Omdat de vaststelling van geschikte diermodellen voor ziekten bij de mens is de sleutel tot het vinden van een remedie, hebben passende dieren modellen lang voortgezet en verbeterd na verloop van tijd. Meerdere stammen van immuungecompromitteerde lymfkliertest modellen zijn ontwikkeld die de engraftment van menselijke cellen en/of weefsels en de latere uitvoering van gehumaniseerd functies1,2 toelaten. Dergelijke gehumaniseerd Muismodellen staan kritisch tegenover voor onderzoek van mens-specifieke ziekten3,4,5.
Acquired immune deficiency syndrome (AIDS) als gevolg van infectie met het humaan immunodeficiëntie virus (HIV) is een voorbeeld. Voorafgaand aan de vaststelling van gehumaniseerd Muismodellen beperkt ethische en technische beperkingen HIV/AIDS preklinische dierstudies naar niet-menselijke primaten3. Echter, de hoge kosten en vereisten voor gespecialiseerde zorg voor zo'n dier een belemmering vormen voor HIV/AIDS studies in typische academische instellingen. HIV voornamelijk infecteert menselijke CD4 + T-cellen en impact van de ontwikkeling en de immuunrespons van andere menselijke immune cellen zoals B-cellen, macrofagen en dendritische cellen6; kleine dierlijke modellen met een functionele menselijk immuunsysteem getransplanteerd zijn daarom in hoge vraag.
Een doorbraak kwam in 1988, toen CB17 -scid muizen met een mutatie Prkdcscid ontwikkeld en succesvolle engraftment van het menselijke immuunsysteem1toonde. De resultaten van de mutatie Prkdcscid in defecte T - en B-cel-functies en een ablated adaptieve immuunsysteem in muizen, waardoor de engraftment van menselijke perifere bloed mononucleaire cellen (PBMCs), hematopoietische stamcellen (HSCs), en foetale hematopoietische weefsels7,8. Niettemin, lage niveaus van engraftment worden vaak waargenomen bij dit model; mogelijke oorzaken zijn 1) residuele aangeboren immuun activiteit gemoduleerd via natural killer (NK)-cellen en 2) de ontwikkeling van de late-stadium van muis T - en B-cellen (leakiness)5. De verdere ontwikkeling van de nietzwaarlijvige diabetische (NOD)-scid muismodel bereikt dramatische down-regulatie van de activiteit van de NK-cel; het is dus een hoger niveau en meer duurzame engraftment van menselijke immuunsysteem componenten9kunnen steunen. Verder onderdrukken of belemmeren de ontwikkeling van aangeboren immuniteit, Muismodellen truncation of totale knockout van de interleukin-2 receptor γ-keten (Il2rg) rekening houdend met in de (NOD) -scid achtergrond werden opgericht. Il2rg, ook bekend als gemeenschappelijk cytokine-receptor γ-keten, is een onmisbaar onderdeel van verschillende cytokine receptoren10,11,12,13. Stammen zoals de KNIPOOG. CG -PrkdcscidIl2rgtm1Wji (NSG) en NODShi.Cg -PrkdcscidIl2rgtm1Sug (NOG) presenteren robuuste verstoring van muis cytokine signalering en volledige ablatie van NK-cel ontwikkeling, in aanvulling op ernstige aantasting van adaptieve immuniteit14,15,16.
Drie gehumaniseerd muismodellen, rekening houdend met een scid mutatie en Il2rg knock-out in HIV/AIDS-onderzoek vaak werkzaam zijn: de BLT (beenmerg/lever/zwezerik)-model, het model van de PGO (perifere bloed leukocyten) en de SRC (SCID repopulatie cel) model 3. de BLT model is gemaakt via chirurgische transplantatie van menselijke foetale lever en thymus onder de muis nier capsule vergezeld met intraveneuze injectie van foetale lever HSCs3,17,18. De BLT muismodel biedt hoge engraftment werkzaamheid, ontwikkeling van menselijke hematopoietische cellen in alle geslachten, en totstandbrenging van een sterke menselijke immuunsysteem; Daarnaast is T-cellen zijn opgeleid in een menselijke autologe zwezerik en vertonen immuunresponsen HLA-beperkte4,5,17,19. De eis voor chirurgische procedures blijft echter het belangrijkste nadeel van de BLT-model. De muismodel van PGO is opgericht door intraveneuze injectie met menselijke perifere lymfoïde cellen. Het PGO-model biedt gemak en succesvolle T-cel engraftment levert, maar de toepassing ervan is beperkt als gevolg van onvoldoende B-cel en myeloïde cel engraftment, laag engraftment niveau algemene en het ontstaan van ernstige graft - versus - host-ziekte (GVGH)3 ,20. De SRC muismodel is door injectie van menselijke HSCs in pasgeboren of jonge volwassen SCID muizen aangetoond. Het gemiddelde engraftment efficiëntie boven 25% (beoordeeld als perifeer bloed CD45 percentage) vertoont en ondersteunt de ontwikkeling van meerdere-bloedlijn van geïnjecteerd HSCs en het uitwerken van een aangeboren menselijke immuunsysteem. De beperking van het SRC-model is echter dat de T-cel-respons is muis H2-beperkt in plaats van menselijke HLA-beperkte14,21.
De SRC muismodel wordt beschouwd als een model voor facile en betrouwbare voor preklinische HIV/AIDS kleine dierlijke studies, wordt geïllustreerd door het consistent engraftment van een menselijke immuunsysteem en succesvolle hematopoietische ontwikkeling. Wij eerder gemeld van de oprichting van een muismodel NSG Hu-SRC-SCID (hu-NSG) en de toepassing ervan in de HIV-replicatie en latentie studies22,23,24beschreven. Deze hu-NSG muismodel vertoont hoge niveaus van beenmerg homing, gevoeligheid voor HIV-infectie, en de recapitulatie van HIV-infectie en pathogenese. Bovendien, de hu-NSG muismodel correct reageert op combinatorische antiretrovirale therapie (cART) en recapituleert de virale rebound plasma bij kar geldopname, bevestiging van de oprichting van een HIV latency reservoir25,26 ,27. Dit reservoir van HIV latency wordt verder onderbouwd door overlegging van replicatie-bevoegde HIV virussen ex vivo geïnduceerd door mens rust CD4 + T-cellen van geïnfecteerde en kar-behandeld hu-NSG muizen geïsoleerd.
Hierin beschrijven we de gedetailleerd protocol voor oprichting van de hu-NSG muismodel van neonatale NSG muizen, met inbegrip van de procedures met betrekking tot HIV-infectie en kar behandeling voor latency ontwikkeling. Wij verwachten dat dit protocol te bieden een nieuwe reeks van benaderingen in HIV dierlijke studies met betrekking tot HIV Virologie, latency en behandeling.

humanized nod/scid/il2r&#947;null (hu-nsg) mouse model for hiv replication and latency studies

Ethische regels en technische uitdagingen voor onderzoek in de menselijke pathologie, immunologie en therapeutische ontwikkeling hebben kleine diermodellen geplaatst in hoge vraag. Met een nauwe genetische en gedragsmatige gelijkenis met mensen zijn kleine dieren zoals de muis goede kandidaten voor ziekten bij de mens modellen, waarlangs mens-achtige symptomen en reacties kunnen worden gerecapituleerd. Verder kan de genetische achtergrond van muis aangepast aan uiteenlopende eisen worden gewijzigd. De KNIPOOG/SCID/IL2rγnull (NSG) muis is één van de meest gebruikte immuungecompromitteerde muis spanningen; Hierdoor engraftment met menselijke hematopoietische stamcellen en/of menselijke weefsels en de verdere ontwikkeling van een functionele menselijk immuun systeem. Dit is een kritieke mijlpaal in het begrip van de prognose en pathofysiologie van mens-specifieke ziekten zoals HIV/AIDS en medeplichtigheid aan het zoeken naar een remedie. Hierin, rapporteren we een gedetailleerd protocol voor het genereren van een gehumaniseerd NSG muismodel (hu-NSG) door hematopoietische stamceltransplantatie in een muis neonatale NSG straling-geconditioneerd. De hu-NSG muismodel toont meerdere lineage ontwikkeling van getransplanteerde menselijke stamcellen en de gevoeligheid voor HIV-1 virale infectie. Het recapituleert ook belangrijke biologische kenmerken in respons op combinatorische antiretrovirale therapie (cART).

Stroom cytometry analyse wordt vaak uitgevoerd om te valideren van de zuiverheid van geïsoleerde HSCs, engraftment niveaus, profiel immuun reacties op virale infectie, evalueren en enquête kar werkzaamheid. Een typische antilichaam deelvenster bevat 4-6 individuele fluorescently gelabelde antilichamen; een cytometer van de stroom met meerdere lasers en een brede selectie van filters is dus cruciaal om accurate resultaten te behalen.

Deze zaken kunnen helpen bij het beantwoorden van belangrijke vragen in hiv/aids-onderzoek, zoals het moleculaire mechanisme en de prognose van hiv-infectie, en hoe en waar het latentiereservoir is gevestigd. Het voordeel van deze techniek is dat het niet-invasief is, en vestigde een hu-NSG muizen die een multi-lineage ontwikkeling over getransplanteerde menselijke stamcellen die vatbaar zijn voor HIV-infectie en ontvankelijk zijn voor cART te behandelen. Begin met het filteren van een verteerd, menselijk, foetaal, leverweefsel, drijfmestmonster door een steriel, 70 micrometer, nylon, mesh-zeef om een enkele celsuspensie te verkrijgen en te verrijken voor CD34+HSCs door magnetische geactiveerde celsortering volgens de instructies van de fabrikant.Na het tellen, opnieuw opschorten de HSC op een twee-keer-tien-tot-de-zevende levensvatbare cellen per milliliter concentratie in verse DPBS op ijs. Plaats vervolgens een hele neonatale menselijke NOD / SCID / gamma of NSG muis strooisel in een steriele, taart-vormige, bestraling kooi, en behandelen de dieren met 200 tot 250 centigrades van gammastraal uit een cesium-137 stralingsbron. Laad vervolgens de cellen in een op maat gemaakte, Hamilton 80508 50-microliter spuit uitgerust met een 30-gauge naald, en injecteer elk neonatale dier direct in de lever met vijf-keer-tien-tot-de-vijfde levensvatbare menselijke CD34 + HSCs en 25 microliters van DPBS.Tien tot twaalf weken na infectie, spin down retro-orbitale, perifere, bloedmonsters van elke muis door centrifugatie, en breng de plasma supernates naar microcentrifuge buizen voor negatieve 80 graden Celsius opslag als experimenteel geschikt. Voor HSC engraftment validatie, opnieuw opschorten van de pellets met 200 microliters van rode bloedcel lyse buffer, en trek de cel suspensies in een 1,5 millimeter microcentrifuge buis voor een tien minuten incubatie bij kamertemperatuur. Verzamel vervolgens de resterende witte bloedcellen door centrifugatie voor twee wasbeurten in 0,5 milliliter verse, 0,01 procent runderserum albumine, of BSA, in DPBS per wasbeurt.Na de tweede wasbeurt, blokkeer de cellen met 100 microliters van het blokkeren van cocktail gedurende twintig minuten op 4 graden Celsius, gevolgd door etikettering met de juiste anti-body cocktail van belang bij een 2 microliter anti-lichaam per een-keer-een-op-de-zesde cellen concentratie. Na 30 minuten bij 4 graden Celsius, was de cellen twee keer in verse DPBS plus BSA, en evalueer de perifere bloedmonsters door stroomcytometrie volgens standaardprotocollen om het percentage menselijke CD45+CD3+CD14+of CD19+cellen per monster te kwantificeren. Bereken vervolgens de verhouding van CD4+-cd8+T-cellen en beoordeel de virale belasting met in de plasmamonsters volgens standaard, kwantitatieve, omgekeerde transcriptase, PCR-protocollen.Om de effecten van HIV-infectie te beoordelen, leveren HIV-baalvirus aan verdoofde, menselijke, NSG muizen met een meer dan 20 procent menselijke CD45 +cel populatie in hun perifere bloed, op een 200 nanogram P24 per muis concentratie via intraperitoneale toediening, het verzamelen van bloedmonsters via retro-orbitale bloeden om de twee weken, en analyseren zowel de CD4-to-CD8 verhouding en de plasma virale belasting , zoals aangetoond. Voor virale onderdrukking validatie leveren de geïnfecteerde, menselijke NSG dieren met verse, combinatorische, anti-retrovirale therapie, of cART, in gezoete water elke week gedurende 4 weken, het verzamelen van bloedmonsters via retro-orbitale bloeden om de twee weken en analyseren zowel de CD4-to-CD8 verhouding en het plasma virale belasting zoals aangetoond. Om de virale rebound te beoordelen wanneer de plasma virale belastingen onder de detectielimiet liggen en de CD4-naar-CD8-verhouding wordt hersteld tussen 1,5 en 2,5 perifere bloedmonsters verzamelen door middel van retro-orbitale bloedingen om de 2 weken na cART terugtrekking en evalueren van de monsters zoals aangetoond.Tijdens de eerste validatie van engraftment moet het aantal menselijke CD45+-cellen variëren van 20 tot 80 procent en moeten de subsets van menselijke leukocyten worden weergegeven als afzonderlijke populaties door stromingscytometrische analyse. De verhouding van CD4+to-CD8+cellen blijft tussen 1,5 en 2,5 bij gezonde personen, terwijl significante CD4+T-celuitputting doorgaans wordt waargenomen bij virale infectie. Met name de gezonde verhouding wordt hersteld in reactie op cART-behandeling.De detectie van plasma virale belastingen door kwantitatieve RT PCR in de loop van de infectie en het cART-regime kan worden uitgezet en gebruikt om de efficiëntie van de virale levering en de antivirale behandeling te evalueren. Hoewel deze methode inzicht kan geven in hiv/aids virologiestudies en therapeutische ontwikkeling, kan het ook worden toegepast op andere studies met betrekking tot menselijke ziekten, zoals kanker, diabetes en auto-immuunziekten. De ontwikkeling van het hu-NSG-muismodel maakte de weg vrij voor onderzoekers in immunologie en oncologie om een nieuwe disfunctie en pathologie bij de mens te onderzoeken, met name mechanistische en therapeutische studies gericht op chronische hiv-infecties en het latentiereservoir.
