De Ca2 +-mobilisatie assay hierin beschreven eerder blijkt te zijn een waardevol instrument te identificeren en te karakteriseren receptorantagonisten targeting CXCR417. Verwacht wordt, echter dat deze methode worden meer in het algemeen op een grote groep van andere GPCRs die leiden een cytosolische Ca2 + release op hun activering, toegepast kan tot zoals geïllustreerd voor de verwante chemokine receptor CCR5. GPCRs zijn een belangrijke superfamilie van cel-oppervlakte-eiwitten die signaaltransductie cascades van extracellulaire ligand bindende activeren. Zij kunnen worden geactiveerd door een grote verscheidenheid van stimuli met inbegrip van peptiden, eiwit hormonen, biogenic amines en lipiden, wat in de opening van uiteenlopende intracellulaire signaalroutes en uiteindelijk biologische reacties1,2 resulteert . Bovendien GPCRs zijn betrokken bij velen, als niet alle, ontwikkelings- en fysiologische processen en vele ziekten bij de mens worden geassocieerd met disfunctionele GPCR signalering of receptor overexpressie. GPCRs behoren daarom tot de meest gevalideerde farmacologische doelen in geneeskunde1,3.
Meestal begint een GPCR workflow voor de ontdekking van de drug met cellulaire screening tests waarmee de identificatie van stoffen zoals kleine molecules, monoklonale antilichamen en peptiden die de activiteit van een bepaalde GPCR kunnen moduleren. In GPCR drugontdekking vele verschillende soorten tests bestaan om te zoeken naar dergelijke verbindingen, zijn waarvan de meeste compatibel met midden - tot high-throughput screening campagnes. De meest gebruikte tests omvatten receptor bindende experimenten, fluorescentie- of luminescentie gebaseerd testen schommelingen in het niveau van de zogenaamde secundaire boodschappers (b.v., Ca2 +, cyclisch adenosinemonofosfaat (AMP)), fenotypische opsporen screening tests, en de aanwerving van de β-arrestin testen4. De keuze voor een bepaald soort assay kan afhangen van meerdere factoren, maar wordt ook bepaald door voorafgaande kennis omtrent de signalering eigenschappen van een bepaalde GPCR. Agonist binden aan een GPCR induceert een conformationele verandering katalyseren van de uitwisseling van guanidine (III) difosfaat (BBP) voor guanidine trifosfaat (GTP) op de α-subunit van heterotrimeric G eiwitten. Vervolgens de Gα-GTP subeenheid distantieert van de G-βγ -subunit en beide subeenheden signaalroutes verder zal starten. Hydrolyse van de GTP-molecuul en de daaropvolgende hernieuwde vereniging van de G-α- BBP en Gβγ subeenheden zal herstellen het G-eiwit in de inactieve conformatie5,6. Gebaseerd op volgorde gelijkenis van de Gα subeenheid verschillende soorten G eiwitten zijn gedefinieerd (Gs, GikGq, G12/13)7. Signalering via de G-α subeenheid geeft aanleiding tot verschillende typische reacties zoals de verhoging (via Gs) of daling (via Gik) van cyclisch ADENOSINEMONOFOSFAAT productie en intracellulaire Ca2 + mobilisatie (via Gq)5 ,7. Gβγ subeenheden kunnen ook voor het opwekken van intracellulaire effector trajecten. Bijvoorbeeld, na activatie van Gik-gekoppelde GPCRs, Gβγ kunnen direct stimuleren phospholipase C (PLC-β) te produceren inositol trifosfaat (IP-3) die als trigger de vrijlating van Ca2 + van intracellulaire winkels7 fungeert . Na receptor activering, zijn GPCRs phosphorylated door GPCR kinases (GRKs) ter bevordering van de interactie met β-arrestins. Dit proces wordt beëindigd G eiwit signalering en leidt tot receptor desensibilisatie en uiteindelijk internalisering. Β-arrestins kunnen ook multi moleculaire complexen die kunnen leiden tot andere signaalroutes onafhankelijk van signalering van8G-proteïne vormen.
Binnen de onderfamilie van chemokine receptoren, de Gik-gekoppelde CXCR4 is een GPCR die veel belangstelling heeft opgeworpen als een veelbelovende doelwit voor drugontdekking. Gelet op haar gevestigde rol als een belangrijke co receptor voor humaan immunodeficiëntie virus (HIV-1) 1 werden virale ingang en infectie, verbindingen richten CXCR4 in eerste instantie ontwikkeld als anti-HIV drug kandidaten9. Meer recentelijk, een groeiend lichaam van bewijsmateriaal heeft gewezen op een belangrijke rol voor CXCR4 tumorvorming en kanker uitzaaiingen, waardoor het een goed gevalideerde therapeutisch doel in oncologie evenals10. CXCR4 wordt sterk uitgedrukt in meer dan twintig soorten menselijke kanker en overleving van de cel van de tumor van besturingselementen, proliferatie, en migratie alsmede tumor-gerelateerde angiogenese10. CXCR4 antagonisten van verschillende chemische klassen geweest eerder beschreven11,12, maar alleen het kleine molecuul dat amd3100 is momenteel goedgekeurd voor gebruik in de kliniek als een stamcel mobilisatie agent gebruikt tijdens de behandeling van het lymfoom en myeloom patiënten13,14. Klinische proeven zijn aan de gang om de veiligheid en de werkzaamheid van verschillende andere CXCR4 antagonisten in verschillende ziekten bij de mens, maar met een sterke focus op oncologie12. Gezien de vele mogelijke toepassingen voor CXCR4 antagonisten, is het zoeken naar nieuwe verbindingen met verbeterde farmacokinetische eigenschappen, verbeterde biologische beschikbaarheid of potentieel minder bijwerkingen gerechtvaardigd.
Hierin een kinetische fluorescentie gebaseerde cellulaire assay hoofdzakelijk gebruikt voor scherm voor verbindingen kunnen remmen CXCR4 wordt beschreven. De fluorescerende meting van deze methode is gebaseerd op de tijdelijke verhoging van de intracellulaire Ca2 + concentratie opgeroepen op CXCR4 activering door de endogene agonist, de chemokine ligand CXCL12 (voorheen bekend als stromale cel afgeleide factor 1α) SDF1-α)), en de potentiële inhibitie van deze CXCL12-geïnduceerde Ca2 + reactie door bepaalde verbindingen. In deze test, worden U87 menselijke glioblastoma cellen stabiel uiting van de menselijke CXCR4 receptor gebruikt. Deze cellen missen op hetzelfde moment, endogene uitdrukking van CXCR7, een verwante chemokine receptor die ook CXCL1215,16,17 bindt. CXCR7 is eerder ook aangetoond te kunnen vormen van de heterodimers met CXCR4, waardoor het moduleren van de signalering eigenschappen van deze laatste receptor18. Schommelingen in de hoeveelheid intracellulair Ca2 + gemedieerd door CXCR4 worden gecontroleerd door het laden van de CXCR4+ cellen met fluo-2 acetoxymethyl (AM) ester, een cel-permeabele hoge affiniteit fluorescerende Ca2 +-bindende kleurstof. Fluo-2 AM is een één golflengte fluorescerende molecule die kan worden enthousiast op 490 nm, terwijl de fluorescentie van de emissie is gemeten op 520 nm. Deze emissie-fluorescentie verhoogt op Ca2 + binding, met een groot dynamisch bereik tussen de Ca2 +-gebonden en ongebonden staat. De toename van fluorescent signaal is van voorbijgaande aard, die zich voordoen binnen een tijdspanne van een paar minuten, en daarna zal vergaan. De piekhoogte van de verdere correlaten van fluorescerende emissie met het niveau van de receptor activering. De test zelf wordt uitgevoerd met behulp van een fluorescentie microplate lezer uitgerust met een CCD geïntensiveerd (ICCD) camera die beschikt over een geïntegreerde pipetting systeem waarmee de standaardisatie van de pipetting stappen in de analyse (Zie Tabel van materialen) . Bovendien, is de gelijktijdige meting van het fluorescerende signaal in ieder putje van een microplate een ander belangrijk voordeel van de lezer van de fluorescentie die wordt gebruikt. Tijdens het eerste deel van de bepaling die de verbindingen onderzochte (bijvoorbeeld, een panel van kleine moleculen met een vaste concentratie of in een verdunningsreeks) worden toegevoegd aan de fluo-2 AM geladen CXCR4+ U87 cellen gevolgd door een ~ 10 min incubatietijd tijdens welke de mogelijke agonistic effect van de verbindingen wordt continu gemeten in real-time. Vervolgens de endogene agonist (d.w.z.CXCL12) wordt toegevoegd aan de cellen om te roepen een CXCR4-gemedieerde voorbijgaande verhoging van het niveau van intracellulaire Ca2 +. Tijdens dit deel van de bepaling kan de potentiële antagonistische activiteit van de geteste stoffen worden geëvalueerd. Hoewel deze Ca2 + mobilisatie test is vooral gebruikt voor het identificeren en bepalen de remmende potentie van concurrerende CXCR4 antagonisten (d.w.z., verbindingen, waardoor de endogene agonist om te binden en stimuleren de receptor), het ook receptor agonisten kan identificeren en, bovendien, verbindingen die oefenen hun functie door inbinden op allosteric sites (dat wil zeggen, websites die topografisch van de orthosteric binding site bezet door de endogene agonist afwijken). De laatstgenoemde categorie van verbindingen en voorbeelden van allosteric agonisten PAMs en NAMs19,20. Overwegende dat specifieke receptor antagonisten evenals NAMs de CXCL12-geïnduceerde Ca2 + reactie remmen zou, PAMs dit antwoord zou verbeteren (Zie ook bespreking sectie). Hoewel de bepaling hierin specifiek CXCR4 doelen beschreven, wordt verwacht dat deze methode kan worden toegepast op andere GPCRs met minimale optimalisatie inspanning, tenminste als ze het signaal via de vrijlating van intracellulaire Ca2 +.

G eiwit-gekoppelde receptoren (GPCRs) zijn van groot belang voor de farmaceutische industrie, omdat ze betrokken bij vele ziekten bij de mens zijn en goed gevalideerde streefniveaus voor therapeutische interventie. Ontdekking van loodverbindingen, met inbegrip van kleine synthetische moleculen, die een specifiek remmende werking van de receptor functie, is een belangrijke eerste stap in de Geneesmiddelenontwikkeling en is afhankelijk van robuuste, gevoelige en specifieke cel-gebaseerde testen. Hier beschrijven we een kinetische cellulaire assay met een fluorescerende uitlezing hoofdzakelijk ontworpen om te identificeren specifieke receptor antagonisten die een remmende werking van de intracellulaire Ca2 + release opgeroepen bij de activering van de CXC chemokine receptor 4 (CXCR4) door haar endogene ligand, de CXC chemokine ligand 12 (CXCL12). Een belangrijk voordeel van deze methode is dat ook hierdoor screening van verbindingen begiftigd met intrinsieke agonistic eigenschappen (d.w.z., verbindingen die aanleiding kunnen geven tot een toename van de intracellulaire Ca2 + concentratie in de afwezigheid van CXCL12) of verbindingen moduleren van de receptor functie via interactie met allosteric bandplaatsen (d.w.z., positieve en negatieve allosteric modulatoren (PAMs en NAMs, respectievelijk)). Op de keerzijde, kunnen autofluorescent verbindingen interfereren met de bepaling van uitlezing, waardoor het belemmeren van betrouwbare data interpretatie. The overall goal of this cell-based assay is to identify and characterize molecules capable of stimulating or inhibiting G protein-coupled receptor-mediated intracellular calcium mobilization. This method can be a valuable assay for G protein-coupled receptor drug discovery because it can be used to screen for molecules that modify receptor-mediated intracellular calcium bodies. The main advantage of this technique is that it allows molecules with agonistic or antagonistic properties to be identified within the same assay.This assay may help in the discovery of novel drug candidates that act on G protein-coupled receptors promising therapeutic targets that play a role in many human diseases. On the day of cell passaging, coat each well of a black walled polystyrene 96-well plate with clear bottoms with 100 microliters of 0.1%gelatin solution and incubate the plates for two hours at room temperature. While the gelatin is solidifying, use a trypsin EDTA solution to lift the cells of interest from the bottom of their culture flask, resuspending the cells in fresh growth medium in a 50 milliliter conical tube once they have detached.After counting, collect the cells by centrifugation and resuspend the pellet in fresh growth medium to a one times 10 to the fifth viable cells per milliliter concentration. When the plates are ready, discard any excess gelatin solution and dry the plates on a lab tissue. Then wash the wells with 200 microliters of PBS and replace the wash with 200 microliters of cells per well for an overnight incubation at 37 degrees Celsius and 5%carbon dioxide.The next morning, tap the plate to discard the growth medium from the wells and dry the plate upside on a paper towel. Then label the cells in each well with 100 microliters of loading dye solution per well and incubate the plate for 45 minutes in the dark at room temperature. While the cells are incubating, add 75 microliters of CXCL12 per well to the chemokine plate and 50 microliters of the compound of interest to each well of the compound plate.Next, turn on the cooler unit of the fluorescence plate reader followed by the plate reader itself. After allowing the system a few minutes to initiate, open the plate reader software. To set up the assay protocol, under the settings tab select read mode and the appropriate excitation and emission wavelengths for the loading dye.Select the mix with transfer fluid tab for automatic mixing of the compounds in the compound plate. In the transfer fluid tab, set 20 microliters of each well from the compound plate to be transferred into the measurement plate and position the pipette tips at 20 microliters below the liquid surface. Set the speed of aspiration to 50 microliters per second and the speed of dispensing to 25 microliters per second and click read with transfer fluid.During the first interval, select 60 reads with a read interval time of one second with 10 reads to be recorded before the compounds are dispensed into the measurement plate and 50 reads to be completed after. Set the second interval with a read interval time of 30 seconds and 18 reads. Select three tip washes for the protocol, setting each wash step to include the appropriate wash fluid, one wash cycle, a fast pump speed, and five strokes per wash.Use the pause pipettor function to include the pipettor pause of 300 seconds and use the mix with transfer fluid setting to allow automatic mixing of the CXCL12 solution within the chemokine plate. In the subsequent transfer fluid tab, set 25 microliters from each well of the chemokine plate to be transferred to the measurement plate and position the tips 20 microliters below the liquid surface. Set the aspiration speed to 50 microliters per second and the dispensing speed to 25 microliters per second.Use the read with transfer fluid button to set the first interval to include 145 reads with a read interval time of one second. Set the second interval to a read interval time of six seconds and 20 reads. Select three more wash tip cycles.Then click set stage temperature and set the device temperature to 37 degrees Celsius. At the end of the loading dye incubation, discard the buffer from the measurement plate and dry the plate on a lab tissue. Wash the cells with 150 microliters per well of assay buffer replacing the buffer with 80 microliters of assay buffer after two minutes.Place the measurement plate in the read position, the compound plate at the source two position, the chemokine plate at the source three position, and a box of black tips at the source one position. Close the device and click protocol signal test to determine the background relative light units and fluorescence variance over the plate. Select test signal in the popup window.If background values of 8, 000 to 10, 000 relative light units are obtained, select update and click save to save the main protocol. Then push run to start the assay. In this graph, a dose-dependent increase in fluorescence correlating with the dose-dependent increase in the release of calcium can be observed.Increasing levels of CXCL12-induced fluorescence allow the generation of a dose response curve based on the difference between the minimum and maximum response over baseline between the first measurement after addition addition of the chemokine and the final measurement in the protocol. No fluorescent calcium-related response was evoked by CXCL12 even at a high concentration when cells lacking the corresponding functional chemokine receptor CXCR4 were used demonstrating the receptor specificity of the CXCL12-evoked measurement. A key application for this cell-based assay is to identify small molecules that, following pre-incubation, dose dependently inhibit CXCL12-induced calcium mobilization in the cells as compared to control non-active compounds that do not inhibit the response.To determine the inhibitory potency of small molecules, a dose response curve can be generated for the inhibitory molecules using nonlinear regression. This assay can be used to study other G protein-coupled receptors with no or only minimal further optimization as well as in this experiment in which a dose-dependent increase in fluorescence was observed when another chemokine receptor, CCR5, was stimulated with its natural chemokine agonist CCL5. Once mastered, this technique can be used to study many other G-protein coupled receptors that signal via the mobilization of intracellular calcium ions upon receptor activation.After watching this video, you should have a good understanding of how this cellular assay can be used to identify agonists and antagonists of the CXCL4 chemokine receptor.
