La méthode présentée ici montre comment la combinaison de deux photons Ca2 + imagerie et électrophysiologie patch clamp peut être utilisée pour l’étude dendritiques Ca2 + la signalisation dans les dendrites neuronales dans des tranches de cerveau aiguë. La contribution d’un neurone à l’activité du réseau est en grande partie déterminée par la nature dynamique des entrées synaptiques qu’il reçoit. Traditionnellement, la principale méthode de caractérisation de l’activité synaptique dans les neurones s’est fondé sur les enregistrements de patch-clamp de la cellule entière somatique des courants postsynaptiques évoqués par stimulation électrique des axones de passage. Toutefois, la seule activité des synapses situées dans la partie proximale est véridiquement signalée dans ce cas1. En outre, afin d’évaluer les mécanismes spécifiques à synapse, enregistrements de paires de neurones et de dendrites à un endroit précis ont été utilisés pour cibler les synapses d’intérêt et les mécanismes d’intégration dendritique, respectivement. Une percée majeure dans le domaine de la physiologie synaptique a été atteint par un mariage des techniques optiques et électrophysiologiques. Excitation biphotonique microscopie à balayage laser (2PLSM) en combinaison avec les outils d’optogenetic et de Ca2 + imagerie peut révéler des détails extraordinaires de l’organisation dynamique de l’activité synaptique à connexions neuronales spécifiques dans des tranches de cerveau dans vitro et in vivo.
Plusieurs avantages principaux faits 2PLSM se démarquer de l’excitation classiques, un photon microscopy2: (1) en raison de la nature non linéaire d’excitation à deux photons, la fluorescence est générée uniquement dans le volume focal, et représentent tous les photons émis signaux utiles (pas besoin de trou d’épingle) ; (2) longueurs d’onde, utilisés en 2PLSM, pénètrent dans les tissus de diffusion plus efficacement ; en outre, les photons dispersés sont trop diluées pour produire l’excitation biphotonique et fluorescence de fond ; (3) le photovieillissement et phototoxicité sont également limités à plan focal. Par conséquent, malgré le coût élevé des systèmes 2 photons par rapport aux microscopes confocaux classiques, la 2PLSM reste une méthode de choix pour enquête à haute résolution de structure neuronale et la fonction dans les tissus vivants épais.
La première application de 2PLSM dans les tissus de diffusion a été à l’image de la structure et la fonction des épines dendritiques3. 2PLSM en combinaison avec Ca2 + imagerie a révélé cette fonction d’épines comme des compartiments isolés biochimiques. Puisque de nombreux types de neurones, il y a une correspondance biunivoque entre les épines et les synapses individuels4, deux photons Ca2 + imagerie bientôt est devenu un outil de reporting de l’activité des synapses individuelles en tissu intact5, 6,7,8. En outre, 2PLSM Ca2 + imagerie a été utilisée avec succès pour surveiller l’activité des canaux calciques unique et les interactions non linéaires entre les conductivités intrinsèques et synaptiques, ainsi que d’évaluer l’État et l’activité dépendante- régulation du Ca2 + signalisation neuronale dendrites5,6,7,8,9,10,11.
Le calcium est un omniprésent second messager intracellulaire, et son organisation spatio-temporelle subcellulaire détermine la direction de réactions physiologiques, de modifications de la force synaptique à la régulation des ions canaux, croissance dendritique et la colonne vertébrale, comme Bien que la mort cellulaire et la survie. Dendritiques Ca2 + élévations se produisent via l’activation de multiples voies. Deux photons Ca2 + imagerie en combinaison avec les enregistrements électrophysiologiques de patch clamp emploie synthétique Ca2 +-indicateurs fluorescents, qui sont généralement livrés par le biais de l’électrode de patch pendant les enregistrements de cellules entières . Une méthode standard pour la quantification de Ca2 + dynamique repose sur l’indicateur de double méthode17,18. Il utilise deux fluorophores avec des spectres d’émission bien séparées (par exemple, une combinaison d’un rouge Ca2 +-insensible colorant vert Ca2 + indicateurs, tels que l’Oregon vert BAPTA-1 ou Fluo-4) et présente plusieurs avantages par rapport à la méthode de l’indicateur unique. Tout d’abord, un Ca2 +-colorant insensible est utilisé pour localiser les petites structures d’intérêt (ramifications dendritiques et épines) où l’imagerie2 + Ca sera effectuée. Deuxièmement, le rapport entre la variation de la fluorescence verte et rouge (ΔG/R) est calculé en guise de [Ca2 +], qui est pratiquement insensible aux variations de fluorescence de base en raison de fluctuations [Ca2 +]017 , 18. en outre, les changements de fluorescence peuvent être calibrés en termes absolus, Ca2 + concentration19.
Une préoccupation générale lors de l’exécution de deux photons Ca2 + imageries expériences en tranches aiguës est la cellule santé et la stabilité de l’acquisition d’image en raison de la puissance du laser haute généralement utilisée. En outre, dans les2 + expériences d’imagerie, de Ca on se préoccupe de la perturbation et l’importante surestimation de subcellulaire Ca2 + dynamique dû au fait que servent d’indicateurs de Ca2 + très mobiles exogène Ca2 + mémoires tampons. Nous avons adapté la méthode biphotonique Ca2 + imagerie pour recherche de Ca2 + fluctuations de dendrites de GABAergique interneurones9,10,11,20,21 , 22. alors que la majeure partie des premiers Ca2 + études d’imagerie ont été effectuées dans les principaux neurones, interneurones inhibiteurs présenter une grande variété de mécanismes2 + Ca fonctionnels qui sont distincts de ceux des cellules pyramidales20 , 23 , 24. ces mécanismes interneurone spécifiques (par exemple, Ca2 + perméables récepteurs AMPA) pourraient jouer un rôle spécifique dans la régulation de l’activité des cellules. Dendritiques Ca2 + signalisation dans des interneurones est une cible tentante pour complément d’enquête, deux photons Ca2 + imagerie dans les dendrites des cellules présente des difficultés supplémentaires, d’un diamètre plus mince des dendrites et manque d’épines pour une particulièrement élevé Ca2 + liaison capacité endogène. Comme nos recherches portent sur l’étude des interneurones hippocampe, le protocole suivant, alors qu’il y a lieu à différentes populations de neurones, a été adapté pour faire face à ces défis.
Ce protocole a été exécuté avec un microscope confocal de deux photons commercial, qui était équipé de deux détecteurs externes, non-descanned (NDDS™), modulateur électro-optique (MOE) et un Dodt balayage dégradé contraste (CGT) et installé sur une table optique. Le microscope a été couplé avec un laser Ti : Sapphire de multiphoton mode bloqué à 800 nm (> 3 W, 140 impulsions fs, 80 Hz fréquence de répétition). Le système d’imagerie était équipé d’une plate-forme d’électrophysiologie standard, y compris une chambre de perfusion avec contrôle de température, une plateforme de traduction avec deux micromanipulateurs, un amplificateur de la microélectrode contrôlé par ordinateur, un numériseur, une stimulation appareil et logiciel d’acquisition de données.

two-photon calcium imaging in neuronal dendrites in brain slices

Imagerie de calcium (Ca2 +) est un outil puissant pour étudier la dynamique spatio-temporelle de Ca2 + signaux intracellulaires dans les dendrites neuronales. Ca2 + fluctuations peuvent se produire à travers une variété de membrane et mécanismes intracellulaires et jouent un rôle crucial dans l’induction de la plasticité synaptique et la régulation de l’excitabilité dendritique. La capacité d’enregistrer différents types de signaux de Ca2 + en ramifications dendritiques est donc précieuse pour des groupes d’étudier comment dendrites intègrent l’information. L’avènement de la microscopie biphotonique a fait ces études beaucoup plus facile en résolvant les problèmes inhérents à l’imagerie dans les tissus vivants, tels que la diffusion de la lumière et le photovieillissement. En outre, grâce à la combinaison de techniques électrophysiologiques classiques avec deux photons Ca2 + imagerie, il est possible d’étudier Ca2 + fluctuations locales dans les dendrites neuronales en parallèle avec les enregistrements de l’activité synaptique Soma. Nous décrivons ici comment utiliser cette méthode pour étudier la dynamique des locaux Ca2 + transitoires (chats) dans les dendrites des interneurones inhibiteurs GABAergiques. La méthode peut être également appliquée à l’étude dendritiques Ca2 + signalisation dans différents types de neurones dans des tranches de cerveau aiguë.

En utilisant le protocole présenté ici, nous avons obtenu des chats évoquées par injection de courant somatique et par la stimulation électrique dans les dendrites des interneurones oriens/alveus dans la région CA1 de l’hippocampe. Imagerie biphotonique Calcium en Dendrites neuronales dans des tranches de cerveau

The overall goal of this experiment is to study calcium elevations in neuronal dendrites in response to different stimulation paradigms using whole-cell patch-clamp recordings in neuronal soma in parallel with two-photon calcium imaging in dendrites. This method can be employed to monitor calcium signaling in small dendritic compartments, allowing for close observation of the processes that occurs in dendritic branches during integration of synaptic inputs. The main advantage of this technique is that it enables calcium signaling patterns to be observed in high spatiotemporal resolution and specificity.Place the mouse brain onto a Petri dish containing continuously oxygenated, cold ACSF-sucrose. To prepare hippocampal slices from the extracted brain, let the brain chill for around two minutes. Afterward, place a piece of filter paper on an ice-packed Petri dish, and transfer the brain onto the filter paper.Then, dissect the brain into two hemispheres. Glue the dissected hemispheres onto a vibratome sectioning platform, and immerse it into the sectioning tray filled with ice-cold oxygenated ACSF-sucrose. Subsequently, section 300-micrometer-thick slices at one degree Celsius using a vibratome cooler or placing the ice around the vibratome tray.Using a paint brush, transfer the slices containing the hippocampus to a bath of oxygenated ACSF-recovery heated to 37 degrees Celsius. In this step, transfer a hippocampal slice to a bath under the objective of a laser-scanning two-photon microscope. Continuously perfuse the bath with oxygenated ACSF-normal.Then, use infrared differential interference contrast microscopy to locate a cell of interest using its size, shape, and position. After that, fill a stimulating pipette with an ACSF-normal solution containing the red fluorophore. Lower it on top of the slice so that the tip is in the same region as the cell of interest.Following that, fill the patch pipette with patch solution, and attach it to the head stage. Set it over the slice so that its tip is directly above the cell of interest. Subsequently, fit a syringe into the three-way stopcock, and connect it to the patch pipette through a plastic tube.Inject constant positive pressure into the patch pipette, and keep the stopcock in close position. Next, open the MultiClamp amplifier control software module, and switch to Voltage Clamp mode by clicking on the VC button. Open the electrophysiology Clampex data acquisition software for recording electrophysiological signals, and click on the Membrane Test icon to send out a constant square voltage pulse for monitoring changes in the pipette resistance.Now, lower the patch pipette until it is right on top of the targeted cell. Upon making contact with the cell membrane, remove the pressure by turning the stopcock into the open position. Then, apply a slight negative pressure to the patch pipette using an empty syringe fitted into the stopcock until the pipette resistance reaches one gigaohm.In the Membrane Test window of the data acquisition software, clamp the cell at negative 60 millivolts. Continue applying negative pressure until the pipette resistance drops and the whole-cell configuration is achieved. In the MultiClamp amplifier control software module, set the patch configuration to current clamp by clicking on the IC button, and record membrane properties in response to somatic injections of hyperpolarizing and depolarizing currents.Wait for at least 30 minutes for the cell to be filled with the indicators present in the patch solution. To acquire images, locate a dendrite of interest using the red fluorescence signal. Ensure a visible response by choosing a proximal dendrite shorter than 50 micrometers, as the efficiency of AP backpropagation may significantly decline with distance in GABAergic interneurons, and switch to the xt mode.With the laser intensity controllers, set the two-photon laser at a minimal power level where baseline green fluorescence is just slightly visible, to avoid phototoxicity. Subsequently, in the Clampex electrophysiology data acquisition software Protocol window and image acquisition software, create a recording trial of the desired duration containing a somatic current injection of the desired amplitude. After that, click the Start Record button, and acquire the fluorescence continuously for one to two seconds.Repeat the image acquisition three to 10 times. Wait at least 30 seconds between single scans to avoid photodamage. To record dendritic calcium transients induced by electrical stimulation, locate a dendrite of interest using the red fluorescence signal.Set the stimulating pipette on the surface of the slice above the dendrite of interest. Slowly lower the stimulation pipette 10 to 15 micrometers from the dendrite, minimizing movement to avoid disturbing the whole-cell configuration. To visualize the location of synaptic microdomains in aspiny neurons, in the image acquisition software, switch to the xt mode and position the line along the dendritic branch of interest.In the Protocol window, create a recording trial that triggers the stimulation. Using the acquisition software, scan along the dendrite continuously for one to two seconds. Repeat acquisition three to five times, waiting 30 seconds between scans to prevent photodamage.To obtain preliminary information on the cell's morphology and keep a record of the location of the stimulation pipette, acquire a Z-stack of the cell in the red channel. Using the acquisition software in xyz mode, set the upper and lower stack limits to image the entire cell with all processes included. Then, set the step size at one micrometer, and initiate the stack acquisition using the Start Record button.When the Z-stack acquisition is complete, use the Maximum Projection option in the software to superimpose all focal plans of the stack and verify the quality of acquisition. Then, slowly retract the patch pipette out of the slice. To fix the slice for post hoc morphological identification, quickly remove the slice from the bath using a paint brush, and place it between two filter papers in an ACSF-filled Petri dish.Replace ACSF with a 4%paraformaldehyde solution, and leave the dish in a four degrees Celsius overnight. In this figure, the maximum projection of a two-photon Z-stack shows a patched interneuron filled with Alexa-594. White arrowheads point to the location of axonal terminals within the pyramidal layer, suggesting the interneuron is a basket cell.This figure shows the dendritic branch near which the stimulating electrode was positioned. The dashed line shows the location of the line scan. The images here illustrate the result of a line scan in the red and green channels.The rise in green fluorescence at the time of stimulation indicates a rise in calcium concentration in that area. The images can be binned into smaller segments, allowing the analysis of the spread of the response. The traces seen here show the calcium transients elicited in different segments in response to electrical stimulation recorded at the somatic level during the imaging trial.The amplitude of the calcium transients declines with distance from the central hotspot, indicating that the response is spatially localized. Following this procedure, other methods like voltage-sensitive dye imaging or optogenetics can be performed in order to answer additional questions, like local changes in the memory potential within dendritic branches or integration of specific inputs. After watching this video, you should have a good understanding of how to monitor activity-dependent calcium fluctuations in neuronal dendrites using a combination of optophysiological techniques, which will be useful to explore the intricacies of dendritic input integration and plasticity.
