AMs zijn lang-leven Long-ingezeten macrofagen die bevolken de longen beginnen bij de geboorte en blijvende over de gehele levensduur26. Hun rol in de pulmonaire fysiologie7 en pathologie12 en hun potentieel voor het voorspellen van pulmonaire autoimmuniteit24 zijn bekroond. Omdat AMs een langdurige aanwezigheid in longen11,27 hebben en omdat zij betrokken bij de active...

De Long-communicatie is een uniek complex ecosysteem met een uitgebreide lucht conduit en therapieën. De ingeademde lucht reist via de luchtpijp en vele vertakkingen van de bronchiën en bronchioli alvorens de longblaasjes, waar het bloed-lucht gasuitwisseling plaatsvindt. Als gevolg van directe interactie met de atmosfeer vereist het respiratoire oppervlak bescherming tegen de potentieel schadelijke effecten van deeltjes in de lucht en verontreinigende stoffen. Een aantal fysieke, chemische en immunologische belemmeringen beschermen de longen. Implementatie van fagocyten aan de luchtwegen oppervlakte dient met name een belangrijke eerste regel verdedigingssysteem. Alveolaire macrofagen (AMs) zijn een soort Long-ingezeten fagocyten, en ze vormen de overgrote meerderheid van de longkanker macrofaag zwembad. Zoals hun naam al doet vermoeden, AMs zijn voornamelijk gelokaliseerd op de alveolaire lumen en optreden als sessiele cellen die voortdurend de omgevingsklimaat proeven en communiceren met de alveolaire epitheel1. In stationaire toestand longen zijn meer dan 95% van de fagocyten in de alveolaire ruimte AMs2, waarvan de samenstelling als gevolg van ontsteking, infectie of chronische blootstelling aan verontreinigende stoffen wijzigen kan.
AMs deelnemen aan een breed scala van functies die mogelijk lokale naar de longen en/of systemische belang. Bijvoorbeeld, zijn AMs van essentieel belang in de ontwikkeling en de optimale werking van de longen; immuun toezicht; en de ontmijning van cellulaire puin, invasie van ziekteverwekkers, en geïnhaleerde deeltjes3,4,5,6,7. Gerichte uitputting van AMs heet schade kunnen toebrengen aan de goedkeuring van de respiratoire virussen en bacteriën4,8. Naast hun rol als fagocyten en een verdedigers van de eerste regel van de pulmonaire homeostase, AMs zijn gekend om te functioneren als antigeen-presenteren cellen in het opwekken van T cel immuniteit9, potentiating van de werkzaamheid van intranasale vaccin10 en Long-beperkte autoimmuniteit na Long transplantatie11,12te beïnvloeden. Tekort aan AM functie is gekoppeld aan de pulmonaire alveolaire proteinosis (PAP), een aandoening als gevolg van een genetische mutatie, maligniteit of infectie die Goedkeuringvande pulmonaire oppervlakteactieve stoffen13,14 schaadt. Transplantatie van AMs is nu onderzocht als een therapeutische benadering voor de behandeling van PAP 15,16.
AMs zijn bekend ontstaan tijdens de embryogenese en te volharden in de longen gedurende het hele leven zonder verdrongen door het circuleren van leukocyten2,17. Hoewel, AM omzet is niet detecteerbaar in homeostatische longen, zijn verschillende niveaus van AM omzet gemeld in bepaalde klinische voorwaarden met inbegrip van infectie door het influenza virus4, myeloablative bestraling18, blootstelling aan endotoxinen 19en20van de ouderdom. AMs worden verondersteld zelf vernieuwen via een low-grade proliferatie17,21, maar sommige recente studies beweren dat monocyten aanleiding kunnen geven tot een bevolking intravasculaire Long macrofagen22,23 onder experimentele omstandigheden, maar de functionaliteit van deze nieuwe geconverteerde pulmonaire macrofagen moeten nog worden omschreven in longziekten. Bovendien begrijpen de drempel van stimulus in het kader van de activering van de AM is een potentieel interessant gebied, zoals de Long probeert te behouden een evenwicht tussen de inflammatoire signalen en de immunoregulerende machines.
De fysiologische of pathologische veranderingen die tot verlies van immuunregulatie leiden zijn belangrijk om te evalueren in verschillende klinische settings (bijvoorbeeld infecties van de luchtwegen, inflammatoire longziekte en fibrotische longziekte). AMs zijn echter steeds meer erkend als indicatoren of zelfs determinanten van de gezondheid van de pulmonaire11,24. Momenteel zijn er geen uniforme protocollen beschikbaar voor oogsten, karakteriseren, en/of onderhouden AMs van mens en preklinische lymfkliertest modellen. Ontbreken van een consensus op AM precursoren en fenotypes en het ontbreken van een gedetailleerde methodologie hadden de grote wegversperring in ontcijferen rol(len) van AM bij pulmonaire gezondheid en ziekte. Het volgende protocol biedt een definitieve identificatie, isolatie en in vitro cultuur strategie die sterk zal verder van het begrijpen van AM gedrag en AM-gerichte diagnostische en therapeutische studies te vergemakkelijken.

Alveolaire macrofagen zijn terminaal gedifferentieerde, Long-inwoner van macrofagen van prenatale oorsprong. Alveolaire macrofagen zijn uniek in hun lange levensduur en hun belangrijke rol in de ontwikkeling van de Long en functie, evenals hun Long-gelokaliseerde reacties op infecties en ontstekingen. Tot op heden bestaat er geen uniforme methode voor de identificatie, isolatie, en behandeling van de alveolaire macrofagen van mensen en muizen. Deze methode is nodig voor studies over deze belangrijke aangeboren immuun cellen in verschillende experimentele instellingen. De methode die hier beschreven, die kan gemakkelijk door elk laboratorium worden aangenomen, is een vereenvoudigde aanpak voor het oogsten van de alveolaire macrofagen uit bronchoalveolar lavage vloeistof of longweefsel en onderhouden hen in vitro. Omdat alveolaire macrofagen voornamelijk als Adherente cellen in de longblaasjes optreden, is de focus van deze methode op hen verjagend voorafgaand aan de oogst en identificatie. De Long is een zeer gevacuoliseerd orgaan, en verschillende celtypes van myeloïde of lymfoïde oorsprong bewonen, interactie en worden beïnvloed door de communicatie van de longen. Met behulp van de set van oppervlakte markers beschreven hier, kunnen onderzoekers gemakkelijk ondubbelzinnig alveolaire macrofagen onderscheiden van andere leukocyten en zuiveren ze voor downstream toepassingen. De methode van de cultuur ontwikkeld hierin ondersteunt beide mens muis alveolaire macrofagen in vitro groeimogelijkheden en is compatibel met cellulaire en moleculaire studies.

De stroom cytometrische aanpak om te identificeren muis AMs is afgebeeld in Figuur 1. Dit omvat analyse van een minimumpakket van oppervlakte markers in AMs onderscheiden van andere Long-ingezeten of Long-infiltreren fagocyten noodzakelijk.
