En el siguiente protocolo, describimos un método conveniente para visualizar la segregación cromosómica y la progresión mitótica durante el ciclo celular en fibroblastos embrionarios de ratón usando histona 2B-GFP, etiquetado BacMam 2.0 y proyección de imagen de células vivas.
Puestos de control de ciclo celular monitor segregación cromosómica y juegan un papel importante en el mantenimiento de la integridad genética de la célula 1,2,3. Acumulación de los cromosomas segregados mal puede llevar a aneuploidía, que es una característica de los tumores sólidos más 4. Por lo tanto, la detección de los cromosomas del revestimiento puede utilizarse como un método para el estudio de inestabilidad cromosómica.
Etiquetado fluorescente proteínas pueden ser utilizado para visualizar segregación cromosómica vivo y cromosomas rezagados pero la generación de etiquetas mCherry o proteína H2B-GFP etiquetado requiere conocimiento substancial del gene entrega y biología molecular 5. Aquí describimos el uso del reactivo de CellLight Histone2B-GFP BacMam 2.0, regente de CL-HB, por simplicidad en lo sucesivo. Este reactivo puede utilizarse inmediatamente y elimina así las preocupaciones sobre la integridad y calidad de vector. Además, este reactivo no requiere el uso de tratamientos potencialmente dañinos o lípidos y productos químicos del colorante de carga. A diferencia de las etiquetas fluorescentes convencionales, el regente de la CL-HB manchas independientemente de la función (es decir., potencial de membrana). El regente CL-HB puede ser simplemente añadido a las células y se incubó toda la noche para la expresión de la proteína. El regente de la CL-HB no se replica en células de mamíferos y puede ser utilizado en la configuración de nivel de bioseguridad (BSL) 1 laboratorio. También, se puede detectar esta transfección transitoria después de la incubación durante la noche durante 5 días, tiempo suficiente para llevar a cabo análisis celulares más dinámico.
Por otra parte, anomalías cromosómicas podrían ser estudiados por varias técnicas como la citometría de flujo, inmunohistoquímica o fluorescencia in situ hibridación (pescado) 6. Citometría de flujo puede utilizarse para el estudio de la aneuploidía, que puede ser medido basado en contenido de ADN y la fase de células en ciclo celular. Aunque citometría de flujo puede utilizarse para medir aneuploidía, no proporciona información sobre segregación de errores cromosómica. PESCADO con técnicas de inmunohistoquímica utilizan sondas fluorescentes para obligar a ADN o los cromosomas. Mientras que estas técnicas proporcionan una instantánea de la situación de una población de células, no permiten el vivo de la célula de tal modo que falta cualquier información obtenida a través de la visualización espaciotemporal de citocinesis en células específicas durante un período de tiempo.
Este protocolo fue utilizado para estudiar los cromosomas de revestimiento o segregación errónea cromosómica en fibroblastos embrionarios de nocodazole tratado ratón (MEFs) aislado de PP2A-B56γ-ratones. Además de la anterior aplicación, este protocolo proporciona una herramienta sencilla para marcar y visualizar la segregación cromosómica en diversos tipos celulares que pueden utilizarse para estudiar la regulación del ciclo celular o inestabilidad cromosómica en las células del tumor. Además, también puede utilizado para el estudio de inestabilidad cromosómica causada por varios tratamientos con fármacos o para estudiar los efectos del gen golpee hacia fuera dando por resultado la segregación cromosómica de mal.

Los cromosomas deben ser fiable y uniforme separados en células de la hija durante la división celular mitótica.
