Dit protocol beschrijft het volledige proces voor Europese aal rijping, handling en sperma cryopreservatie. De hier beschreven voorwaarden van veehouderij zijn optimaal voor snelle rijping en productie van grote volumes kwalitatief hoogwaardige sperma in dit soort 6,7,25. Het succes van dit protocol cryopreservatie en het mogelijke gebruik voor bevruchting na het ontdooien, afhangen sterk af van de kwaliteit van vers sperma <sup...

In de afgelopen 25 jaar, het aantal Europese aal (Anguilla anguilla) aankomen op de Europese kust met 90% 1,2,3gestaag gedaald. Er zijn verschillende factoren die deze drastische daling verklaren, met inbegrip van vervuiling, infecties, vernietiging van de habitat en overbevissing. Dit alles heeft een diepgaand effect op deze soort, wat leidt tot de opneming van de Europese aal op de Internationale Unie voor behoud van Nature (IUCN) lijst als een kritisch bedreigde 4. De ontwikkeling van technieken en protocollen voor de voortplanting in gevangenschap zijn dus noodzakelijk.
De rijping van de Europese aal in gevangenschap wordt bereikt door hormonale behandeling 5,6,7 , maar de productie van de gameten bij beide geslachten is moeilijk om te synchroniseren van 8. Hoewel de ontwikkeling van nieuwe androgeen implantaten is gebleken voor het versnellen van de oögenese in aal 9,is10, de timing van definitieve rijping in vrouwtjes nog steeds zeer variabele en moeilijk te controleren van 11. Daarom op korte termijn opslag van sperma 12,13,14 en cryopreservatie technieken zijn nodig voor het beheer van de reproductie, gameet synchronisatie geen onnodige 8maken.
Cryopreservatie van Europese aal sperma heeft ontwikkeld sinds 2003 15,16. Verschillende onderzoekers ontworpen succesvolle protocollen met dimethylsulfoxide (DMSO) of methanol als cryoprotectant 16,17,18,19,20. Hoewel beide protocollen met succes hebben ingezet, is de levensvatbaarheid van de verkregen cellen van gedooid sperma cryopreserved met DMSO lager dan met methanol 20,21. Bovendien paling zaadcellen in contact komt met DMSO is geactiveerd en vereist meer vervelend sperma manipulatie 19, methanol is daarom een geschikter cryoprotectant voor Europese aal sperma dan DMSO.
Hier, zal het protocol voor optimale behandeling en hormonale behandeling van de Europese aal hieronder worden beschreven. Naast dit, zal de beste Europese aal sperma cryopreservatie protocol gebruik van methanol als een cryoprotectant en een protocol voor de beoordeling van de kwaliteit van het sperma in deze soort ook worden beschreven.

handling and treatment of male european eels (anguilla anguilla) for hormonal maturation and sperm cryopreservation

De laatste jaren hebben verschillende onderzoeksgroepen gewerkt aan de ontwikkeling en verbetering van nieuwe protocollen voor het hanteren van de Europese aal en rijping. Als van nog, wekelijkse injecties van humaan choriongonadotrofine (hCG) gebleken rijp mannetjes na slechts 5-6 weken van de behandeling, produceren grote hoeveelheden hoogwaardige sperma gedurende enkele weken. Daarnaast zijn sperma cryopreservatie protocollen met verschillende extenders, cryoprotectant koeling en keer ontdooien eerder beschreven voor Europese aal. Hier, laten we dat Tanaka´s extender oplossing direct gebruikt worden kan voor bevruchting of voor cryopreservatie, waardoor het gebruik van verschillende soorten oplossingen en verdunningen niet nodig. Bovendien, het gebruik van methanol als een cryoprotectant maakt dit protocol makkelijk te gebruiken zoals methanol lage toxiciteit heeft en het sperma niet wordt geactiveerd. Het sperma niet hoeft te worden cryopreserved onmiddellijk na toevoeging van de cryoprotectant, en kan het lang na wordt ontdooid worden gebruikt. Bovendien is de beweeglijkheid van de zaadcellen na het ontdooien, hoewel het is lager dan dat van vers sperma nog steeds hoog. Het doel van dit werk is om te laten zien van de beste beschikbare protocol voor Europese aal behandeling, rijping en sperma cryopreservatie.

Sperma van 18 paling met een beweeglijkheid van de zaadcellen van 70% of hoger, werd geselecteerd voor deze studie. De resultaten toonden een vermindering in alle kwaliteitsparameters na ontdooien met die van verse zaadcellen (tabel 1 en Figuur 2 vergeleken).
