Der er flere kritiske trin til vellykket venøs blodprop dannelse ved hjælp af stasis model. Induktion af venøs trombose er mere udfordrende i gamle mus på grund af ophobning af fedt omkring den ringere vena cava og aorta. Mus under denne procedure, bør ideelt set være 8 - 10 uger gamle. Venøs tromboemboli (VTE), som består af dyb venøs trombose (DVT) og lungeemboli, er den tredje hyppigste årsag til hjerte-kar-død efter myokardieinfarkt og slagtilfælde. Det er en almindelig tilstand, der påvirker 1-2% af befolkningen, med en årlig incidens på 1 i 5001. VTE kan føre til: 1) død gennem lungeemboli; 2) post-trombotiske syndrom, præget af kroniske smerter i benene, hævelse og sårdannelse; eller 3) kronisk pulmonal hypertension resulterer i betydelig kronisk respiratorisk kompromis. VTE er en multifaktorielle sygdomme og kan skyldes stasis af blodgennemstrømningen, skade på fartøjet vægge, og/eller hypercoagulable stater som følge af en forstyrrelse af balance mellem koagulation og de fibrinolytiske systemer, som det er blevet beskrevet over hundrede år siden af Virchow og er kendt som den Virchow treklang.
Fordi i de fleste tilfælde er det umuligt at få menneskelige DVT prøver, har forskere udviklet eksperimentelle dyremodeller for DVT. Flere dyr herunder rotte2, mus3, kanin4, gris5, hund6og primat7 har været brugt. Mus kan være genetisk modificerede og er den hyppigst anvendte dyr at studere DVT. Men som i alle dyr, spontane DVT er ikke observeret i mus. Således, fysiske eller kemiske ændringer af vene væg er brugt til at oprette trombose hos mus. Vi har tidligere brugt ferrichloridopløsning model til at fremkalde blodpropper i den ringere vena cava (IVC) mus8,9,10. Denne model har fordelen at producere pålideligt okklusiv trombi inden for få minutter og kan bruges til at undersøge rollen af antikoagulerende og anti-trombocyttal medicin under akut DVT. Det er imidlertid en terminal procedure. Således, for at studere akut og kronisk DVT, stasis model er mere velegnet. I denne model, er blodprop dannelse fremkaldt af den fuldstændige afbrydelse af blodgennemstrømningen i IVC, en af faktorer i Virchow's triad for udvikling af DVT. Denne model kan bruges til at studere DVT dannelse og opløsning, hvilket er en fordel i forhold til FeCl3 model11.
Vi har udviklet en ikke-invasiv metode til at følge blodprop dannelse og opløsning over tid ved hjælp af en mikro-imaging høj frekvens ultralyd system12. Vi har tidligere vist, at måling af venøs trombose af ultralyd korrelerer positivt med trombi fremkommer patologisk. I to efterfølgende undersøgelser har vi bekræftet, at målinger fremstillet med ultralyd korrelat med blodprop vægt og blodprop område kvantificeres ved histokemi9,10. Vigtigere er, har vi viste at høj frekvens ultralyd kan bruges til at overvåge dannelsen af dyb venøs trombose i mus12. Det kan også bruges til at kvantificere Trombe opløsning i en ikke-invasiv måde.
Her vil vi beskrive protokollen tillader blodprop dannelse ved hjælp af stasis-induceret trombose musen model og hvordan blodprop dannelse kan overvåges ikke-invasivt over tid ved hjælp af højfrekvente ultralyd.

Venøs trombose er en almindelig tilstand, der påvirker 1-2% af befolkningen, med en årlig incidens på 1 i 500. Venøs trombose kan føre til døden gennem lungeemboli eller resultater i den post trombotiske syndrom, præget af kroniske smerter i benene, hævelse og ulceration eller kronisk pulmonal hypertension resulterer i betydelig kronisk respiratorisk kompromis. Dette er den mest almindelige hjerte-kar-sygdom efter myokardieinfarkt og iskæmisk slagtilfælde og er en klinisk udfordring for alle medicinske discipliner, som det kan komplicere forløbet af andre lidelser som kræft, systemisk sygdom, operation og større traumer.
Eksperimentelle modeller er nødvendige for at undersøge disse mekanismer. Stasis model inducerer konsekvent Trombe størrelse og en kvantificerbar mængde af blodprop. Det er imidlertid nødvendigt at systematisk ligate sidegrene af den ringere vena cava at undgå variabilitet i Trombe størrelser og eventuelle fejlagtige data fortolkning. Vi har udviklet en ikke-invasiv teknik til at måle Trombe størrelse ved hjælp af ultralyd. Brug af denne teknik kan vi vurdere Trombe udvikling og opløsning over tid i de samme dyr. Denne tilgang begrænser antallet af mus kræves til kvantificering af venøs trombose i overensstemmelse med princippet om erstatning, reduktion og forfinelse af dyr i forskning. Vi har demonstreret, at blodprop vægt og histologiske analyse af Trombe størrelse korrelat med måling fremstillet med ultralyd. Den nuværende undersøgelse beskrives derfor, hvordan til at fremkalde dyb venetrombose i mus ved hjælp af den ringere vena cava stasis model og hvordan man kan overvåge det ved hjælp af højfrekvente ultralyd.
