De gepresenteerde gedetailleerde protocol mogelijk maakt betrouwbare inrichting van muis primaire RPE culturen die confluentie bereiken na 1 week en presenteren van de belangrijkste kenmerken van de RPE zoals zeshoekige vorm en pigmentatie na 2 weken. De verkregen RPE cellen kunnen worden gebruikt voor een aantal downstream toepassingen zoals vitamine A metabolisme9, fagocytose van de fotoreceptor buitenste segmenten10 en ion vervoer11, epitheliale c...

De retinale pigment epitheel (RPE) is een enkellaags gepolariseerde epitheliale cellen, die zich bevindt tussen het neurale netvlies en het vaatvlies van het oog. De functionaliteit van de RPE cellen en de integriteit van de RPE enkelgelaagde zijn cruciaal voor de visie, omdat de RPE speelt een belangrijke rol in meerdere processen zoals het behoud van de buitenste bloed-retinale barrière, vervoer van water en ionen tussen het netvlies en de choroideus, lichte absorptie, bescherming tegen oxidatieve stress, controle van retinoid metabolisme en fagocytose van de buitenste segmenten van de researchdieren1,2. De locatie van de RPE aan de achterkant van het oog, evenals haar barrièrefunctie voorkomen van drugs systemisch beheerd vanuit doorgeven van het bloed naar het glasvocht humor, maken het moeilijk om te bestuderen van de complexiteit van de RPE functie in vivo. Dus, is er een grote behoefte aan de oprichting van RPE celculturen te bestuderen van de RPE cellen in een flexibele, gecontroleerde omgeving3,4.
Een aantal gevestigde RPE cellijnen bestaat, bieden een eenvoudige en handige manier van verkrijgen en opslaan van de cellen; gepasseerd cellen hebben echter enkele nadelen ten opzichte van primaire cellen2,3,4. Ten eerste, worden zij vaak gekenmerkt door veranderingen in de morfologie van de cel. Bijvoorbeeld, werden geen van de bestaande cellijnen gevonden te zijn geschikt voor een betrouwbare studie van de RPE barrière eigenschappen als gevolg van het verlies van cel polariteit fenotype en gedeeltelijke verdwijning van strakke kruispunten4. Naast het verlies van de polariteit en de juiste cel-naar-cel verbindingen verliezen de cellijnen RPE snel hun pigmentatie als gevolg van het ontbreken van de enzymen van de belangrijkste melanogenesis in de volwassen RPE5. De pigmentatie kan worden hersteld, maar de uitgebreide analyse van het mechanisme van nieuwe pigmentatie die zou bestaan uit een combinatie van transmissie-elektronenmicroscopie, gen expressie analyse en chemische tests ter bevestiging van de aanwezigheid van melanine heeft nooit zijn uitgevoerd6. Een meer beperking is dat de cellijnen RPE uitgebreide cel leven (soms - onsterfelijkheid hebben) en onder bepaalde voorwaarden kunnen transformeren in een zichzelf vernieuwende multipotente stamcellen die loskoppelen van het substraat en het formulier drijvende kolonies7, 8. deze beperking maakt het onmogelijk om het gebruik van de cellijnen voor transplantatie3 experimenten.
Gezien de nadelen van de gevestigde cellijnen van RPE, primaire RPE celculturen verkregen uit verse weefsels kunnen dienen als een meer biologisch relevante model te bestuderen van de RPE. Primaire RPE cellen zijn gebruikt niet alleen studeren RPE-specifieke functies zoals vitamine A metabolisme9, fagocytose van de fotoreceptor buitenste segmenten10 en ion transport11, maar ook om te studeren fundamentele celbiologie zoals epithelial cel polariteit2 lysosomale homeostase en autophagy12,13.
In de laatste paar jaar zijn er een aantal publicaties over de oprichting van primaire RPE culturen, die aangeeft een groeiende belangstelling op dit gebied onderzoek3,14,15. Talrijke protocollen voor menselijke RPE cellen en niet-menselijke RPE cellen zoals runderen en varkens RPE cellen werden gepubliceerd16,17,18,19. Het is echter moeilijker te behandelen muis RPE cellen vanwege het veel kleinere formaat. Hoewel er nogal een aantal publicaties beschreven protocollen om te isoleren RPE cellen uit de muis14,20,21, zijn er nog vele onderzoekers worstelen om te isoleren RPE cellen zonder verontreiniging van choroideus cellen of van cellen uit neurale netvlies puin. Hier presenteren we het protocol voor de vaststelling van de primaire muisknop RPE celcultuur, waaronder het verkrijgen van de ogen van de muis, de dissectie van de ogen en isolatie van de RPE bladen opleveren van de cellen voor het kweken. Dit video protocol zou vooral nuttig zijn voor onderzoekers die beginnen te werken met de muis primaire RPE culturen en begeleiding op de dissectie technieken nodig hebben.

primary cell cultures from the mouse retinal pigment epithelium

De retinale pigment epitheel (RPE) is een sterk gepolariseerde multifunctionele epitheel, die is gelegen tussen het neurale netvlies en het vaatvlies van het oog. Het is een vel van gepigmenteerde cellen die hexagonally zijn verpakt en verbonden door strakke kruispunten. De belangrijkste functies van de RPE omvatten absorptie van licht fagocytose van de schuur fotoreceptor buitenste segmenten, ruimtelijke bufferen van ionen, vervoer van voedingsstoffen, ionen en water evenals actieve betrokkenheid in de visuele cyclus. Met deze belangrijke en diverse functies is het uitermate belangrijk om te bestuderen van de biologie van RPE cellen. Een aantal RPE cellijnen zijn vastgesteld; gepasseerd en vereeuwigd cellen zijn echter bekend om snel verliest een deel van de morfologische en fysiologische kenmerken van natuurlijke RPE cellen. Primaire cellen zijn dus meer geschikt voor de studie van de verschillende aspecten van de biologie van de cel van de RPE en functie. Muis primaire RPE celkweek is erg handig voor onderzoekers sinds Muismodellen worden veel gebruikt in biologische studies, maar verzamelen RPE cellen van muis is ook zeer uitdagend vanwege hun kleine omvang. Hier presenteren we een protocol voor de vaststelling van de primaire muisknop RPE celculturen, waaronder enucleation en dissectie van de ogen en isolatie van de RPE bladen opleveren van de cellen voor het kweken. Deze methode kan efficiënt cel worden hersteld. De cellen van de RPE twee muizen verkregen kunnen confluentie op een 12 mm polyester membraan invoegen voorgeladen in cultuur plaat na een week van de cultuur en display enkele van de originele eigenschappen van bonafide RPE cellen zoals zeshoekige vorm en pigmentatie na twee bereiken weken van cultuur.

De muis primaire RPE cultuur van 2 muizen in 12 mm polyester membraan invoegen bereikt 90-95% confluentie opgericht na 1 week in cultuur. Na 2 weken van de cultuur, de cellen bereikt 100% confluentie en begon te vormen een mozaïek van cellen zeshoekige, gepigmenteerde en bi-nucleated. 3 weken van de cultuur, de cellen blijven vormen van de vorm en de pigmentatie, echter kreeg na 4 weken een deel van de cellen hyperpigmented (Figuur 2).
