We geïntroduceerd hierin, het ontwerp en gebruik van een toegankelijke, effectieve Caenorhabditis zeef als een instrument voor het sorteren en het onderhouden van C. elegans. De nematode worm, Caenorhabditis elegans, is een premier model-organisme. Naast het eenvoudig en gecontroleerde karakter van hun teelt in het laboratorium, hun gehele genoom is gesequenceerd1 en het ontwikkelings lot van elke cel2is bekend. Als gevolg van deze functies is C. elegans een veel gebruikte modelorganisme voor genetische studies. Nochtans, samen met deze gunstige kenmerken komen sommige uitdagingen voor onderzoekers. Als gevolg van hun snelle generatietijd, C. elegans populaties kunnen snel opraken van voedsel en/of populaties met meerdere generaties worden gemengd en ontwikkelingsstadia presenteren in een keer. Zo vereisen experimenten uitgevoerd op solide nematode groei media (NGM) onderzoekers om fysiek naar dieren verse platen voordat de bacteriële voedselbron uitput en nieuwe larven ontwikkelen. Dit kan vervelend zijn als een frequente overbrengen van de dieren is nodig om te voorkomen dat de experimentele populaties van steeds vermengd met generaties nakomelingen. Enkele experimenten vereisen nog, beide grote aantallen dieren en langere tijd punten (bijvoorbeeld, DNA of RNA extractie in volwassenheid). Deze verbindingen de uitdagingen van nauwkeurig vasthouden van een gesynchroniseerde bevolking en de overdracht van grote aantallen dieren.
Huidige methoden van de overdracht van C. elegans gekweekte op NGM zijn plukken of wassen van de dieren uit plaat op plaat; chemisch behandeling van de dieren (bijvoorbeeldmet de DNA-replicatie remmer fluorodeoxyuridine of FUDR); of stroom cytometry gebruikt voor het sorteren van de dieren in de multi goed platen. Picken impliceert het gebruik van een handje, gemaakt met een dunne draad van platina of een wimper, handmatig overbrengen individu of meerdere dieren3,4. Deze methode is nauwkeurig maar vereist zowel de vaardigheid en de tijd en is een beperking voor studies waarbij grote aantallen dieren. Mei ook plukken worden fysiek schadelijk en stressvolle tot aan de dieren door individuen mogelijk te onderwerpen aan onnatuurlijk en inconsistent bedragen van verstoring en kracht. Wassen gaat spoelen van een cultuur-schotel met een bufferoplossing en overbrengen van de oplossing met de dieren via glazen pipet van Pasteur aan een nieuwe plaat van de cultuur. Deze methode is snel en efficiënt, maar is niet juist als meerdere generaties en ontwikkelingsstadia van dieren worden overgebracht in bulk. Chemische behandelingen, zoals FUDR, kunnen worden ontbonden in de kweken media om te voorkomen dat de productie van nakomelingen door het blokkeren van een DNA-replicatie, en dus de gameet-productie en ei-ontwikkeling. Tijdje effectief, deze methode moet worden toegepast na developmental rijping over niet verstoren de normale ontwikkelings processen, en dit betekent dat er nog steeds sprake is van een eis tot overdracht van de dieren vóór haar administratie3. Deze methode beïnvloedt ook meerdere mobiele signalering van trajecten, wat resulteert in merkbare gevolgen voor de dieren, naarmate zij ouder worden (b.v., een verlenging van de levensduur of een gewijzigde proteostasis) afhankelijk van de stam van C. elegans gebruikt5, 6,7,,8,,9,10. Stroom cytometry methoden automatisch sorteren en individuele C. elegans van één Multi goed bord overbrengen in een andere11. Hoewel deze methode zeer effectief en efficiënt is, is stroom cytometry apparatuur onbetaalbaar duur en ontoegankelijk voor veel onderzoekers. Een alternatief voor het overbrengen van de dieren is het gebruik van mutant modellen die zijn temperatuur gevoelig, zoals fer-15 en fem-1, die steriel met temperatuur aanpassing12geworden. Terwijl met behulp van mutant dieren nuttig in sommige situaties is, deze specifieke stammen groeien langzamer dan wild-type dieren en ze rekenen op een gewijzigde genoom, arme vertegenwoordigers voor veroudering of gezonde wormen bijeenkomen. Bovendien, de afhankelijkheid van een verschuiving van de temperatuur ertoe steriliteit resulteert ook in het ontbreken van een statische omgeving en temperatuurveranderingen gemakkelijk is aangetoond dat de invloed van gen expressies13,14, 15. onderzoeksgroepen technieken met een beschrijving van het gebruik van een Maas wilt filteren C. elegans grootte16eerder hebt gepubliceerd. Maar waren we niet in staat om vorige werk testen op eventuele wijzigingen in de algemene gezondheidsresultaten dat geassocieerd met het gebruik van deze filters worden kunnen te vinden.
Er is dus behoefte aan een binnen de Gemeenschap van C. elegans onderzoek een betaalbare, efficiënte, snelle en nauwkeurige methode voor het overbrengen van grote aantallen dieren tussen cultuur platen. We hebben een verbeterde, toegankelijke stuk van apparatuur (genoemd de Caenorhabditis zeef) en een bijbehorende protocol voor de fabricage en de werking die voldoet aan de behoeften van de onderzoekgemeenschap C. elegans ontwikkeld. Hierin delen we het ontwerp van de Caenorhabditis zeef en methoden voor het gebruik ervan, en we laten zien dat het gebruik ervan heeft geen gevolgen voor de gezondheid van de gemeenschappelijk of eventuele stress markeringen in vergelijking met standaard manuele picking en een behandeling met de veelgebruikte, beperking van de vruchtbaarheid chemische FUDR.

caenorhabditis sieve: a low-tech instrument and methodology for sorting small multicellular organisms

Caenorhabditis elegans (C. elegans) is een reeds lang gevestigde model-organisme gebruikt in een heel scala van fundamenteel en biomedisch onderzoek. Binnen de nematode onderzoeksgemeenschap is er een behoefte aan een betaalbare en effectieve manier om grote, leeftijd-matched populaties van C. elegans. Hier presenteren we een methode voor het mechanisch sorteren en reiniging van C. elegans. Ons doel is om een rendabel, efficiënt, snel en eenvoudig proces om te verkrijgen van dieren van uniforme afmetingen en levensfasen voor hun gebruik in experimenten. Deze tool, zeef Caenorhabditis hanteert een custom-built deksel die draden op gemeenschappelijke conische lab buizen en sorteert C. elegans gebaseerd op lichaamsgrootte. We laten ook zien dat de Caenorhabditis zeef effectief dieren van één cultuur plaat brengt naar een ander toe te staan voor een snelle sorteren, synchroniseren, en het schoonmaken zonder de beïnvloedende markers van gezondheid, met inbegrip van de motiliteit en stress-afleidbare gen de verslaggevers. Deze toegankelijk en innovatieve tool is een snelle, efficiënte en niet-stressvolle optie voor het handhaven van C. elegans populaties.

Caenorhabditis Sieve: A Low-tech Instrument and Methodology for Sorting Small Multicellular Organisms

This method can help answer key questions in molecular biology, involving assays that require specific developmental stages of C.elegans. Like assays of gene and protein expression levels. The advantage of this technique is that it allows for accurate and efficient selection of large numbers of C.elegans by body size, this facilitates the use of assays that would normally be time and cost prohibitive.Generally, individuals new to this method will struggle because they can't perform the arching turnover step or they don't keep solution flow in the center of the mesh. Visual demonstration of this method is critical, as the washing and flipping steps are difficult to learn. Some may have trouble interpreting and visualizing specific motions in text.To begin, acquire two 50 mL conical tubes with lids, then use a stepped drill bit to remove the center from the inner lip of the lids. If necessary, use a file, sandpaper, or a rotary tool to clean and sand the edges and tops of the lids. Then cut a disc of nylon monofilament woven mesh, slightly smaller than the diameter of the lid.Texture the top surfaces of the lids, and clean them with ethanol. After allowing the lids to dry, apply cyanoacrylate glue to the top surfaces of both lids. Next, lay the monofilament mesh disc on one glued lid.Invert the second lid on top of the mesh, and press the lids firmly together. After the first layer of glue dries, apply a generous ring of cyanoacrylate glue around the outer gap between the lids. Next, label the filter with its respective mesh pore size.First, place the sieve over a labeled 50 mL conical tube. Then pipette saline solution through the center of the sieve until a droplet forms, coalesces, and drips off of the center of the filter. Using M9 buffer, wash a population of C.elegans off of an auger plate, then use a glass pasteur pipette to transfer the buffer containing the worms on the top side of the monofilament mesh.Operating in the center of the mesh, rinse the filter with M9 buffer, repeat this rinse step as needed to pass all bacteria and smaller worms through the mesh. Next, attach a new 50 mL conical tube to the top of the sieve, the previously collected worms larger than the pores in the mesh will be facing the inside of the new tube. Remove the waste tube and quickly flip the sieve.Then rinse the mesh with M9 buffer into the new collection tube. Allow the collected worms to settle for approximately one minute, then carefully aspirate the buffer solution away from the pellet of worms. Using a glass pasteur pipette, create droplets of worms on an NGM plate and allow them to dry.Finally, rinse the sieve with reverse-osmosis water and ethanol. After the sieve dries, store it in a clean container for future use. Discard the sieve when it develops a sagging appearance.In this protocol a sieve was constructed to sort and clean age-matched populations of C.elegans. Animals in the sieve treatment group exhibited normal and spontaneous movement pattern throughout adulthood. Additionally, the pick and sieve groups did not show any significant differences in pharyngeal pump rate.Animals in the sieve group also responded normally to anterior or posterior mechanical stimulus when compared to all of the treatment groups on any of the testing days. Finally, the fecundity of the treatment groups was compared;the use of the sieve did not significantly impact the number of progeny when compared to the pick treatment group. Once mastered, this technique can be done in less than ten minutes.While attempting this procedure, it's important to remember to operate in the center of the mesh area. After watching this video, you should have a good understanding of how to assemble and properly use a Caenorhabditis sieve to sort worms by body size.
