Matrix-assistert laser desorpsjon/ionisering massespektrometri imaging (MALDI MSI) har vært ansatt som en basert teknikk for to tiår1,2, analysere en rekke biomolecules inkludert: lipider3, peptider2 ,4, proteiner2,5, metabolitter6,7, N-glykaner8og syntetisk molekyler som terapeutiske narkotika9,10. Hvor mange publikasjoner demonstrere nytten av denne teknikken har vokst betydelig de siste tiår6,11,12,13. Visse molekyler, som lipider, er relativt enkle å analysere via MALDI MSI, som de ionisere lett på grunn av deres kjemiske natur og krever derfor litt tidligere forberedelse3. Men for mer vanskelig mål som peptider er trinn kreves for å effektivt ionisere disse molekylene omfattende og generelt kompliserte14. Det er svært få publikasjoner som mål å adresse eller demonstrere reproduserbarhet i metoder som er ansatt for å forberede vev denne unike visuelle teknikk15. Derfor har vi samlet observasjoner og implementert optimaliseringer i én enkelt å implementere, krysskoblet metodikk som bør kreve liten eller ingen endring, for analyse av peptider fra en formaldehyd vev kilde14.
I dette manuskriptet har vi beskrev en validert, lavpris reproduserbar metode av romlige kartlegging av peptider, generert fra formalin-fast frosne (FFF) og formalin-fast parafin-embedded (FFPE) vev inndelinger. Denne metoden krever ikke eller stole på noen spesialisert instrumentering3. Spesielt ta vi mange aspekter av spesialiserte eksempel forberedelse nødvendig å analysere peptider; trinn som antigen henting16 og matrix belegg. Våre protokollen utnytter også billig utstyr og reagenser, og dermed gjør denne metodikken tilgjengelig for et bredere fellesskap som ellers ikke råd til de alternative robot apparater17.
Begrunnelsen bak utviklet en manuell eksempel forberedelse metoden var todelt: først, bruk av en sublimator skaper et enhetlig og homogene belegg av matrix krystaller som er ~ 1 µm lengde18, noe uoppnåelig med mer vanlig sprøyting teknikker. Dernest den relativt liten etableringskostnader: den totale kostnaden av tilpassede apparater var <$ 1500 AUD. Vi merke, i form av kosteffektivitet, prisen per eksempel er langt billigere når det er ingen robotsveising maskiner involvert. Bruk av sublimering har rapportert tidligere, men best av vår kunnskap, trinnvise metoder som beskriver denne prosessen og prøve forberedelse ikke har vært rapportert eller beskrevet i litteraturen.
Denne protokollen er ment å hjelpe forskere som har tilgang til en MALDI masse spectrometer og som er innstilt på å generere romlig informasjon i forbindelse med en bio-molekylet interesse19. I hovedsak er MALDI MSI en form for histologiske screening som ikke er avhengig av antistoffer eller flekker2.

optimal preparation of formalin fixed samples for peptide based matrix assisted laser desorption/ionization mass spectrometry imaging workflows

Bruk av matrix-assistert laser desorpsjon/ionisering, massespektrometri imaging (MALDI MSI) har ekspandert, siden denne teknikken analyserer en rekke biomolecules fra narkotika og lipider til N-glykaner. Selv om ulike eksempel forberedelser teknikker finnes, fortsatt oppdage peptider fra formaldehyd bevart vev en av de vanskeligste utfordringene for denne massen spectrometric analyse. Derfor har vi opprettet og optimalisert robust metode som bevarer romlige informasjonen i utvalget, mens fremlokkende flest ionizable peptider. Vi har også som mål å oppnå dette på en kostnadseffektiv og enkel måte, og dermed eliminere potensielle skjevhet eller forberedelse feil som kan oppstå når ved hjelp av automatisert instrumentering. Sluttresultatet er en reproduserbar og rimelig protokoll.
