Foto-attractie bioassays zijn een belangrijk instrument om te bepalen van de optimale aantrekkelijke kleur(en) en minimaliseren van de opties voor veldproeven van deze kleuren. Echter verschillende factoren moeten worden overwogen bij het optimaliseren van de bioassay voor een specifieke insecten met inbegrip van: enkel licht vs. concurrerende lichte experimenten, helderheid, optimale spectraal bereik, ambient licht interferentie, stand van de insecten en de natuurlijke gedragingen die kunnen beperken de mogelij...

Bijna alle entomologische toezicht is afhankelijk van olfaction of visuele lokstoffen en vaak beide. Vluchtige olfactorische lokstoffen kan verspreiden doorheen de omgeving wat resulteert in een groot aantrekkelijk gebied. Visuele lokstoffen kunnen echter een beperkter bereik hebben vanwege de ongewervelde compound eye oplossen afbeeldingen1,2,3. Visuele lokstoffen moet daarom worden geoptimaliseerd om het insect van belang om attractie en de val ontworpen om te profiteren van het doel insect natuurlijke gedragingen te maximaliseren.
Visuele attractie is gebaseerd op de golflengten van de zon of andere lichtbronnen die worden geabsorbeerd of gereflecteerd door de oppervlakte van een object; organismen bekijken deze absorptie/breking van golflengtes als kleur. Insect visie heeft gevonden om op te nemen van de blauwe, groene en ultraviolet (UV) golflengten1. Insecten gebruiken hun visie om hulp bij het vinden van stuurlieden, voedsel en onderdak4. Insecten kunnen visueel object maten, kleuren, vormen, bewegingen en contrasten5,6definiëren. Nocturnally actieve insecten zijn over het algemeen aangetrokken door licht van uiteenlopende contrast en intensiteit4, terwijl dagverloop insecten achtergrondkleuren en -afbeeldingen, naast contrast vanwege de grotere foton beschikbaarheid gedurende de dag oplossen kunnen. Toezicht vallen gebruiken het insect visuele aanwijzingen in hun voordeel te optimaliseren attractie en vangen van7.
De meest voorkomende methode voor de evaluatie van de foto-attractie was waarneming van insecten beweging naar verschillende gekleurde vormen zoals bloemen8 of objecten (zoals kleverige kaarten9,10). Visuele bioassays met behulp van gekoloniseerde insecten kunnen helpen identificeren van het optimale aantal golflengtes en/of intensiteit, waardoor het aantal veldproeven. Visuele bioassays zoals de "dubbelzijdig licht Tunnel" werden ontworpen voor het testen van vliegen11. Het probleem met twee zijdig lichte tunnels zijn dat ze niet verantwoordelijk zijn voor insecten die niet worden verzameld. De meeste insecten zal blijven steken op interne hoeken en langs de randen in arena's. Slechts twee kleuren kunnen ook in één keer worden getest. Andere testen zijn de methoden van Steverding & Troscianko (2004)12, waardoor werd teruggebracht van de tseetseevlieg aantrekkingskracht op brede banden (±50 nm) van lichte kleuren. Light emitting diodes (LED's) zijn opgenomen in overlappen naar insecten attractie te verbeteren door het optimaliseren van de golflengtes van de uitgezonden licht1,13,14. Optimaliseren van de visuele aantrekkingskracht van deze traps of toezicht apparaten zal efficiënter insect collectie met behulp van het insect aangeboren gedrag om insecten te lokken. Op deze manier worden bioassay resultaten gebruikt voor het optimaliseren van bestaande technologie van de overlapping. De "terrestrische Arthropod Trap" dat verbeterd de industrie standaard koepel-type val voor rode bloem kever surveillance (US patent # US8276314B2)) en de "methode en composities voor verbeterde licht Traps" die opgenomen van light emitting diodes in antenne insect traps (US patent # US2009/0025275A1). De twee octrooien gebruiken LED-technologie die is geoptimaliseerd door het gebruik van de resultaten van de bioassay tot een aanzienlijke verbetering insect vallen.
Deze studie beschrijft een foto attractie bioassay arena en methoden waarmee onderzoekers te evalueren van het insect antwoord als u wilt beperken golflengten als een concurrerende of één aantrekkelijke kleur. Apparatuur en experimentele wijzigingen worden uitgereikt aan nachtelijke dagverloop, terrestrische en luchtfoto insecten.

Geoptimaliseerde visuele lokstoffen zal insect overlapping efficiëntie verhogen met behulp van het doel insect aangeboren gedrag (positieve foto-taxi's) als een middel om het insect verleiden met een bevolking controle of toezicht val. Onderzoekers moeten vervolgens bevestigen de resultaten van de bioassay in het veld en de beperkte aantrekkelijke afstand van de visuele lokstoffen begrijpen.
De klaverblad bioassay-arena is een flexibele methode te beoordelen foto attractie terwijl ook beoordeling van een aantal natuurlijke insectenwerende gedrag zoals ontsnappen en voederen van reacties. De arena kan worden gebruikt voor terrestrische of luchtfoto insect experimenten, evenals dagelijkse en nachtelijke insecten. Data collectie technieken met de arena zijn video-opnamen, tellen van contact met de lichten, of fysiek het verzamelen van de insecten als ze aangetrokken naar de lichten worden. De assay rekeningen voor insecten die neen-keuze en de arena's maken kunnen enkele (concurrerend) kleur of meerdere (concurrerende) kleuren. This method can help answer key questions in insect surveillance and control, by improving the attraction of existing traps and incorporating colors or lights into the design of future monitoring and control traps. The main advantage of this technique is the initial evaluation of attractive colors and insect behaviors in the lab, prior to the costly and time consuming tests in the field. Insects have innate behaviors or instincts that should be used to make monitoring and trapping more effective.The bioassay arena is designed to be a flexible method to evaluate behaviors in any insect species, prior to field trials. This protocol uses an aerial arena, but a terrestrial arena design is also available. The critical difference between the aerial and terrestrial arena's is the use of vertical space, which changes the size of the arena.When selecting an arena type for an experiment consider the insect species being studied. The aerial arena has the ability to capture insects in a collection container, whereas the terrestrial arena is a two dimensional object and really is designed for recording visits to the lights. To begin, disassemble and clean the aerial arena with an odorless, non abrasive soap and warm water.Then dry the arena and allow it to finish air drying. Before each experiment record the pertinent environmental conditions, and monitor any trends that occur from experiment to experiment. First, use four supports and black linen cloth to set up a pedestal.Then place the arena baseplate on top of the pedestal and assemble each piece of the arena on the baseplate. Next, place the cloverleaf arena around the central release point in the baseplate. Then install the LEDs into the LED holders of the four collection containers, and set up the electrical equipment to control the lights.To prepare the arena for a competitive experiment, position LED lights with distinct characteristics on each cloverleaf. The arena set up is the most important and critical factor, because simple changes in the arena can alter the insect's behaviors. Therefore, many factors can be changed or altered to address insect behaviors, such as photo attraction.Behaviors with feeding, so blood fed, or sugar fed or starved. Different life history stages or different physiological responses. Next, to control for potential effect of light interference or environmental preferences, rotate the light positions after every replicate.Refer to the text protocol for further information on filming devices. First place the clear lid over the arena, parallel to the baseplate. Then introduce immobilized midges to the arena.After the experimental period record the pertinent environmental conditions. Finally, after euthanizing the midges count the responders in each collection container. In this study the movement of red flower beetles was evaluated in a competitive style terrestrial arena for five minutes, and filmed with an infrared camera.Test colors included blue, and three wavelengths of UV light. The trial data indicated that the beetles were most attracted to near UV light with a wavelength of 390 nanometers. The movement of Hessian flies was also evaluated in a competitive style aerial arena with a diurnal setting.The lights were most attracted to lights in green wavelengths with high intensities. Female flies preferred the green spectra of 502 and 525 nanometers. While both sexes preferred the high intensity 16 watts per square meter light.Biting midges were also tested in the aerial arena under nocturnal conditions, to determine optimal attractive colors. The highest proportions of midges were attracted to UV light, with the brightest lights being the most attractive. Once mastered, the photo attraction bioassay can be used to rapidly assess insect behavior, prior to costly and labor intensive field trials.
