RCPG est une super-famille important de protéines de surface de cellules qui activent les cascades de transduction de signal lors de la liaison du ligand extracellulaire. Ils peuvent être activés par une grande variété de stimuli, y compris les peptides, hormones protéiques, les amines biogènes et lipides, qui se traduit par l’ouverture de diverses voies de signalisation intracellulaire et réponses biologiques éventuellement1,2 . En outre, RCPG est impliqués dans un grand nombre, si pas tous, les processus physiologiques et du développement et plusieurs maladies humaines sont associées à dysfonctionnelle surexpression de signalisation ou récepteur de RCPG. RCPG est donc parmi les cibles pharmacologiques plus validés en médecine1,3.
Typiquement, un flux de travail découverte de drogue GPCR commence avec des essais de criblage cellulaire permettant l’identification de composés tels que de petites molécules, des anticorps monoclonaux et des peptides qui peuvent moduler l’activité de l’un RCPG particulière. Dans la découverte de médicaments GPCR beaucoup de différents types de tests existe pour rechercher ces composés, dont la plupart est compatible avec les campagnes de milieu - de criblage à haut débit. Les analyses les plus utilisés sont des expériences de liaison du récepteur, fluorescence ou luminescence base de tests de détection des fluctuations du niveau de ce que l'on appelle messagers secondaires (p. ex., Ca2 +, adénosine monophosphate cyclique (AMP)), phénotypiques tests de dépistage et β-arrestine recrutement assays4. Le choix d’un type particulier de test peut dépendre de plusieurs facteurs, mais est également déterminé par la connaissance préalable des propriétés de signalisation d’un RCPG donnée. Agoniste liant aux un RCPG induit un changement conformationnel catalysant l’échange de diphosphate de guanidine (PIB) pour triphosphate (GTP) de la guanidine sur la sous-unité α des protéines G hétérotrimériques. Par la suite, la sous-unité de GTP - Gαse dissocie de la sous-unitéβγ G et les deux sous-unités lancera également les voies de signalisation. Hydrolyse de la GTP-molécule et re-l’association subséquente du Gα- PIB et sous-unitésβγ G restaurera la protéine G dans sa conformation inactive5,6. Basé sur la similarité de séquence de la sous-unitéα G désignent les différents types de protéines G (G,s, Gje, Gq, G12/13)7. Signalisation via le Gα sous-unité donne lieu à plusieurs réponses typiques tels que l’augmentation (via Gs) ou diminuer (via Gj’ai) de la production d’AMP cyclique et des mobilisation intracellulaire de Ca2 + (via Gq)5 ,,7. Sous-unitésβγ G sont également capables d’induire des voies effecteurs intracellulaires. Par exemple, lors de l’activation de Gj’ai-GPCRs couplés, Gβγ peuvent stimuler directement la phospholipase C (PLC-β) pour produire le triphosphate d’inositol (IP-3) qui déclenche la libération de Ca2 + des réserves intracellulaires7 . Suite à l’activation des récepteurs, les RCPG est phosphorylés par kinases GPCR (krg) qui favorise l’interaction avec le β-arrestins. Ce processus termine G protéines de signalisation et mène à l’internalisation et finalement la désensibilisation du récepteur. Β-arrestins sont également capables de former des complexes moléculaires multiples qui peuvent déclencher des autres voies de signalisation indépendantes de protéine G8de signalisation.
Au sein de la sous-famille des récepteurs de chimiokines, le Gj’ai-couplé CXCR4 est un RCPG qui a soulevé beaucoup d’intérêt comme une cible prometteuse pour la découverte de médicaments. Compte tenu de son rôle établi comme un corécepteur majeur pour le virus d’immunodéficience humaine 1 (VIH-1) l’entrée virale et infection, composés ciblant CXCR4 étaient initialement développés comme de candidats médicaments anti-VIH9. Plus récemment, un nombre croissant de preuves a conduit à un rôle important pour CXCR4 dans la tumorigenèse et cancer du sein métastase, ce qui en fait une cible thérapeutique validée en oncologie ainsi10. CXCR4 est fortement exprimée dans plus d’une vingtaine de types de cancer chez l’homme et la survie des cellules tumorales contrôles, prolifération et migration ainsi que l’angiogenèse tumorale liés10. CXCR4 antagonistes des différentes classes chimiques ont été précédemment décrits11,12, mais seulement la petite molécule qu'amd3100 n’est actuellement homologué pour une utilisation en clinique comme agent de mobilisation des cellules souches utilisée pendant le traitement du lymphome et myélome patients13,14. Les essais cliniques sont en cours pour évaluer l’innocuité et l’efficacité de plusieurs autres antagonistes CXCR4 dans les maladies humaines différentes, mais avec un fort accent sur l’oncologie12. Étant donné les nombreuses applications potentielles pour CXCR4 antagonistes, la recherche de nouveaux composés avec des propriétés pharmacocinétiques améliorées, une biodisponibilité améliorée ou potentiellement moins d’effets secondaires est justifiée.
Dans les présentes, un cinétique dosage cellulaire à base de fluorescence est principalement utilisé pour dépister composés capables d’inhiber CXCR4 est décrite. La mesure fluorescente de cette méthode repose sur l’augmentation transitoire de la concentration2 + Ca intracellulaire évoquée lors de l’activation de CXCR4 par son agoniste endogène, le ligand de chimiokine CXCL12 (autrefois appelés cellules stromales dérivées (1α) facteur SDF1-α)) et l’inhibition éventuelle de cette réponse2 + Ca induite par CXCL12 de composés particuliers. Dans cet essai, U87 glioblastome humain cellules exprimant le récepteur CXCR4 humain de manière stable sont utilisés. Dans le même temps, ces cellules ne possèdent pas une expression endogène de CXCR7, un récepteur de chimiokine connexes qui lie également CXCL1215,16,17. CXCR7 a auparavant également avéré capable de former des hétérodimères avec CXCR4, modulant ainsi les propriétés de signalisation de ce récepteur ce dernier18. Fluctuations du niveau d’intracellulaire Ca2 + médiée par CXCR4 sont surveillées en chargeant le CXCR4 des cellules avec ester acétoxyméthyl fluo-2 (AM), une cellule perméable haute affinité fluorescent Ca2 ++ -lie de colorant. Fluo-2 AM est une molécule fluorescente de longueur d’onde unique qui peut être excitée à 490 nm tandis que sa fluorescence d’émission est mesurée à 520 nm. Cette fluorescence d’émission augmente à la fixation Ca2 + , avec une large gamme dynamique entre le Ca2 +-lié et non consolidé État. L’augmentation du signal fluorescent est transitoire, qui se produisent dans un intervalle de quelques minutes et décroîtra par la suite. La hauteur du pic des corrélats fluorescent d’émission supplémentaires au niveau de l’activation du récepteur. L’analyse elle-même est réalisée à l’aide d’un lecteur de microplaques de fluorescence équipé d’une caméra CCD intensifié (ICCD) qui possède un système de pipetage intégré qui permet la normalisation du pipetage lors de l’essai (voir Table des matières) . En outre, la mesure simultanée du signal fluorescent dans tous les puits d’une microplaque est un autre avantage majeur du lecteur de fluorescence qui est utilisé. Au cours de la première partie du dosage les composés incriminés (p. ex., un panel de petites molécules à une concentration fixe ou dans une série de dilutions) sont ajoutés à la fluo-2 AM chargé CXCR4+ U87 cellules suivie d’un ~ 10 min d’incubation au cours de laquelle l’effet agoniste potentiel des composés est continuellement mesurée en temps réel. Ensuite, l’agoniste endogène (c'est-à-direCXCL12) est ajouté aux cellules pour évoquer une augmentation transitoire induite par le CXCR4 au niveau intracellulaire Ca2 +. Durant cette partie de l’analyse de l’activité antagoniste potentielle des composés testés peut être évaluée. Un aperçu schématique du déroulement général de l’essai est présenté dans la Figure 1.
Bien que ce test Ca2 + mobilisation a d’abord été utilisé pour identifier et déterminer la puissance inhibitrice des antagonistes compétitifs de CXCR4 (c.-à-d., les composés qui empêchent l’agoniste endogène pour lier et stimuler le récepteur), elle aussi peuvent identifier les agonistes des récepteurs et, en outre, les composés qui exercent leurs fonctions en se liant à des sites allostériques (c.-à-d., les sites qui diffèrent sur le plan topographique du site de liaison d’orthosteric occupé par l’agoniste endogène). Exemples de cette dernière catégorie de composés agonistes allostériques et PAMs et NAMs19,20. Considérant que les antagonistes des récepteurs spécifiques, mais aussi les NAMs inhiberait la réponse induite par CXCL12 Ca2 + , PAMs renforcerait cette réponse ( Voir aussi section). Bien que le test décrit ci-après spécifiquement des cibles CXCR4, on prévoit que cette méthode peut être appliquée aux autres RCPG avec un effort minimal d’optimisation, au moins si ils le signal par l’intermédiaire de la libération d’intracellulaire Ca2 +.

Les récepteurs couplés aux protéines G (RCPG) sont d’une grande importance pour l’industrie pharmaceutique car ils sont impliqués dans plusieurs maladies humaines et comprennent bien validées cibles d’intervention thérapeutique. Découverte de composés de plomb, y compris les petites molécules synthétiques, inhibent spécifiquement la fonction de receveur, est un premier pas important dans le développement de médicaments et s’appuie sur des analyses cellulaires sensibles, spécifiques et robustes. Nous décrivons ici une analyse cinétique cellulaire avec un afficheur fluorescent principalement destinée à identifier les antagonistes des récepteurs spécifiques qui inhibent la libération2 + Ca intracellulaire évoquée lors de l’activation du récepteur chimiokines CXC 4 (CXCR4) par ses ligand endogène, le ligand de chimiokines CXC 12 (CXCL12). Un avantage majeur de cette méthode est qu’elle permet aussi de dépister des composés dotés des propriétés agonistes intrinsèques (c'est-à-dire, composés provoquant une augmentation de Ca2 + concentration intracellulaire en l’absence de CXCL12) ou composés modulant la fonction du récepteur via l’interaction avec des sites de liaison allostérique (c.-à-d., des MODULATEURS ALLOSTÉRIQUES positifs et négatifs (PAMs et NAMs, respectivement)). Sur le côté, auto-fluorescente composés pourraient interférer avec une lecture de l’essai, entravant ainsi l’interprétation des données fiables. Probablement ce dosage peut être implémenté, avec des adaptations minimes, comme un test de découverte de médicaments génériques pour de nombreux autres RCPG dont l’activation entraîne une libération de Ca intracellulaire2 +.

The overall goal of this cell-based assay is to identify and characterize molecules capable of stimulating or inhibiting G protein-coupled receptor-mediated intracellular calcium mobilization. This method can be a valuable assay for G protein-coupled receptor drug discovery because it can be used to screen for molecules that modify receptor-mediated intracellular calcium bodies. The main advantage of this technique is that it allows molecules with agonistic or antagonistic properties to be identified within the same assay.This assay may help in the discovery of novel drug candidates that act on G protein-coupled receptors promising therapeutic targets that play a role in many human diseases. On the day of cell passaging, coat each well of a black walled polystyrene 96-well plate with clear bottoms with 100 microliters of 0.1%gelatin solution and incubate the plates for two hours at room temperature. While the gelatin is solidifying, use a trypsin EDTA solution to lift the cells of interest from the bottom of their culture flask, resuspending the cells in fresh growth medium in a 50 milliliter conical tube once they have detached.After counting, collect the cells by centrifugation and resuspend the pellet in fresh growth medium to a one times 10 to the fifth viable cells per milliliter concentration. When the plates are ready, discard any excess gelatin solution and dry the plates on a lab tissue. Then wash the wells with 200 microliters of PBS and replace the wash with 200 microliters of cells per well for an overnight incubation at 37 degrees Celsius and 5%carbon dioxide.The next morning, tap the plate to discard the growth medium from the wells and dry the plate upside on a paper towel. Then label the cells in each well with 100 microliters of loading dye solution per well and incubate the plate for 45 minutes in the dark at room temperature. While the cells are incubating, add 75 microliters of CXCL12 per well to the chemokine plate and 50 microliters of the compound of interest to each well of the compound plate.Next, turn on the cooler unit of the fluorescence plate reader followed by the plate reader itself. After allowing the system a few minutes to initiate, open the plate reader software. To set up the assay protocol, under the settings tab select read mode and the appropriate excitation and emission wavelengths for the loading dye.Select the mix with transfer fluid tab for automatic mixing of the compounds in the compound plate. In the transfer fluid tab, set 20 microliters of each well from the compound plate to be transferred into the measurement plate and position the pipette tips at 20 microliters below the liquid surface. Set the speed of aspiration to 50 microliters per second and the speed of dispensing to 25 microliters per second and click read with transfer fluid.During the first interval, select 60 reads with a read interval time of one second with 10 reads to be recorded before the compounds are dispensed into the measurement plate and 50 reads to be completed after. Set the second interval with a read interval time of 30 seconds and 18 reads. Select three tip washes for the protocol, setting each wash step to include the appropriate wash fluid, one wash cycle, a fast pump speed, and five strokes per wash.Use the pause pipettor function to include the pipettor pause of 300 seconds and use the mix with transfer fluid setting to allow automatic mixing of the CXCL12 solution within the chemokine plate. In the subsequent transfer fluid tab, set 25 microliters from each well of the chemokine plate to be transferred to the measurement plate and position the tips 20 microliters below the liquid surface. Set the aspiration speed to 50 microliters per second and the dispensing speed to 25 microliters per second.Use the read with transfer fluid button to set the first interval to include 145 reads with a read interval time of one second. Set the second interval to a read interval time of six seconds and 20 reads. Select three more wash tip cycles.Then click set stage temperature and set the device temperature to 37 degrees Celsius. At the end of the loading dye incubation, discard the buffer from the measurement plate and dry the plate on a lab tissue. Wash the cells with 150 microliters per well of assay buffer replacing the buffer with 80 microliters of assay buffer after two minutes.Place the measurement plate in the read position, the compound plate at the source two position, the chemokine plate at the source three position, and a box of black tips at the source one position. Close the device and click protocol signal test to determine the background relative light units and fluorescence variance over the plate. Select test signal in the popup window.If background values of 8, 000 to 10, 000 relative light units are obtained, select update and click save to save the main protocol. Then push run to start the assay. In this graph, a dose-dependent increase in fluorescence correlating with the dose-dependent increase in the release of calcium can be observed.Increasing levels of CXCL12-induced fluorescence allow the generation of a dose response curve based on the difference between the minimum and maximum response over baseline between the first measurement after addition addition of the chemokine and the final measurement in the protocol. No fluorescent calcium-related response was evoked by CXCL12 even at a high concentration when cells lacking the corresponding functional chemokine receptor CXCR4 were used demonstrating the receptor specificity of the CXCL12-evoked measurement. A key application for this cell-based assay is to identify small molecules that, following pre-incubation, dose dependently inhibit CXCL12-induced calcium mobilization in the cells as compared to control non-active compounds that do not inhibit the response.To determine the inhibitory potency of small molecules, a dose response curve can be generated for the inhibitory molecules using nonlinear regression. This assay can be used to study other G protein-coupled receptors with no or only minimal further optimization as well as in this experiment in which a dose-dependent increase in fluorescence was observed when another chemokine receptor, CCR5, was stimulated with its natural chemokine agonist CCL5. Once mastered, this technique can be used to study many other G-protein coupled receptors that signal via the mobilization of intracellular calcium ions upon receptor activation.After watching this video, you should have a good understanding of how this cellular assay can be used to identify agonists and antagonists of the CXCL4 chemokine receptor.
