Het succesvolle gebruik van dit protocol, is afhankelijk van de optimalisering van de verschillende stappen voor de plantensoorten, weefseltype en doel verbinding(en) van belang. De parameters die worden beschreven in het protocol vormen een goed uitgangspunt. De volgende experimentele beslissingen moeten worden gemaakt en getest: wel of niet gebruik (1) knippen of Onbesneden weefsel en (2) oplosmiddel of geen oplosmiddel, (3) welke oplosmiddel te gebruiken en in welke omvang, (4) wat de afstand van weefsel van de inlaat...

Planten bevatten een breed scala van kleine moleculen met uiteenlopende chemische eigenschappen. MS is een krachtige techniek voor het analyseren van plantenstoffen omdat het elementaire composities met een hoge gevoeligheid en specificiteit voor de detectie en de identificatie van metabolieten1 bieden kan. MS is meestal uitgevoerd op oplosmiddel-geëxtraheerde monsters, die worden gescheiden door chromatografie vóór de MS analyse1. Echter, het gebruik van vloeistofchromatografie (LC) vereist lange analyse keer en wordt vaak geassocieerd met een uitgebreide sample voorbereiding1. In tegenstelling, is directe chemische analyse van intacte weefsels die chromatografie omzeilt een zeer snelle techniek, minimal steekproef voorbereiding2vereisen. Dus, in gevallen waar de chromatografische stappen kunnen worden afstand, een directe chemische analyse kan nuttig zeer.
LC-MS (normaal) voor natuurlijke producten en metabolomica onderzoek berust op lange bulk extracties van gedroogde of diepgevroren plantaardig materiaal met meerdere weefsels en cel typen3. Anderzijds kan directe chemische analyse, zoals de MS detectie van metabolieten van plant weefsel, isoleren celtypes en voorbereiding artefacten4voorkomen. Blad spray MS, ook wel aangeduid als weefsel-spray5,6, is een directe ambient ionisatie MS-techniek, die in wezen geen monster voorbereiding5,7vereist. Blad spray die MS is nauw verwant aan papier spray MS, een ambient ionisatie-techniek met kenmerken van electrospray ionisatie die het mogelijk maakt voor de detectie van analyten dat zijn neergelegd op papier7. Ondanks de naam, blad spray MS is van toepassing op verschillende soorten plantaardige weefsels niet alleen bladeren en is aangetoond op fruit, zaden, wortels, bloemen weefsels en knollen, o.a.6,8,9, 10,11,12. De techniek vergemakkelijkt de ionisatie van endogene fytochemicaliën rechtstreeks vanuit het plantaardig materiaal in de massaspectrometer voor de detectie van8. Blad spray MS kan ook informatie bevatten over de ruimtelijke spreiding van chemische stoffen in verschillende weefseltypes (HLA) in planten13. Blad spray MS is in vergelijking met solvent extractie en LC-MS, suggereren de resultaten blad spray die MS voor de snelle detectie van oppervlakte metabolieten van unieke celtypes zoals plantenharen13 zorgt. Figuur 1 illustreert het blad spray MS experimentele opstelling. Directe electrospray ionisatie optreedt na slechts kleine bron wijzigingen. Een hoge spanning wordt toegepast op de plant weefsel via een metalen klem, produceren een spray van zeer geladen druppels vormen een Taylor-kegel die de ionen bij de inlaat van de ion van het MS. Electrospray draagt ionisatie optreedt vanaf de natuurlijke vloeistof van de plant of het oplosmiddel appl IED op het oppervlak van de plant. Een puntige tip op het weefsel vergemakkelijkt de electrospray en kan natuurlijk voorkomende of gemaakt door te snijden.
Blad spray MS is een snelle methode voor de kwalitatieve en semi-kwantitatieve analyse van intacte plantaardige weefsels die nut hebben gevonden voor een breed scala aan toepassingen. Bijvoorbeeld, is de techniek gebruikt om op te sporen van endogene stoffen te onderscheiden van verwante soorten, en zelfs om te evalueren van de wijzigingen in de dezelfde soort gegroeid onder verschillende omstandigheden. Eerdere studies hebben deze aanpak aangetoond door het meten van metabolieten in beautyberry (Callicarpa L.) 12 en Amerikaanse ginseng (Panax quinquefolium L.) 6. in het laatste voorbeeld, ginsenosides, amino zuren en oligosachariden na bevochtiging van ruwe ginseng weefsel kon worden opgespoord. Wilde en gekweekte Amerikaans ginseng werden onderscheiden van Knol segmenten6. Ginseng Knol integriteit werd volgende blad spray MS, waardoor voor een latere morfologische en microscopische inspectie6bewaard. Exogene stoffen op plant monsters kunnen bovendien ook worden gedetecteerd. Een aantal pesticiden (acetamiprid, difenylamine imazalil, linuron en thiabendazool) zijn op peel of pulp van fruit en groenten9ontdekt. Hoewel deze studies en vele anderen hebben het nut van blad spray MS aangetoond voor verschillende specifieke doeleinden, is een gedetailleerd protocol niet eerder gemeld.
De beschrijving van het protocol zal hier niet ingaan op de optimalisering van de methode voor een specifieke weefsel of samengestelde. Eerder, de detectie van mesembrine alkaloïden van Sceletium tortuosum (L.) N.E.Br (IJskruidfamilie) wordt gebruikt als een voorbeeld te bespreken van de nodige optimalisatie maatregelen die moeten worden genomen bij het opzetten van een blad spray MS experiment voor een soort, weefsel, of verbinding(en) voor de eerste keer. S. tortuosum is een succulente endemisch aan de semi-aride regio van het Karroo van Zuid-Afrika. Een traditionele geneeskunde van het San en de Khoi Khoi volkeren, het werd gebruikt voor de eetlust en dorst onderdrukking alsook wat betreft de psychotrope en pijnstillende effecten14,15. Momenteel, worden gestandaardiseerde extracten gebruikt voor de behandeling van neuropsychiatrische en neuropsychologische stoornissen16,17. De primaire verbindingen van belang bevatten het alkaloïde mesembrine en derivaten daarvan, waarvan vele ook in verwante Sceletium soorten15 gevonden zijn. Zowel wilde als gecultiveerde bevolkingsgroepen van S. tortuosum hebben variabele concentraties van mesembrine-alkaloïden, dus presenteren een kwaliteitscontrole uitdaging18. Een methode voor de snelle detectie van mesembrine-alkaloïden, zoals blad spray MS, kan zinvol zijn bij het toezicht op Sceletium producten. Omdat eerder, was er geen gedetailleerde visuele experimentele protocol voor het blad spray MS techniek, zullen we illustreren de methode met behulp van het voorbeeld van S. tortuosum, en het volgende wordt beschreven: de wijziging van de bron van een nanospray, de selectie en voorbereiding van de plantaardige weefsels, het verwerven van de data, de interpretatie van de resultaten, en het optimaliseren van de parameters van MS.

leaf spray mass spectrometry: a rapid ambient ionization technique to directly assess metabolites from plant tissues

Planten produceren duizenden kleine moleculen die divers in hun chemische eigenschappen zijn. De Spectrometrie van de massa (MS) is een krachtige techniek voor het analyseren van plant metabolieten omdat het molecuulgewicht met hoge gevoeligheid en specificiteit biedt. Blad spray MS is een ambient ionisatie techniek waar plant weefsel wordt gebruikt voor directe chemische analyse via electrospray, elimineren van chromatografie van het proces. Deze benadering van bemonstering van metabolieten zorgt voor een breed scala van chemische klassen gelijktijdig uit intact plantaardige weefsels, minimaliseren van de hoeveelheid van de bereiding van de monsters die nodig zijn worden gedetecteerd. Bij gebruik in combinatie met een hoge resolutie, nauwkeurige massa MS, vergemakkelijkt blad spray MS de snelle detectie van metabolieten van belang. Het is ook mogelijk om versnippering van de tandem massa gegevens met deze techniek om een samengestelde identificatie te vergemakkelijken te verzamelen. De combinatie van nauwkeurige metingen van de massa en versnippering is nuttig bij de bevestiging van de samengestelde identiteiten. Het blad spray MS techniek vereist slechts kleine wijzigingen aan een nanospray ionisatie bron en is een nuttig instrument om de mogelijkheden van een massaspectrometer verder uitbreiden. Hier, wordt vers blad weefsel van Sceletium tortuosum (Portulacaceae), een traditionele medicinale plant uit Zuid-Afrika, geanalyseerd; talrijke mesembrine-alkaloïden worden gedetecteerd met blad spray MS.

Op 10 weken die na ontkieming, vers verzameld serre gekweekte S. tortuosum verlaat werden geanalyseerd door blad spray MS.
