Im Protokoll beschriebenen Parameter bieten einen guten Ausgangspunkt dar. Pflanzen enthalten eine Vielzahl von kleinen Molekülen mit unterschiedlichen chemischen Eigenschaften. MS ist eine leistungsfähige Technik für Pflanzenstoffe zu analysieren, da es elementare Kompositionen eine hohe Sensitivität und Spezifität für die Detektion und Identifizierung von Metaboliten1verschaffen kann. Am häufigsten wird MS auf Lösungsmittel extrahierte Proben durchgeführt, die durch Chromatographie vor MS Analysis1getrennt sind. Jedoch der Flüssigchromatographie (LC) langen Analysenzeiten benötigt und ist oft verbunden mit einem umfangreichen Probe Vorbereitung1. Im Gegensatz dazu ist direkte chemische Analyse von intakten Gewebe, die Chromatographie umgeht eine sehr schnelle Technik, minimales Beispiel Vorbereitung2erfordern. In Fällen wo chromatographische Schritte verzichtet werden können, kann somit eine direkte chemische Analyse sehr vorteilhaft sein.
Typische LC-MS für natürliche Produkte und Metabolomics-Forschung stützt sich auf langwierige Bulk Extraktionen von getrockneten oder gefrorenen Pflanzenmaterial, enthält mehrere Gewebe und Zelle Arten3. Alternativ kann direkte chemische Analyse, wie die MS-Detektion von Metaboliten von Pflanzengewebe, Zelltypen zu isolieren und Vorbereitung Artefakte4vermeiden. Blatt-Spray MS, auch bezeichnet als Gewebe-Spray5,6, ist eine direkte ambient Ionisation MS-Technik, die im wesentlichen keine Probe Vorbereitung5,7erfordert. Blatt-Spray, die MS auf Papier Spray MS, eine ambient Ionisation Technik mit Merkmalen der Elektrospray-Ionisation verwandt ist, die für die Erkennung von Analyten ermöglicht, die auf Papier7abgelegt werden. Trotz des Namens Blatt Spray MS ist für verschiedene Arten von Pflanzengewebe, nicht nur Blätter und wurde auf Obst, Samen, Wurzeln, floral Gewebe und Knollen unter anderem6,8,9, demonstriert 10,11,12. Die Technik erleichtert die Ionisierung der endogenen Phytochemicals direkt aus pflanzlichen Stoffen in das Massenspektrometer für Erkennung8. Blatt-Spray MS bieten auch Informationen über die räumliche Verteilung der Chemikalien in verschiedenen Gewebetypen in Pflanzen13. Wenn Blatt Spray MS mit solvent-Extraktion und LC-MS verglichen wird, deuten die Ergebnisse Blatt-Spray, die MS für die schnelle Erkennung von Oberfläche Metaboliten von einzigartigen Zelltypen wie Trichome13ermöglicht. Abbildung 1 zeigt den Blatt Spray MS Versuchsaufbau. Direkte Elektrospray-Ionisation tritt nach nur geringfügigen Quelle Änderungen. Liegt eine hohe Spannung auf das pflanzliche Gewebe über eine Metallklammer, produzieren einen Spray von hoch geladenen Tröpfchen bilden einen Taylor-Kegel, der trägt die Ionen mit dem Ion-Einlass der MS. Electrospray Ionisierung tritt aus der natürlichen Flüssigkeit der Pflanze oder vom Lösungsmittel appl IED auf der Pflanzenoberfläche. Eine Spitze auf das Gewebe der Elektrospray erleichtert und kann natürlich vorkommende oder geschaffene durch Schneiden.
Blatt-Spray MS ist eine schnelle Methode für die semi-quantitative und qualitative Analyse der intakten Pflanzengewebe, die Dienstprogramm für vielfältige Anwendungen gefunden haben. Zum Beispiel wurde die Technik zur endogene Verbindungen, verwandte Arten zu unterscheiden und auch zu Änderungen in der gleichen Spezies, die unter verschiedenen Bedingungen gewachsen bewerten zu erkennen. Frühere Studien haben hierbei gezeigt, durch die Messung der Metaboliten in Beautyberry (Callicarpa L.) 12 und amerikanischer Ginseng (Panax Quinquefolium L.) 6. im zweiten Beispiel, ginsenoside, Aminosäuren und Oligosaccharide konnte nach Benetzung rohes Ginseng Gewebe nachgewiesen werden. Wilde und kultivierte amerikanischer Ginseng wurden von Knolle Scheiben6differenziert. Ginseng Knolle Integrität blieb nachfolgenden Blatt Spray MS, die für eine anschließende morphologische und mikroskopische Prüfung6erlaubt. Darüber hinaus können auch exogene Verbindungen auf Pflanzenproben nachgewiesen werden. Eine Reihe von Pestiziden (Acetamiprid, Diphenylamine, Imazalil, Linuron und Thiabendazol) wurden auf Schale und Fruchtfleisch von Obst und Gemüse9gefunden. Während diese Studien und viele andere das Dienstprogramm Blatt Spray MS für verschiedene spezifische Zwecke gezeigt haben, hat ein detailliertes Protokoll bisher nicht gemeldet.
Die Protokollbeschreibung wird hier nicht auf die Optimierung der Methode für eine bestimmte Gewebe oder zusammengesetzte konzentrieren. Vielmehr wird die Erkennung von Mesembrine Alkaloide von Sceletium Tortuosum (L.) N.E.Br. (Mittagsblumengewächsen) als Beispiel verwendet, um notwendigen Optimierungsmaßnahmen besprechen, die getroffen werden sollten, wenn Sie ein Blatt Spray MS Experiment für eine Art Gewebe, einrichten oder zum ersten Mal widerrufen. S. Tortuosum ist eine saftige endemisch in der semi-ariden Karroo-Region in Südafrika. Eine traditionelle Medizin der San und Khoi Khoi Völker, es diente für Appetit und Durst Unterdrückung sowie die psychotropen und analgetische Effekte14,15. Derzeit sind standardisierte Extrakte für die Behandlung von neuropsychiatrischen und neuropsychologischen Störungen16,17verwendet. Die primäre Verbindungen des Interesses enthalten das Alkaloid Mesembrine und seine Derivate, von die viele auch in verwandten Sceletium Arten15gefunden werden. Wilde und kultivierte Populationen von S. Tortuosum haben Variable Konzentrationen der Mesembrine Alkaloide, damit eine Qualitätskontrolle Herausforderung18präsentieren. Eine Methode für die schnelle Erkennung von Mesembrine Alkaloide, wie Blatt-Spray MS, kann bei der Überwachung der Sceletium Produkte hilfreich sein. Denn bisher es keine detaillierte visuelle experimentelles Protokoll für das Blatt-Spray MS Technik gab, wir zeigen die Methode am Beispiel von S. Tortuosumund Folgendes beschrieben: die Änderung einer Nanospray-Quelle, die Auswahl und Vorbereitung von Pflanzengewebe, die Erfassung der Daten, die Interpretation der Ergebnisse und die Optimierung der MS-Parameter.

leaf spray mass spectrometry: a rapid ambient ionization technique to directly assess metabolites from plant tissues

Pflanzen produzieren Tausende von kleinen Molekülen, die in ihren chemischen Eigenschaften unterschiedlich sind. Massenspektrometrie (MS) ist eine leistungsfähige Technik für Pflanze Metaboliten zu analysieren, denn es Molmassen mit hoher Sensitivität und Spezifität bietet. Blatt-Spray MS ist eine ambient Ionisation Technik wo Pflanzengewebe für direkte chemische Analyse über Elektrospray, Beseitigung von Chromatographie aus dem Prozess verwendet. Dieser Ansatz zur Probenahme Metaboliten ermöglicht eine Vielzahl von chemischen Klassen gleichzeitig von intakten Pflanzengewebe, Minimierung der Menge der Probenvorbereitung erforderlich erkannt werden. Bei Verwendung mit einem hochauflösenden, genaue Masse MS erleichtert Blatt Spray MS den Schnellnachweis von Metaboliten von Interesse. Es ist auch möglich, tandem mass Fragmentierung Datensammlung mit dieser Technik um eine zusammengesetzte Identifizierung zu erleichtern. Die Kombination aus präzise massenmessungen und Fragmentierung ist vorteilhaft bei der Bestätigung zusammengesetzte Identitäten. Das Blatt-Spray MS Technik erfordert nur geringfügige Änderungen Nanospray Ionisierung Quelle und ist ein nützliches Werkzeug zum Ausbau der Fähigkeiten eines Massenspektrometers. Hier wird frisches Blattgewebe aus Sceletium Tortuosum (Mittagsblumengewächsen), eine traditionelle Heilpflanze aus Südafrika, analysiert; zahlreiche Mesembrine Alkaloide werden mit Blatt-Spray MS erkannt.

Bei 10 Wochen nach dem Keimen, frisch gesammelten Gewächshaus gewachsen S. Tortuosum Blätter wurden von Blatt Spray MS analysiert. Die experimentelle Workflow zum Nachweis von Metaboliten von S. Tortuosum verlässt mit Blatt-Spray, die MS in Abbildung 2dargestellt ist.
