Ce protocole a été conçu pour maximiser la génération des espèces moléculaires ionisables tout en éliminant les délocalisation d’analytes. Les principaux facteurs impliquent en utilisant le même principe primordial lors de l’application de matrice, digérer l’échantillon ou recristalliser après sublimation24; savoir, qu’un même dépôt de vapeur, de matrice ou autrement, doit être créée et maintenue. Pipetage solvants nettoyants pour recristallisation et digestion, uniform?...

Spectrométrie de masse à désorption-ionisation laser assistée par matrice (MALDI MSI) d’imagerie a été employée comme technique d’image basé pendant deux décennies1,2, analyser une gamme de biomolécules, y compris : lipides3, peptides2 ,4, protéines2,5, métabolites6,7, N-glycans8et des molécules synthétiques tels que les médicaments thérapeutiques9,10. Le nombre de publications démontrant l’utilité de cette technique ont fortement augmenté au cours de la dernière décennie6,11,12,13. Certaines molécules, comme les lipides, sont relativement faciles à analyser par MALDI MSI, comme ils ionisent facilement en raison de leur nature chimique et nécessitent peu de préparation à l’avance3. Toutefois, pour des cibles plus difficiles comme les peptides, les étapes requises pour ioniser efficacement ces molécules sont vastes et généralement compliqué14. Il y a actuellement très peu de publications qui visent à répondre ou à démontrent la reproductibilité dans les méthodologies utilisées pour préparer les tissus à cette technique visuelle unique15. Pour cette raison, nous avons compilé les observations et mises en oeuvre des optimisations en un seul, facile à implémenter, méthodologie ne nécessite peu ou aucun modifications, pour l’analyse des peptides d’un formaldéhyde que réticulé tissu source14.
Dans ce manuscrit, nous avons décrit une méthodologie reproductible validée, à faible coût pour la détection et la cartographie spatiale des peptides, produit des surgelés fixés au formol (FFF) et fixés au formol des sections de tissu (FFIP) inclus en paraffine. Cette méthodologie ne nécessitent ni s’appuyer sur une instrumentation spécialisée3. Plus précisément, nous abordons les nombreux aspects de la préparation de l’échantillon spécialisées nécessaire pour analyser les peptides ; étapes comme antigène récupération16 et revêtement de matrice. Notre protocole utilise aussi peu coûteux matériel et réactifs, ce qui rend cette méthode accessible à toute une communauté qui autrement seraient incapable de payer l' autre appareils robotisés17.
Le raisonnement qui sous-tend l’élaboration d’une méthode de préparation d’échantillon manuel comportait deux volets : Premièrement, l’utilisation d’un sublimator crée un revêtement uniforme et homogène des cristaux de matrice qui sont environ 1 µm de longueur18, quelque chose d’irréalisables avec arrosage plus fréquent techniques. Deuxièmement, l’ensemble relativement faible des coûts : le coût total de l’appareil de mesure était < 1500 $ AUD. Nous notons, en termes de rapport coût-efficacité, que le prix par exemple est beaucoup moins cher quand il n’y a aucun mécanisme robotique impliqué. L’utilisation de sublimation a été signalée précédemment, cependant, au meilleur de nos connaissances, des méthodes pas à pas qui décrivent ce processus et préparation des échantillons n’ont pas été signalés ni décrits dans la littérature.
Ce protocole vise à aider les chercheurs qui ont accès à un spectromètre de masse MALDI et qui ont l’intention sur le développement de l’information spatiale en relation avec une bio-molécule d’intérêt19. En substance, MALDI MSI est une forme de histologique préalable qui ne s’appuie pas sur les anticorps ou taches2.

optimal preparation of formalin fixed samples for peptide based matrix assisted laser desorption/ionization mass spectrometry imaging workflows

L’utilisation de désorption-ionisation laser assistée par matrice, spectrométrie de masse (MALDI MSI) d’imagerie a rapidement élargi, car cette technique analyse une foule de biomolécules de médicaments et de lipides à N-glycans. Bien qu’il existent de différentes techniques de préparation d’échantillon, détection des peptides de formaldéhyde préservé des tissus reste l’un des défis plus difficiles pour ce type d’analyse par spectrométrie de masse. Pour cette raison, nous avons créé et optimisé une méthodologie robuste qui conserve les informations spatiales contenues dans l’échantillon, tout en suscitant le plus grand nombre de peptides ionisables. Nous avons visait également à atteindre cet objectif de manière simple et rentable, éliminant ainsi l’erreur potentielle de partialité ou de la préparation, qui peut se produire lorsque l’utilisation automatisée instrumentation. Le résultat final est un protocole reproductible et peu coûteux.

Si suivies correctement, ce protocole produit des images qui représentent clairement la morphologie générale du tissu sans les égratignures ou autres déformations (Figure 1). La validation idéale pour une préparation de l’échantillon correctement exécutée, est la capacité de distinguer entre les différentes structures physiques en changeant la molécule étant lus (Figure 2).
