Studerer de motoriske koordination er en nyttig tilgang i karakterisering af modeller for neurodegenerative sygdomme, især for sygdomme som PD i som motor koordinering er hårdt ramt. Med hjælp fra en kinematisk gangart analyse funktionel analyse, kan vi identificere subtile ændringer i gangart af dyr i starten af locomotion problemer eller i modeller med svag neurodegeneration og dermed relativt beskedne fænotype. Neurodegenerative sygdomme være en betydelig byrde for patienter, familier og samfund, og vil blive af endnu større bekymring som levealder stiger, og verdensbefolkning fortsat alder. En af de mest almindelige symptomer på neurodegenerative sygdomme er balance og mobilitet problemer. Således karakterisering af motor adfærd i aging pattedyr (f.eks. gnavere) modeller, og/eller modeller viser neurodegenerative fænotyper, er et værdifuldt redskab til at demonstrere i vivo relevans af den specifikke animalske modeller, eller terapeutisk behandlinger, der har til formål at forbedre sygdomssymptomer. Næsten hver tilgang til behandling af neurodegenerative sygdomme i sidste ende kræver test i en dyremodel før indledningen af kliniske forsøg med mennesker. Derfor er det afgørende at have pålidelig og reproducerbar adfærd tests, der kan bruges til at kvantificere konsekvent sygdom-relevante fænotyper langs alder progression, for at sikre, at en kandidat stof, som viste potentiale i en in vitro- model, kan effektivt forbedre fænotype i et levende dyr.
Et aspekt af motor adfærd vurdering i gnavere er kinematiske gangart analyse, som kan udføres af VPI'EN (også kaldet ventrale flyet videography)1,2. Denne etableret metode udnytter kontinuerlig registrering af undersiden af gnavere omvandrende oven på en gennemsigtig og motoriseret løbebånd bælte1,2,3,4. Analyse af video feed data skaber "digital pote print" af alle fire lemmer, der dynamisk og pålideligt sammenfatte de gnaver omvandrende mønster, som oprindeligt beskrevet af grønkål et al. 2 og Amende et al. 3.
Princippet om imaging-baserede gangart analyse er at måle området pote i kontakt med løbebånd bælte over tid, for hver individuel pote. Hver stilling er repræsenteret ved en stigning i pote område (i fasen for bremsning) og et fald i pote område (i fasen for fremdriftssystem). Dette er efterfulgt af swing fase hvor intet signal er konstateret. Swing og holdning sammen danner en skridtlængde. Ud over gangart dynamics parametre udvindes kropsholdning parametre også fra de optagne videoer. Eksemplarisk parametre og deres definition er angivet i tabel 1 og omfatter holdning bredde (SW; kombineret afstanden fra de fore eller hind poter til snude-hale akse), skridtlængde længde (SL; gennemsnitlige afstand mellem to fremskridt af den samme pote) eller paw placering vinkel (vinklen på pote til snude-hale akse). Kropsholdning og gangart dynamics dataene tillade drage konklusioner på animalsk balance (af kropsholdning parametre og deres variation over flere trin) og koordination (efter gangart dynamics parametre). Andre parametre, såsom ataksi koefficient (SL variabiliteten beregnet af [(max. SL−min. SL) / mener SL]), hind lemmer delt holdning tid (tid, der begge bagben lemmer er i kontakt med bæltet), eller pote træk (samlede areal i paw på bælte fra fuld holdning til pote lift-off) kan også udvindes, og er blevet rapporteret til at være ændret i forskellige neurodegenerative di sease modeller5,6,7,8 (Se tabel 1).

Vurdering af motor funktionsmåden for gnavere modeller for neurodegenerative sygdomme kan være udfordrende afhængigt af sværhedsgraden af Fænotypen af en bestemt model i en given alder. Flere sygdomme, de mest fremtrædende PD, Vis stærk motor adfærd (bevægelse) underskud, både hos patienter og i dyremodeller. En af de fire vigtigste symptomer i PD er bradykinesi, som udvikler sig med aldring og manifesterer sig i alvorlige gangart funktionshæmninger allerede i tidlige stadier af PD9. Undersøgelser af akut PD model, rotter behandlet med 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridin (MPTP), har allerede brugt VPI gangart analyse10,11,12. Dog behandler givet den akutte karakter af denne model, disse undersøgelser ikke de aldersrelaterede progression af motor underskud. Flere nyere undersøgelser har gennemført gangart analyse i alderen mus med neurodegenerative ændringer, for eksempel13,14,15, understreger relevansen af forståelse sygdomsprogression med fremrykkende alder .
Ud over motor underskud, Dyremodeller neurodegenerative sygdomme ofte har vanskeligheder med fokus på undersøgelse opgaver og Vis fremtrædende kognitive funktionsnedsættelser, især med fremrykkende alder. Sådan en fænotype kan påvirke resultatet af test, motor adfærd. Nemlig, en af de mest anvendte test til at undersøge motor underskud, rotarod test16, afhængig af kognition, opmærksomhed og stress17,18. Mens vilje til at gå på en motoriseret løbebånd afhænger også af disse faktorer, den registrerede udlæsning kører, som er en mere standardiseret funktion og langt mindre påvirket af ændrede kognition. Effekter af stress og opmærksomhed kan ses i specifikke parametre, som swing/holdning tid for stress, og SL for opmærksomhed19,20, men ikke i samlet kører evne.
Den kinematiske gangart analyse tilgang yderligere tilbyder fordelen at have muligheder for at justere udfordring for gnavere modeller. Løbebånd med justerbar vinkel og hastighed giver mulighed for gå hastigheder fra 0,1 - 99,9 cm/s, således at gnavere med svær omvandrende funktionsnedsættelser kan muligvis stadig køre en langsom hastighed (~ 10 cm/s). Non-nedsat dyr kan måles på hurtigere kører hastigheder (30 - 40 cm/s). Observation af eller ej de testede dyr er i stand til at køre på en bestemt hastighed giver et resultat af sig selv. Yderligere, gnaver kan desuden blive udfordret til at køre op en skråning eller ned en tilbagegang, ved at vippe løbebånd til en ønskede vinkel ved hjælp af en goniometer, eller ved at vedhæfte en vægtet slæde i mus eller rotte hind lemmer.
Ud over talrige undersøgelser af enkelt proteiner, der er muteret i patienter, er der en nylig øget bevidsthed om sammenhængen mellem defekte endocytose proces og neurodegeneration13,21,22, 23,24,25,26,27,28. Mus modeller med reducerede niveauer af endophilin-A (herefter endophilin), en central aktør i begge clathrin-medieret endocytose13,21,29,30,31 , 32 , 33 , 45 og clathrin-uafhængig endocytose34, blev fundet for at vise neurodegeneration og alder afhængige funktionsnedsættelse i bevægeapparatet aktivitet13,21. Tre gener indkode familie af endophilin proteiner: endophilin 1, endophilin 2, og endophilin 3. Navnlig fænotype som følge af nedbrydningen af proteiner, endophilin varierer meget afhængigt af antallet af mangler endophilin gener13,21. Mens tredobbelt knock-out (KO) af alle endophilin gener er dødbringende bare nogle få timer efter fødslen, og mus uden begge endophilin 1 og 2 ikke trives og dø inden 3 uger efter fødslen, viser enkelt KO for nogen af de tre endophilins ingen åbenlyse fænotype til testet betingelser21. Andre endophilin mutant genotyper Vis nedsat levetid og udvikle motoriske funktionsnedsættelser med stigende alder13. For eksempel, endophilin 1KO-2HT-3KO mus display omvandrende ombygninger og motor koordineringsproblemer (som testet af kinematiske gangart analyse og rotarod) allerede på 3 måneder, mens deres littermates, viser endophilin 1KO-2WT-3KO dyr, en betydelig reduktion i motor koordinering kun på 15 måneders alder13. Den store variation af fænotyper i disse modeller er det nødvendigt at identificere og anvende en test, der kan integrere en række udfordringer svarer til dyrets motor og kognition evner, samt alder. Her, detalje vi de eksperimentelle procedurer, at kapitalisere på den kinematiske ganganalyse at vurdere indsættende og progression af motoriske funktionsnedsættelser i en musemodel, der viser neurodegenerative ændringer (dvs. endophilin mutanter). Motor adfærd test er almindeligt anvendt til at bestemme den funktionelle relevansen af en gnaver model og for at teste nyligt udviklet behandlinger hos disse dyr. Specifikt, giver gangart analyse generobre sygdom relevante fænotyper, der er observeret i patienter, især i neurodegenerative sygdomme, der påvirker motoriske evner som Parkinsons sygdom (PD), Alzheimers sygdom (AD), amyotrofisk lateral sklerose (ALS), og andre. I tidlige undersøgelser langs denne linje, måling af gangart parametre var besværlige og af faktorer, der var svære at styre (fx kører hastighed, kontinuerlig kørende). Udviklingen af ventrale flyet imaging (VPI) systemer gjort det muligt at udføre ganganalyse på en stor skala, hvilket gør denne metode et nyttigt redskab til vurdering af motor adfærd i gnavere. For at illustrere brugen af kinematiske gangart analyse, vi har udført ganganalyse på WT C57BL/6J mus med fremrykkende alder, samt flere endophilin mutant linjer, ved hjælp af kommercielt tilgængelige instrumentering og software (der henvises til tabel af Materialer). This method can help answering key questions in the Aging and Neurodegeneration Research such as the onset and progression of gait and motor abnormalities with age and in particular, in mouse models of neurodegenerative diseases. The result of such status can be translated to a much better understanding of motor impairments in patients with neurodegenerative diseases for example, Parkinson's Disease or ataxias. The main advantage of using video based gait analysis is that it allows studying both subtle and severe motor impairments in mouse models of neurodegeneration.Begin by determining the health status of each animal by examining it closely and weighting it on a balance. Gently apply red finger paint to the animal's paws with a brush. Place the animal in a spare, clean cage while the paint dries.Next, set the speed of the treadmill on the top right panel of the apparatus. Then, place the animal in the test chamber and cover it with a dark cloth to allow each animal to adjust for one to two minutes. Turn on the light in the test chamber by turning the treadmill light rotary switch to the on position.Open the cover a little bit so that you can observe the animal during the task. Start the treadmill for about half a second to warn the animal that the task is about to begin. Turn the treadmill rotary switch to forward to start the treadmill.Then, click the Record button in the imager software. Once the animal runs in a stable manner, record for at least five seconds before stopping the treadmill. Stop the recording by clicking Stop on the imager software and turn the treadmill rotary switch back to the off position.Next, click the Edit button in the imager software to open a menu in which the start and end point of the video section can be set. Navigate through the video using the slider on the bottom of the screen. To select the current time point as the start or end point, click From Frame Number and To, respectively.Make sure the section contains at least seven steps per paw or fourteen steps in total of the animal running stably at a constant speed. Finally, enter the animal identification, birthdate, weight, and sex. Save the data on a desired location on the computer or server.The principle of video based gait analysis is to measure the paw area in contact with the surface over time for each individual paw. Many gait dynamics and posture parameters can be extracted from the recorded videos, for example, the swing time, stance time and width, stride length, and frequency. Other gait features such as ataxia coefficient or step angle variability can then be calculated from the primary measurements.Begin the video analysis by starting the analysis software and clicking Select Study Folder to select the folder with the recorded videos. Select one video and click Go.Use the re-draw function to select the area where the mouse is running. Note, this section should only contain the mouse and white background.Next, enter values in the length and width boxes to change the mask that excludes the red area around the snout of the animal for analysis. Adjust the sliders filter noise and filter fur and dark patches to optimize the black and white paw print. Set the filter noise slider to 800 to 950 for black animals and to 700 to 800 for brown or white animals, depending on the exact fur color of the animal, and select Okay.Then select a video that passed the first adjustment and select the Go function to start the analysis of the video. Next, select an analyzed video and click Go.Note that the paw print area in contact with the belt over time for each separate paw, can now be seen. To compare the original video and the calculated paw print for the selected area, select the Play Video function.To correct small mistakes, use either the Correct, Connect, or Delete option. Use the Correct option to delete a wrong signal. Click once to zoom into the relevant area and mark the left border of the object to remove with a second click and the right border with the third click.Use the Connect option to combine two signals. Click once to zoom into the relevant area and double click in the middle of the two objects to combine. Use the Delete option to remove time points from the analysis completely.Click once to zoom into the relevant area and mark the left border of the area to remove with the second click and the right border with the third click. Then select Next Limb to proceed through the four limbs. Note, after Next Limb is clicked after the last paw, the software completes the analysis and shows the results for the animal on four screens.Finally, after all videos from one experiment are analyzed, select all videos and click Reorganize Results to export the results. About 50 gait parameters are calculated and listed in the exported file. Sort the result by group, age, and running speed, and analyze all these conditions independently.In the gait analysis in wild type mice with advancing age, results indicated that the stance with normalized animal width does not change with advancing age. However, hind limb double support and limb loading do increase with age in wild type animals. Furthermore, while all tested animals can run at 30 meters per second at one month, the endophilin mutants develop running deficits as they age.Additionally, gait rhythm is impaired in endophilin mutants. For example, the propel time decreases in the endophilin mutants as the mice age. When attempting the procedure, it's important to remember that the most critical step is the recording of the original video.All further analysis depends on good quality videos. Gait analysis is a useful method to categorize gait and motor behavior in mouse models of neurodegeneration. However, some aspects of motor behavior, for example, muscle strength, are not adequately assessed by this method.But therefore a command complimenting video based gait analysis with a battery of other motor behavior tests, like grip strength, and beam walking. After watching this video, you should have a good understanding of how to perform gait analysis in mouse models that develop motor impairment with aging.
