Cellcykeln kontroll kontrollpunkter som säkerställer korrekt kromosomsegregering förhindra aneuploidi och cell omvandling 1,2,3. I föreliggande studie, fann vi att inaktivering av PP2A-B56γ resulterade i en försvagad spindel montering checkpoint. I följande protokoll beskriver vi en smidig metod för att visualisera kromosomala segregering och mitotiska progression under cellcykeln i mus embryonala fibroblaster använder Histon 2B-GFP, BacMam 2.0 märkning och levande cell imaging.
Cellcykeln kontroll kontrollpunkter övervaka kromosomsegregering och spelar en viktig roll i upprätthållandet av genetisk integritet av cell 1,2,3. Ansamling av mis segregerade kromosomer kan leda till aneuploidi, vilket är ett signum för mest solida tumörer 4. Påvisande av släpar kromosomer kan därför användas som en metod för att studera kromosominstabilitet.
Fluorescently märkt proteiner kan vara används för att visualisera levande kromosomsegregering och kromosomala släpar efter men generationen av mCherry-märkta eller H2B-GFP taggade protein kräver omfattande kunskap om gen leverans och molekylärbiologi 5. Här beskriver vi CellLight Histone2B-GFP BacMam 2.0 reagens, nedan kallad CL-HB regent, för enkelhetens skull. Denna reagens kan användas omedelbart och därmed eliminerar oro vektor kvalitet och integritet. Dessutom kräver denna reagens inte användning av potentiellt skadliga behandlingar eller lipider och dye-lastning kemikalier. Till skillnad från konventionella fluorescerande etiketter, CL-HB regent fläckar oberoende av funktion (dvs., membranpotential). CL-HB regent kan enkelt läggas till cellerna och inkuberas över natten i proteinuttryck. CL-HB regent replikerar inte i däggdjursceller och kan användas i biosäkerhet nivå (BSL) 1 laboratoriemiljö. Också, denna övergående transfection kan upptäckas efter natten inkubation för upp till 5 dagar, tillräckligt med tid för att genomföra mest dynamiska cellulära analyser.
Alternativt kunde kromosomavvikelser studeras av olika tekniker såsom flödescytometri, immunohistokemi eller fluorescens i situ hybridisering (FISH) 6. Flödescytometri kan användas för att studera aneuploidi, som kan mätas utifrån DNA-innehåll och fasen av celler i cellcykeln. Även om flödescytometri kan användas att mäta aneuploidi, ger den inte information om kromosomala felaktig segregation. FISK och immunohistokemi tekniker använda fluorescerande sonder för att binda till DNA eller kromosomer. Medan dessa tekniker ger en ögonblicksbild av status för en population av celler, tillåter de inte levande cell imaging därmed saknas någon information som erhållits genom spatiotemporal visualisering av cytokines i specifika celler följde över en tidsperiod.
Detta protokoll användes för att studera släpar kromosomer eller kromosomala mis segregation i nocodazole behandlade mus embryonala fibroblaster (MEFs) isolerade från PP2A-B56γ-möss. Utöver ovan tillämpning ger detta protokoll ett enkelt verktyg för att märka och visualisera kromosomala segregering i olika celltyper som kan användas för att studera cellcykeln förordning eller kromosomal instabilitet i tumörceller. Det kan dessutom också användas att studera kromosomal instabilitet som orsakas av olika läkemedelsbehandlingar eller studera effekterna av genen knocka resulterar i kromosomala mis segregation.

Kromosomer måste vara tillförlitligt och enhetligt uppdelade i dottercellerna under mitotiska celldelningen. Trohet av kromosomala segregation styrs av flera mekanismer som inkluderar den spindel montering Checkpoint (SAC). MEFs från vildtyp och PP2A-B56γ-möss var seedad i en 2-väl kammaren täckglaset och tillåtet att fästa. Dag 2, MEFs synkroniserades med 0,1% FBS för 24 h.
