Kunststoff wurde erstmals Anfang der 1930er Jahre1im Meer beobachtet. Jüngste Schätzungen des meeresmülls Kunststoff Sortiment aus über 243.000 Tonnen (MT) aus Kunststoff auf der Oberfläche des Ozeans bis 4,8-12,7 Millionen MT aus Kunststoff in den Ozean von terrestrischen Quellen jährlich2,3. Frühe Studien auf Kunststoff meeresmüll konzentrierte sich auf Macroplastics (> 5 mm Durchmesser) sind gut sichtbar und messbar. Jedoch wurde es vor kurzem entdeckt, dass Macroplastics repräsentieren < 10 % Kunststoff Schutt, Graf, im Ozean, darauf hinweist, dass die überwältigende Mehrheit der Kunststoff Schutt Microplastic (< 5 mm Durchmesser)2.
Mikroplastik werden in zwei Gruppen kategorisiert: primäre und sekundäre mikroplastik. Primäre mikroplastik bestehen aus Kunststoffen, die bei einem Durchmesser hergestellt werden < 5 mm und beinhalten Nurdles, die Roh Pellets verwendet, um Konsumgüter, Microbeads als Peelings in Körperpflege-Produkten (z. B. Gesicht waschen, Körper-Peeling, Zahnpasta) und Schleifmittel oder Schmiermittel in der Industrie. Sekundäre mikroplastik sind in der Umgebung erstellt, da größere Kunststoff Schutt durch Photolyse, Abrieb und mikrobielle Zersetzung4,5fragmentiert ist. Synthetische Fasern sind auch sekundäre mikroplastik und ein wachsendes Problem. Eine einzige Kleidungsstück kann loslassen > 1.900 Fasern pro Waschgang in einer haushaltswaschmaschine6. Diese Mikrofasern sowie Microbeads von Körperpflege-Produkten, in die Kanalisation Kanalisation und gewaschen, vor dem Eintritt in Kläranlagen Behandlungen. Murphy (2016) festgestellt, dass eine Kläranlage mit einer Bevölkerung von 650.000 Microplastic Konzentration von 98,4 % von Zufluss auf Abwasser, reduziert noch 65 Millionen mikroplastik in Abwasser blieb und Klärschlamm jeden Tag7. Aufgrund ihrer ökologischen Freilassung wurden über alle trophischen Ebenen12,13,14,15mikroplastik im Verdauungs- und Atemwege Gewebe von marinen Organismen gefunden. Ihre Wirkung nach Aufnahme variabel, mit einigen ist Studien nicht beobachten Schaden, während andere zahlreiche Effekte wie physikalischen und chemischen Gewebe Schaden4,6,14,15zeigen. Aufgrund dieser Erkenntnisse hat Interesse in diesem Bereich in den vergangenen fünf Jahrzehnten zugenommen. Aber erst kürzlich haben Sie Studien begonnen, quantifizieren Kunststoff Schutt, besonders mikroplastik in Süßwasser-Systemen, wie Flüsse und Seen, oder die Auswirkungen auf Organismen, die in diesen Lebensräumen12,16wohnt, 17,18. Flüsse sind eine wichtige Quelle für Kunststoff Trümmer im Meer gefunden, wie sie erhalten Abwasser Abwasser und Oberflächenwasser Abfluss, die mikroplastik und Macroplastics enthalten.
Das Protokoll hier kann verwendet werden, Microplastic Proben zu sammeln, wo die Treibnetze sind nicht möglich; insbesondere Treibgut in aquatischen Umgebungen mit hohen Konzentrationen von suspendierten Sedimenten und großen wie den Mississippi River. Einzugsgebietes des Mississippi River ist einer der größten der Welt und hat eine Bevölkerung von > 90 Millionen Menschen und ist wahrscheinlich damit eine der größten Quellen von Kunststoff Schutt auf dem Ozean19,20. Jedes Jahr den Mississippi River entlädt einen Durchschnitt von 735 Kilometer3 von Süßwasser in den Golf von Mexiko, zusammen mit hohen Konzentrationen von suspendierten Sedimenten (~ 60 bis > 800 mg/L) und großen Trümmer13,21. Wasserproben wurden an verschiedenen Orten entlang des Mississippi River und seinen Nebenflüssen in transluzenten 1 L Low-Density Polyethylen (PE) Container mittels einer peristaltischen Pumpe in zwei tiefen (d. h. Oberfläche und 0,6-Tiefe) gesammelt. Im Labor wurden Proben gefiltert mit Nylon-Mesh-Siebe und gemischte Zellulose Ester Membranfilter gleichzeitig mit einem maßgeschneiderten 63,5 mm (2,5 Zoll) Polyvinylchlorid (PVC) Zylinder mit Fugen, Siebe und Filter22einzufügen. Die Einbeziehung der PVC-Gewerkschaften in der Luftfiltration Gerät ermöglicht für die Filtration durch so viele oder so wenige partikelgrößenklassen wie gewünscht. Darüber hinaus kann verwendet werden, um Microplastic Schutt bis Sub-Mikrometer-Größen mit Membranfilter, bei der Untersuchung von synthetischer Fasern zu erfassen. Sobald gefiltert, Proben wurden getrocknet und vermuteten Kunststoffe wurden identifiziert und aus dem Netz Siebe und Membranfilter unter einem Stereomikroskop sortiert. Mutmaßliche Kunststoffe wurden dann untersucht mit Mikro-gedämpft totale Reflexion Fourier-Transform-Infrarot-Spektroskopie (Mikro ATR-FTIR) zur Beseitigung von nicht-synthetische Materialien oder Polymertyp zu bestimmen. Angesichts der Größe der Microplastic Partikel und Fasern ist die Verschmutzung an der Tagesordnung. Kontaminationsquellen sind atmosphärische Deposition, Kleidung, Feld und Laborgeräte sowie deionisiertes (DI) Wasser Quellen. Mehrere Schritte sind im gesamten Protokoll zur Verringerung der Kontamination aus verschiedenen Quellen während der Durchführung alle Phasen der Studie enthalten.

Die Omnipräsenz des Kunststoff Trümmer im Meer ist von dem öffentlichen, wissenschaftlichen Gemeinschaften und Behörden anerkannt. Jedoch haben erst vor kurzem mikroplastik in Süßwasser-Systemen, wie Flüsse und Seen, quantifiziert worden. Microplastic Probenahme an der Oberfläche in der Regel besteht der Bereitstellung von Treibnetzen hinter entweder einen stationären oder beweglichen Boot, das die Probenahme für Umgebungen mit niedrigen Niveaus der suspendierten Sedimente und unter Wasser oder schwimmend Schutt begrenzt. Filtration erfolgte mit einem maßgeschneiderten Microplastic Filtration Gerät mit abnehmbaren Fugen, die Nylon-Mesh-Siebe untergebracht und Zellulose Ester Membranfilter gemischt. Netz-Siebe und Membranfilter wurden untersucht, mit einem Stereomikroskop zu quantifizieren und Microplastic Partikel und Fasern zu trennen. Diese Materialien wurden dann untersucht mit einer Mikro-gedämpft total Reflexion Fourier-Transform-Infrarot-Spektrometer (Mikro ATR-FTIR), um Microplastic Polymer-Typ zu bestimmen. Erholung wurde durch festnagelnd Proben mit blauen PE-Partikel und grünen Nylon-Fasern gemessen; Prozent Erholung wurde zu 100 % für Partikel und 92 % für Fasern bestimmt. Dieses Protokoll wird ähnliche Studien auf mikroplastik in hoher Geschwindigkeit Flüsse mit hohen Konzentrationen von Sedimenten führen. Mit einfachen Modifikationen der peristaltischen Pumpe und Luftfiltration Gerät können Benutzer sammeln und analysieren verschiedene Probenvolumen und partikuläre Größen.

This method will help researchers quantify microplastic loads in rivers, which often contain high sediment loads, as they are a major source of plastic debris to the ocean. The main advantage of this technique is that it enables filtering and sorting of microplastics and water samples with high sediment loads at sizes not typically included in previous studies. To begin rinse the filtration device and nylon mesh sieves three times with deionized water.Then place the mesh sieves into each union joint, with the pour sizes decreasing from top to bottom. To prevent leaks seal each joint tightly. Next wet a mixed cellulose ester membrane filter with deionized water.While the filter is still damp fold it into a cone. Then place a stainless steel mesh basket into the last union joint. Carefully place the folded membrane filter into the basket.Fold the lip of the filter over the edge of the joint. Next place a mesh sieve on top of the membrane filter in the last union joint. Ensure that all of the union joints are sealed tightly.Then attach the hose form the top of the filtering flask to the base of the filtration device. Turn on the vacuum pump, ensuring that the pressure does not exceed 127 milliliters of mercury. Using a 500 milliliter graduated cylinder measure the total volume of the sample.Next record the volume of the sample and transfer it to the filtration device. To empty the filtering flask turn off the pump and detach the two hoses from the flask. Then empty the flask into a waste container.To continue the filtration cycle reattach the hoses and turn on the pump. Once the entire sample has been filtered use deionized water to rinse the sample container and graduated cylinder three times. After each rinse filter the deionized water used to rinse the container and graduated cylinder.Using deionized water rinse the walls of the filtration device three times. Then turn off the vacuum pump, and use a deionized water wash bottle to rinse the edges of the union joint. Turn the pump off and use forceps to remove the mesh sieve from the union joint.Place the sieve in a covered Petri Dish and dry it at 60 degrees celsius for 24 hours. Repeat this process for each union joint. Then turn the vacuum pump on and rinse the edges of a membrane filter using a deionized water wash bottle.Wash the particulates at the edges of the filter into the center, and ensure that all of the water passes through the filter. Next use forceps to remove and unfold the membrane filter. Place the filter in a foil envelope and dry at 60 degrees celsius for 24 hours.First examine the membrane filter under a stereo microscope. Suspected plastics will not have cellular structure. The fibers will have equal thickness throughout, and the particles will not appear shiny.Remove suspected plastics from the filter and place them into a collection vial containing 70%ethanol. Record the color and shape of each suspected plastic. For nylon mesh sieves stored in Petri Dishes after all suspected plastics have been removed from the filter examine the lid and bottom of the Petri Dish for additional suspected plastics.To validate this protocol three samples from Oso Bay were spiked with 10 blue polyethylene particles and 50 green nylon fibers. On average, 100%of the polyethylene particles and 92%of the nylon fibers were recovered from the samples. The loss of fibers may be due to a small amount of sample loss during filtration or incorrect identification.Once mastered multiple filtration apparatus can be run simultaneously with samples taking less than two hours each to filter. Sample sorting times under the microscope however are sample specific. While attempting this procedure it's important to remember to account for potential contamination using lab equipment and field blanks for each step in the process.Following this procedure other methods like Fourier Transform Infrared Spectroscopy can be used to verify material properties. With its development this technique enables researchers studying environmental contaminates to explore microplastic pollution and waterways with high suspended sediment loads, as well as floating and submerged debris.
