Der Zebrabärbling ist ein relativ neues Modell für die Untersuchung V. Cholerae aber viel verspricht für die Zukunft Entdeckung der bisher unbekannte Aspekte von V. Cholerae Biologie und Pathogenese11,12,13 . Die Erwachsenen Zebrafisch-Modell hat die Vorteile der beiden natürlichen V. Cholerae Host, die intakt, Reife intestinalen Mikrobiota und ein Umweltmodell enthält. Nachteile des Modells sind ...

Vibrio Cholerae ist eine aquatische, gramnegatives Bakterium, das bewirkt, die menschliche Krankheit Cholera sowie sporadische Durchfall1,2 dass. V. Cholerae findet sich in der Umwelt in vielen Bereichen der Welt, oft in Verbindung mit anderen Wasserlebewesen. Diese verknüpfen Organismen gehören Plankton, Insekten Ei Massen, Schalentiere und Wirbeltier Fisch Arten3,4,5,6,7. Mehrere Studien haben V. Cholerae aus Darm-Trakt von Fischen in verschiedenen geographischen Gebieten7,8,9,10isoliert. Das Vorhandensein von V. Cholerae in Fisch zeigt, dass Fische als eine ökologische Reservoir fungieren können. Fisch könnte auch in der Übertragung der Krankheit auf den Menschen und die geographische Verbreitung von V. Cholerae Stämme6verwickelt werden.
Zum besseren Verständnis der Interaktion von V. Cholerae mit Fisch wurde Danio Rerio, besser bekannt als Zebrafisch als Modellsystem zur Untersuchung V. Cholerae11entwickelt. Zebrafisch stammen aus Südasien, einschließlich der Bucht von Bengal Region, die vermutlich das früheste Reservoir von V. Choleraewerden. Vor dem ersten Cholera pandemic Beginn im Jahre 1817 berichtete Cholera nicht außerhalb von, was jetzt Indien und Bangladesch ist. Daher Zebrafisch und V. Cholerae grenzender einander zugeordnet über evolutionäre Zeitskalen, was darauf hindeutet, dass Zebrafisch sind vielfältige V. Cholerae in der natürlichen Umwelt12.
Der Zebrafisch-Modell für V. Cholerae ist einfach durchzuführen und kann verwendet werden, um die gesamte pathogenen studieren V. Cholerae Lebenszyklus. Fische sind V. Cholerae durch Baden im Wasser ausgesetzt, die mit einer bekannten Anzahl von V. Choleraegeimpft worden. Innerhalb von wenigen Stunden findet intestinale Kolonisation statt, gefolgt von der Herstellung von Durchfall. Durchfall besteht aus Mucin, Proteine, ausgeschiedenen Bakterien und anderen Darminhalt. Der Grad der Durchfall kann mit ein paar einfachen Messungen13quantifiziert werden. V. Cholerae , das durch infizierte Fische ausgeschieden worden ist kann dann weiter zu naiv Fische infizieren Abschluss des ansteckenden Zyklus. Daher, rekapituliert der Zebrafisch-Modell der V. Cholerae menschliche Krankheit Prozess12,14.
Die am häufigsten verwendeten V. Cholerae Tiermodelle wurden historisch Mäusen und Kaninchen14,15,16,17,18. Diese Modelle waren maßgeblich an der Zugabe unseres Wissens von V. Cholerae Pathogenese. Jedoch weil Mäuse und Kaninchen nicht natürliche V. Cholerae sind Gastgeber, gibt es Einschränkungen, welche Aspekte des Lebenszyklus V. Cholerae studiert werden können. Die V. Cholerae Besiedlung von Mäusen und Kaninchen erfordert in der Regel das Fehlen von intestinalen Mikrobiota oder eine Vorbehandlung mit Antibiotika, um die intestinale Mikrobiota beschädigen. Beide Modelle erfordern entweder Magensonde einzuführen die Bakterien den Magen-Darm-Trakt oder chirurgische Manipulation die Bakterien direkt in den Darm zu injizieren. Zebrafisch haben den Vorteil, dass Erwachsene Fische mit einem intakten Darm Microbiota sind leicht besiedelt und die infektiöse Prozess geschieht auf natürliche Weise, ohne jede Manipulation erforderlich.
Die vorliegende Arbeit zeigt die Auslastung der Zebrafisch als Model in V. Cholerae Infektion. Die Infektion, Dissektion, Aufzählung der Kolonisation V. Choleraeund die Quantifizierung der Durchfall von V. Cholerae verursacht werden beschrieben12,13. Dieses Modell wird voraussichtlich für Wissenschaftler, die sowohl in den Krankheitsprozess V. Cholerae V. Cholerae ökologischen Lebensstil nützlich sein.

Vibrio Cholerae ist am besten bekannt als das infektiöse Agens, das die menschliche Krankheit Cholera verursacht. Außerhalb der menschlichen Wirt besteht V. Zebrafisch sind eine ideale Modellorganismus für viele Aspekte der Biologie, einschließlich Infektionskrankheiten zu studieren. Zebrafisch kann einfach und schnell von V. Cholerae nach Exposition im Wasser kolonisiert. Intestinale Kolonisation von V. Cholerae führt zu die Produktion von Durchfall und die Ausscheidung von replizierten V. Cholerae.
