この方法は、これらの細胞タイプにおける様々なタンパク質の細胞内局在化とダイナミクスに関するマクロファージおよび樹状細胞生物学の分野における主要な質問に答えるのに役立ちます。この技術の主な利点は、比較的簡単で安価であり、他の利用可能な方法の大部分よりも優れた効率と発現安定性を示していることです。手順を実証することは、私の研究室の大学院生であるヤルミラ・クラロワです。レトロウイルスを生成するには、白金エコ包装細胞の単細胞懸濁液を10センチメートルのシャーレにプレートし、培養物が50〜60%コンフルエントになるまで10%のウシ胎児血清(FBS)を補充して15ミリリットルのDMEMで細胞を培養する。翌日、関心のあるレトロウイルス構造の20マイクログラムとPCLエコパッケージングベクターの10マイクログラムを、穏やかな混合で血清なしで1ミリリットルのDMEMに加えます。血清と抗生物質を含まない1ミリリットルのDMEMに75マイクロリットルのPEIを加え、室温で5分間インキュベーションする2番目のチューブに2番目のチューブに加え、両方のチューブの内容物を混ぜ合わせて室温でさらに10分間インキュベーションします。次に、白金エコセルの培地を、2%FBSで補った新鮮な37°CのDMEMを8ミリリットルに慎重に交換し、トランスフェクション混合物を滴下でプレートに慎重に加えます。摂氏37度で4時間のインキュベーションを行った後、スーパーナテントを摂氏37度のDMEMの10ミリリットルに置き換え、摂氏37度で24時間インキュベーションを行う10%FBSを補います。翌日、5ミリリットルのピペットを使用して、エコトロピックレトロウイルス粒子含有スーパーナテントを15ミリリットル遠心管に移し、遠心分離によって残骸および含有細胞を除去する。一方、摂氏37度のDMEMと10%FBSの10ミリリットルを37°Cでさらに24時間インキュベーションして培養皿に加え、ちょうど実証したようにウイルス粒子の第2ラウンドを収集します。骨髄を収穫するには、組織培養フードで、ピンセットとハサミを使用して後肢から皮膚と筋肉の一部を取り除き、大腿骨を壊すことなく股関節からアセタブラムを慎重に取り除きます。足首の関節で足を切り、骨に70%エタノールを吹き付けます。その後、ペーパータオルで筋肉の残りの部分を取り除き、2%FBSを補ったPBSの5センチメートルのシャーレに骨を入れます。次に、各骨の膝関節を曲げ、慎重にはさみで骨を分離します。はさみを使用して1本の骨の各骨端部の約1〜2ミリメートルを取り除き、新鮮なPBSとFBSを搭載した30ゲージ針を装備した2〜5ミリリットルの注射器を使用して、骨の両端から骨髄を15ミリリットルの遠心管に洗い流します。各骨から骨髄を採取すると、骨細胞を遠心分離によりペレット化し、ペレットを2.5ミリリットルの赤血球リシスバッファーに室温で2〜3分間再懸濁する。lysingの後、100マイクロメートルの細胞ストレーナーを通して新しい15ミリリットル遠心分離管に細胞をフィルターし、12ミリリットルのPBSプラスFBSで粘り強さを回復させます。DMEMでペレットを再懸濁し、10センチメートルで6番目の骨髄細胞に5〜10回を数え、プレートするための10%FBSおよび抗生物質を補充し、非組織培養は血清およびマクロファージコロニー刺激因子、またはMCSFを補ったDMEMの10ミリリットルでペトリ皿を処理した。培養5日目には、細胞培養皿を慎重に傾け、約5ミリリットルの培地を除く全ての培地を皿の端近くの表面から取り除きます。次に、FBSとMCSFを添加した新鮮な37度の摂氏培地を20ミリリットル加え、細胞を細胞培養インキュベーターに戻します。細胞を収穫するには、6日目から7日目に、培地および浮遊細胞のすべてを取り除き、37°CPBSで培養液を洗浄する。組織培養インキュベーターで5ミリリットルの0.02%EDTAとPBSを3〜5分間摂氏37度で取り外し、5ミリリットルのピペットを使用して解離した細胞をプレート底から静かに洗い流します。次に、新鮮なPBSの25ミリリットルを含む50ミリリットル遠心管に細胞を移す。骨髄由来抗原提示細胞におけるEGFPタグタンパク質発現を誘導するために、細胞分化プロトコルの開始時に、骨髄細胞をDMEMプラスFBSおよび抗生物質の1ミリリットル当たり6番目の細胞に2〜5倍に希釈し、1ミリリットルの培地の細胞をプレート化し、適切な分化サイトカインを補充する。細胞培養インキュベーターで4〜6時間後、ポリブレンを添加した収集されたウイルスの第1ラウンドの2ミリリットルを加え、遠心分離と細胞培養インキュベーター内の4時間の培養によって細胞培養をトランスデュースする。伝達期間の終わりに、非接着性細胞を各ウェルから個々の15ミリリットル遠心分離チューブに移し、遠心分離によって細胞を採取する。より多くの細胞が必要な場合、細胞は複数のウェルで同じコンストラクトに感染し、分化前にプールすることができます。次に、適切な分化サイトカインを添加した培養培地の10ミリリットルでペレットを再懸濁し、ちょうど実証したように、チューブ当たりのシャーレを処理した10センチメートルの非組織培養物に細胞をプレートする。スーパーナテントを含む第2ウイルスの回収後のフローサイトメトリーによる白金エコトランスフェクションの有効性の評価により、PSTPIP2-EGFPでは62%、OPAL1-EGFPでは53%の平均トランスフェクション効率を明らかにしています。レトロウイルス伝達は、両方の培養物の細胞の90%以上を有する骨髄由来細胞の分化状態に影響を及ぼさない。そして、適切な特徴的な形態変化を用いて、両方のタイプの分化細胞に対して観察された。なお、EGFPの全体的な発現は、レトロウイルスの種類に関係なく、骨髄由来マクロファージEGFP発現と比較して骨髄由来樹状細胞培養において低かった。この手順を試みる間、実験のために皿から細胞を取り除く予定の場合、細菌を培養するために通常使用される非組織、培養処理プラスチック皿を使用することを忘れないでください。この手順に従って、他の方法は、生細胞イメージング、超解像顕微鏡または他のタイプの顕微鏡検査を使用して、対象となる樹状細胞およびマクロファージタンパク質の細胞内局在化およびダイナミクスに関する追加の質問に答えることができる。この技術は、免疫学分野の研究者がマクロファージや樹状細胞によって発現するタンパク質の分布と容量関係を探求し、免疫応答中のこれらのタンパク質の機能の理解を深める道を開いた。ウイルス粒子の処理は非常に危険であり、エアロゾル発生の最小化、保護具の着用、ラミナーフローフードの使用などの予防措置は、常にこの手順を実行する間に取られるべきであることを忘れないでください。
