosforilação específico (ou seja, reconhecendo Rab10 total) após ser transferido nas membranas, que é geralmente mais específica, sensível e disponível de fontes comerciais/acadêmico. Outra vantagem de usar P-marca SDS-PAGE é que um pode obter a estimativa aproximada da estequiometria da fosforilação, que é impossível pelo immunoblotting com anticorpos específicos fosforilação ou através da marcação metabólica das células com radioativo fosfatos.
Aparte o uso de barato P-marca acrilamida e algumas pequenas modificações relacionadas a ela, o presente método de detecção de fosforilação de Rab10 por LRRK2 segue um protocolo geral de immunoblotting.Portanto, deve ser simples e facilmente executável em alguns laboratórios onde immunoblotting é uma prática usual, com todos os tipos de amostras, incluindo proteínas purified, lisados celulares e tecido homogenates.

Mutações no rico em leucina repetição quinase 2 (LRRK2) foram mostradas para ser associada com a doença de Parkinson familiar (FPD). Como ativação anormal da atividade da quinase de LRRK2 tem sido implicada na patogênese da DP, é essencial estabelecer um método para avaliar os níveis fisiológicos da atividade da quinase de LRRK2. Estudos recentes revelaram que LRRK2 fosforila membros da família Rab GTPase, incluindo Rab10, sob condições fisiológicas. Embora a fosforilação de Rab10 endógeno por LRRK2 em pilhas cultivadas pode ser detectada por espectrometria de massa, tem sido difícil detectá-lo pelo immunoblotting devido à sensibilidade pobre de anticorpos de fosforilação específicos disponíveis atualmente para Rab10. Aqui, descrevemos um simples método de detectar os níveis endógenos de fosforilação de Rab10 por LRRK2 baseado na immunoblotting utilizando sódio Dodecil sulfato poliacrilamida electroforese do gel (SDS-PAGE) combinado com uma marca de ligação de fosfato (P-tag), que é N-(5-(2-aminoethylcarbamoyl)pyridin-2-ylmetyl) -N,N «,N «- tris (piridin-2-yl-metil) - 1,3 - diaminopropan-2-ol. O presente protocolo não só fornece um exemplo da metodologia utilizando a tag-P, mas também permite a avaliação de como mutações, bem como inibidor de tratamento/administração ou quaisquer outros fatores alteram a sinalização a jusante de LRRK2 em células e tecidos .
