Matrix-unterstützte Laser Desorption/Ionisierung Massenspektrometrie imaging (MALDI MSI) als Bild-basierte Technik für zwei Jahrzehnte1,2, eine Reihe von Biomolekülen einschließlich Analyse beschäftigt: Lipide3, Peptide2 ,4, Proteine2,5, Metaboliten6,7, N-Glykane8und synthetische Moleküle wie z. B. Medikamente9,10. Die Anzahl der Publikationen zeigen die Nützlichkeit dieser Technik sind deutlich über der letzten Dekade6,11,12,13gewachsen. Bestimmte Moleküle wie Lipide, sind relativ einfach, über MALDI MSI zu analysieren, wie sie leicht aufgrund ihrer chemischen Natur ionisieren und erfordern daher wenig Vorbereitung3. Jedoch sind die Schritte erforderlich, um effektiv diese Moleküle ionisieren für schwieriger Ziele wie Peptide, umfangreich und in der Regel kompliziert14. Derzeit gibt es nur wenige Veröffentlichungen, die darauf abzielen, Adresse oder demonstrieren Reproduzierbarkeit in den Methoden, die eingesetzt werden, um Gewebe für dieses einzigartige visuelle Technik15vorzubereiten. Aus diesem Grund haben wir zusammengestellt, Beobachtungen und Optimierungen umgesetzt in einer einzigen, einfach zu implementieren, Methodik, die wenig bis gar keine Änderung für die Analyse von Peptiden aus einem Formaldehyd erfordern sollte vernetzt Gewebe Quelle14.
In dieser Handschrift haben wir eine validierte, low-cost reproduzierbare Methode zum Nachweis und zur räumlichen Zuordnung von Peptiden, generiert von Formalin fixiert gefroren (FFF) und Formalin fixierten Paraffin-eingebetteten (FFPE) Gewebeschnitte beschrieben. Diese Methode erfordert und verlassen Sie sich auf spezielle Instrumentierung3. Insbesondere gehen wir auf die vielen Aspekte der spezialisierten Probenvorbereitung notwendig, Peptide zu analysieren; Schritte wie Antigen-Retrieval-16 und Matrix-Beschichtung. Unser Protokoll nutzt auch preiswerte Geräte und Reagenzien, wodurch diese Methodik zugänglich zu einer größeren Gemeinschaft, die sonst nicht die Alternative Roboter Apparat17leisten.
Die Beweggründe für die Entwicklung einer manuellen Probe Vorbereitung Methode hatte zwei Ziele: Erstens die Verwendung von einem Sublimator schafft eine konsistente und homogene Beschichtung von Matrix-Kristalle, ~ 1 µm Länge18, etwas Unerreichbares mit häufiger Spritzen sind, Techniken. Zweitens die relativ kleine Einrichtung Kosten: die Gesamtkosten für den benutzerdefinierten Apparat war <$ 1500 AUD. Wir beachten Sie, dass in Bezug auf Wirtschaftlichkeit, der Preis pro Probe viel billiger ist, wenn keine Roboter Maschinen beteiligt ist. optimal preparation of formalin fixed samples for peptide based matrix assisted laser desorption/ionization mass spectrometry imaging workflows

Die Verwendung von Matrix-unterstützte Laser Desorption/Ionisierung hat Massenspektrometrie imaging (MALDI MSI) schnell erweitert, da diese Technik eine Vielzahl von Biomolekülen von Drogen und Lipide zu N-Glykane analysiert. Obwohl verschiedene Probe Präparationstechniken vorhanden, bleibt Erkennung Peptide aus Formaldehyd konserviert Gewebe eine der schwierigsten Herausforderungen für diese Art der massenspektrometrischen Analyse. Aus diesem Grund haben wir erstellt und optimiert eine robuste Methode, die die räumliche Informationen innerhalb der Probe, während die größte Anzahl an ionizable Peptide zu entlocken bewahrt. Wir haben auch zum Ziel, zu erreichen dies in eine kostengünstige und einfache Möglichkeit, wodurch möglichen Voreingenommenheit oder Vorbereitung Fehler, die auftreten kann, wenn mit Instrumenten automatisierten. Das Endergebnis ist eine reproduzierbare und kostengünstige Protokoll.

Wenn korrekt befolgt, produziert dieses Protokoll Bilder, die eindeutig die grobe Morphologie des Gewebes ohne jegliche Kratzer oder andere Deformationen (Abbildung 1). Die ideale Validierung für eine korrekt durchgeführte Probenvorbereitung ist, dass die Fähigkeit zur Unterscheidung zwischen verschiedenen physikalischen Strukturen durch eine Änderung des Moleküls wird angezeigt (Abbildung 2).
